Abstract

Sperm plasma membrane was rich unsaturated fatty acid and vulnerable to peroxydative damage. Vulnerability sperm against lipid peroksidation could be caused by increased cold shock. The peroxydative processed alter the structure of. sperm, particularly in the membrane and akrosom, lost motility, metabolic changed and the rapid released intraseluler component. This condition could be prevented by adding antioxidants to semen extender, such as vitamin E and BHT. Vitamin E has been proven to protected sperm plasma membrane during freezing to thawing, while Butylated Hydroxytoluene (BHT) prevent the sperm plasma membrane damage caused cold shock and provide protection against changes due to freezing. Vitamin E and BHT prevent lipid peroksidation through its the hidrogen atom to the radical peroksil rapidly. It can be concluded that an antioxidant could be prevent damage the sperm plasma membrane was caused cold sho(:k and provide protection against changes due to freezing and to prevent lipid peroksidation.

Abstract

Research on utilization sugar cane water combined with egg yolk as Bali bull semen extenMr have been implemented. Research was conducted to determine the level ofutilization ofsugar cane water as semen exten~r combined with egg yolk on the quality ofBali bull semen. Variable observed in this study were the percentage ofmotility, the percentage of live count and intact plasma membrane of sperm. Based on the Complete Random Design (RAL) with $ix treatments and three replications. Treatment A = skim milk extender (control), B = 10% sugar cane water extender, C"" 20% sugar cane water extender, D =30% sugar cane water extender, E =40% sugar cane water extender and F = 50% sugar cane water extender. Differences between treatment tested with Duncan's Multiple Range Test (DMRT). Results of this research indicated that treatment 30% and 40% sugar cane water extender that could maintain the quality of the semen longger than the other treatment (96 hours), with the percentage ofsperm motility above 40% and the percentage oflive sperm above 50%. It can be concluded that : (1) a combination ofsugar cane water with the egg yolk could maintain ofsperm motility, sperm Viability and sperm intact pl~ma membrane stored at a temperature of SoC up to 96 hours, (2) sugar cane water could be used as natural extender with the concentration of30% to 40%.

Abstract

A study about the use ofvitamin E and BHT in frozen ram senten extender has done. Tris citrate, lactose and skim milk as extenders and vitamin Eand BHT as antioxidants with ceitain doses were used in this experiment. The quality ofsemen such as motility, live sperm, intact plasma membrane and intact acrosom membrane were evaluated. The objectives ofthis study are to determine the best extender and antioxidant with certain doses in cryopreservation of ram semen. Based on" this study it was found that the skim milk extender (50.0 and 62.5% respectively) showed the best quality ofsperm motility and live sperm after thawing. The mean percentage ofintact plasma membrane and intact acrosome on skim milk extender also showed the best result(58.3 and 61.0% respectiDely). The supplement of0.2 g vitamin Eshowed higher rate ofsperm motility, live sperm, intact plasma membrane and intact acrosome after thawing than the other treatments (51.9, 65.4, 61.3 and 64.2 percent respectiDely). It can be concluded that the supplementation of 0.2 g vitamin E using the skim milk extender gave the best quality in ram semencryapreservation.
