Abstract

Salah satu target untuk menghambat penyebaran penyakit kanker adalah dengan menghambat proses proliferasi sel kanker yaitu dengan menghambat pengatur aktivitas siklus sel kanker  Cyclin dependent kinase 4 (CDK4).  Hingga saat ini, struktur kristal CDK4 belum tersedia. Struktur CDK4 yang mirip CDK2  belum tervalidasi terhadap inhibitor ligan native inhibitor reseptor CDK4, sehingga virtual skrining untuk menemukan senyawa bahan alam  untuk menghambat proliferasi sel kanker masih belum valid dalam pemakaian struktur kristal CDK4 dan sulit dilakukan pengembangan inhibitor CDK4. Penelitian ini bertujuan untuk memvalidasi metode protokol skrining terhadap struktur CDK4 mirip CDK2 agar dapat digunakan sebagai struktur kristal dalam pengembangan inhibitor CDK4 dan melakukan virtual skrining senyawa bahan alam yang berpotensi sebagai inhibitor CDK4 sebagai antiproliferasi sel kanker. Metode yang digunakan untuk validasi protokol skrining virtual dengan cara redocking. Struktur kristal CDK4 mirip enzim CDK2 dengan kode PDB ID 1GII, 1GIH, dan 1GIJ akan diredocking dan pengamatan validasi berdasarkan nilai Root Mean Square Deviation (RMSD) < 2, sehingga protokol skrining yang di kembangkan ini diterima dan dapat dikembangkan untuk penelitian lebih lanjut untuk skrining virtual dalam usaha penemuan senyawa baru inhibitor antiproliferasi CDK4. Setelah itu dilakukan virtual skrining dan analisis interaksi antara 16 senyawa bahan alam dengan reseptor hasil validasi. Hasil validasi metode protokol skrining terhadap struktur CDK4 mirip CDK2 secara internal yang  diterima yaitu reseptor 1GIH dengan ligand native 1PU dan RMSD 0,8072. Virtual skrining dan analisis dari 16 senyawa bahan alam yang memiliki skor CHEMPLP terendah yaitu Naringin dengan skor CHEMPLP sebesar -94,1781 Kkal/mol yang jauh lebih baik dibandingkan senyawa pembanding flavopiridol namun masih lebih baik senyawa pembanding abemaciclib, ribociclib dan palbociclib. Interaksi ligand-reseptor pada naringin lebih didominasi oleh ikatan Hidrogen dibandingkan dengan senyawa pembanding abemaciclib, ribociclib dan palbociclib yang didominasi oleh interaksi hidrofobik, sehingga Naringin berpotensi sebagai inhibitor CDK4 sebagai antiproliferasi sel kanker.

Abstract

Adolescence is prone to deviations and impropriety should be grown a sense of creativity. Creativity of students to be skilled in making VCO so that will grow entrepreneurship spirit for students. Skills in the way of making VCO is expected to increase students' creativity and applied in economic activities to increase student income. The solution offered from the problems that have been mentioned above, then training the manufacture of Virgin Coconut Oil (VCO) is very necessary. Program activities offered in the form of socialization or introduction of VCO products produced both from the benefits and advantages they have compared with other similar products, and there is also a demonstration later in the making of VCO and explanation of how the right and proper marketing. Implementation of community service is specifically a promotive effort in the field of business development to create skilled and productive student students in producing a health product that can later be utilized both for themselves, the family and the surrounding community. Community service activities, also relevant to economics and business, where the product will be marketed so that this can be an income or additional income for local students. The participants' response to this counseling is quite high, indicated by the attendance rate of 80% of the number of invitations. Understanding high school students and SMK Mutiara to the benefits and content of VCO and how to manufacture the correct VCO using this simple and cheap enzymatic method was responded with great enthusiasm by the participants.

Abstract

EMPOWERMENT OF PKK WOMEN AND POSYANDU CADRE OF RW 022 TELUK PUCUNG, NORTH BEKASI IN THE APPLICATION OF GEMA CERMAT (INTELLIGENT COMMUNITIES USING MEDICINE). Until now in the midst of society often encountered various problems in the use of drugs, including lack of understanding of the use of appropriate and rational drugs, excessive use of over-the-counter drugs, and a lack of understanding of how to store and dispose of drugs properly. While health workers are still perceived as lacking adequate information about drug use. This is based on the appropriateness of data and information sources collected through visits and good discussions with several representatives of PKK women members and Posyandu Cadres RW 22 Teluk Pucung, North Bekasi. There are still many found, especially North Bekasi residents who sell or exchange used drugs to collectors of used goods in their residential areas or there are still many people who store liquid preparations such as syrup in the refrigerator. The purpose of this activity is to increase understanding of the treatment of drugs in how to get, use, store and dispose of and increase the rational use of drugs through Gema Cermat. The method of activities to solve the problems faced by PKK women and Posyandu cadres is by socializing about the Gema Cermat by filling out the questionnaire before and after the socialization. Based on the results of the pre-test and post-test questionnaires, the participants became more aware of the management of drugs at home and increased knowledge of the use of rational drugs such as antibiotics. The conclusion of this activity is to increase participants’ understanding of drug management properly and is expected to be implemented in the family and community environment.
