Abstract

The high content of α-galactooligosaccharide in mung beans has the potential for supporting the growth of Lactic Acid Bacteria (LAB), therefore it can be developed as fermented plant-based milk that substituted Ultra High Temperature (UHT) milk. The purposes of this research is to investigate the effectiveness of LAB Lactococcus lactis NBRC 12007 in the fermentation process of mung bean milk and to observe the quality differences of fermented mung bean milk with UHT milk.  Previously, preparations were made for starter L. lactis NBRC 12007 in MRSB medium and then sub cultured to UHT milk. Furthermore, 3% of the culture was inoculated into each of the mung bean milk substrate and UHT milk as a starter for the fermentation process and incubated for 24 hours at 37oC. During this process, the effectiveness of LAB was tested (total cell, total acid, total protein, pH, antibacterial) and the quality test of fermented products (organoleptic and physical).  Based on total cells, total acid, total protein, pH, and antibacterial test findings, fermented mung bean milk products had a higher value than UHT milk products. During the process, it was proven that L. lactis NBRC 12007 had high effectiveness in producing and improving the quality of fermented mung bean milk compared to UHT milk. It is assumed that the content of oligosaccharides supports the growth of LAB and the fermentation process.

Abstract

The present research was conducted to investigate sulphate and manganese removal from synthetic wastewater. The continuous laboratory scale of down-flow fluidized-bed reactor (DFBR) using sulphate reducing bacteria (SRB) consortium and Indonesian natural zeolite as a bacterial support material was designed. At 9 days operation, maximum sulphate and manganese removal was observed to be 23% and 15.4%, respectively. The pH values were also changed to neutral. The population of SRB increased which effect on the raising of their activity for removing sulphate and manganese. Using the scanning electronic microscopy (SEM), it was observed that natural zeolite possesses excellent physical characteristics as a bacterial support material in DFBR. The imaging SEM result of SRB consortium on zeolite surface clearly showed the developed SRB biofilm on that particle. Analysis result of EDX confirmed that manganese was precipitated as manganese–sulfides.

Abstract

Pedukuhan Kraton, Desa Mulyodadi, Kecamatan Bambanglipuro, Kabupaten Bantul merupakan salah satu wilayah dengan jumlah rumah potong ayam (RPA) cukup banyak, satu RPA skala besar dan 3 lainnya skala kecil. Selama kegiatan industri RPA akan dikeluarkan limbah organik berupa limbah cair dan padat dengan kandungan senyawa organik yang sangat tinggi serta bakteri patogen. Selama ini baik limbah cair maupun padat limbah RPA di Pedukuhan Kraton, Desa Mulyodadi tersebut telah diolah, akan tetapi sistem pengolahannya masih sangat sederhana belum menggunakan prinsip zero waste, sehingga masih sangat potensial mencemari lingkungan sekitar. Melalui kegiatan Pengabdian Merdeka Belajar 3 mahasiswa Fakultas Biologi UGM melakukan pendampingan dan fasilitasi kepada anggota kelompok peternak dan petani di Pedukuhan Kraton untuk mengolah limbah tersebut dengan prinsip zero waste. Limbah organik industri RPA diolah dengan prinsip zero waste melalui pemberian nutrien mikrobia, starter mikrobia aktif berupa EM4 dan zeolit alam sebagai biofilter sekaligus katalis. Limbah industri pabrik gula (molase) dan limbah pertanian yang sangat melimpah juga dimanfaatkan sebagai nutrien mikrobia. Kegiatan ini sangat berguna dalam meningkatkan kemandirian dan kinerja kelompok peternak dan petani dalam mengelola limbah RPA di Desa Mulyodadi sehingga beban pencemar limbah hilang sekaligus diperoleh pupuk organik cair dan kompos. Kegiatan ini akan dapat meningkatkan perekonomian sekaligus menjaga kelestarian lingkungan.
