Penelitian ini bertujuan untuk mendapatkan bakteri asam laktat penghasileksopolisakarida (EPS).37 spesies bakteri asam laktat dari yogurt komersial, kultur indigenous danLeuconostoc mesenteroiodes FNCC 0023 yang diseleksi kemampuannya untukmenghasilkan eksopolisakarida menggunakan medium (skim 2%,skim2%+sukrosa5%, dan skim2%+sukrosa10%) dan medium agar (whey agar, wheyagar+sukrosa5% and whey agar+sukrosa10%) menggunakan metode pour plate andspread plate dan diinkubasi pada suhu 30C.Pada medium cair tidak didapatkan bakteri asam laktat penghasileksopolisakarida. Leuconostoc mesenteroides FNCC0023 pada whey agar+sukrosadengan metode pour plate dan spread plate menghasilkan EPS mucoid, pada wheyagar dan whey agar+sukrosa dengan metode spread plate menghasilkan EPS ropy.Lactobacillus sp (Dad1) pada whey agar and whey agar+sukrosa produce EPS ropynamun tidak konsisten, dan Lactobacillus reuteri (M11), Lactobacillus plantarum(Mut4), Lactobacillus sake (Mut5) dan Lactobacillus sake (Mut13) pada whey agardan whey agar+sukrosa menghasilkan EPS mucoid namun tidak konsisten.Disimpulkan bahwa L. mesenteroide FNCC 0023 menghasilkan 2 jenis EPS,mucoid dan ropy. Terdapat beberapa kultur indigenous yang menghasilkan EPS yangtidak konsisten.Kata kunci: Bakteri asam laktat, Leuconostoc mesenteroides FNCC0023,Eksopolisakarida.ABSTRACTThe purpose of this study was to find out lactic acid bacteria producingexopolysaccharides.37 spesies of lactic acid bacteria from commercial yogurt, indigenous cultureand Leuconostoc mesenteroiodes FNCC 0023 was selected their ability to produceexopolysaccharides using liquid medium (2% skim, 2% skim+5%sucrose, and 2%skim+10% sucrose) and agar medium (whey agar, whey agar+5%sukrosa and wheyagar+10%sukrosa) using pour plate and spread plate which incubated at 30C.The liquid medium not find out lactic acid bacteria producingexopolysaccharides.Leuconostoc mesenteroides FNCC0023 on whey agar+sukrosawith pour plate and spread plate method produce mucoid EPS, on whey agar andwhey agar+sukrosa with spread plate method produce ropy EPS. Lactobacillus sp(Dad1) on whey agar and whey agar+sukrosa produce unconsistent ropy EPS, andLactobacillus reuteri (M11), Lactobacillus plantarum (Mut4), Lactobacillus sake(Mut5) and Lactobacillus sake (Mut13) on whey agar and whey agar+sukrosaproduce unconsistent EPS mucoid.We concluded that L. mesenteroides FNCC 0023 produced 2 type of EPS,mucoid and ropy.
