Abstract

Watermelon (Citrullus lanatus (Thunb.) Matsum. & Nakai) is a horticultural plant that belongs to the Cucurbitaceae family with high public demand, however, local markets sometimes have limited supply. The existence of watermelon varieties that are not pest-resistant causes its production to be erratic. Therefore, plant breeding efforts are required to produce superior varieties through the stability test of plant characters. For watermelon to be certified as a new variety, it needs to possess a stable and adaptive character to various conditions. The F1 watermelon from crossing ♀ ‘Putri Delima’ with ♂ ‘Maduri’ produces inole-shaped fruit, red flesh, and a sweet taste. Therefore, this study aims to determine the stability of the phenotypic character of watermelon F2 from crossing ♀ ‘Maduri’ with cultivar ♂ ‘Putri Delima’ and was conducted in Jamusan, Bokoharjo, Prambanan, Sleman, D.I.Yogyakarta fields from August to December. There are five samples of ripe watermelons that were selected randomly while their phenotypic characters were observed qualitatively and quantitatively. Each F2 watermelon character was compared to F1, and the quantitative analysis was conducted using one-factor ANOVA with a confidence level of 5%. The results of quantitative character analysis between F2 and F1 showed a P (P-value)> 0.05. Meanwhile, the results of qualitative observations of F2 watermelon showed different flesh and skin color, while the harvest time from F1 was caused by the segregation of heterozygous crosses. Therefore, it is necessary to select superior phenotypic characters as desired for the next breeding.

Abstract

Indonesia is an agrarian country whose territory is partly used as an agricultural sector, has especially watermelon (Citrullus lanatus (Thunb.) Matsum & Nakai). At present, the development of watermelons in Indonesia still requires a variety of colors, superior watermelons need to be developed by making triploid orange watermelons. The new watermelon cultivar (F₁) is the result of crossing 'Jelita' (♀) and 'BallyBall' (♂) which are expected to provide a combination of characters that can produce large and sweet fruits. The purpose of this study was to identify ISSR markers that were associated with ploidy levels of horticultural crops, particularly watermelon. The watermelons were growns on the agricultural land of Kebondalem Hamlet, Madurejo Village, Prambanan District, D.I.Y. and analyzed at the Laboratory of Biotechnology PAU, UGM and Laboratory of Genetics and Breeding, Faculty of Biology UGM. Analysis of genetic variation was carried out using the PCR-ISSR method and the DNA bandwidth was calculated using Paint Apps, Microsoft Excel 2013, and Microsoft Word 2013. The results of crossing 'Jelita' and 'King Quality 'is sterile. The result of 'Jelita' with 'BallyBall' produces tillers until sterile harvest time. The results of the analysis of genetic variation using the PCR-ISSR method indicate that the ISSR BI, B3, B5, and CBTC 1 molecular markers cannot be used to distinguish ploidy from the tested watermelons.

Abstract

Pandemi Covid-19 menyebabkan perubahan kondisi sosial ekonomi masyarakat Indonesia. Sekitar 40% pekerja atau pelaku usaha mengalami perubahan pendapatan. Hal ini sedikit banyak mempengaruhi kemampuan masyarakat dalam memenuhi kebutuhan pangan keluarga. Pengabdian masyarakat ini bertujuan untuk memberikan edukasi kepada masyarakat untuk mengoptimalkan pemanfaatan pekarangan dan limbah rumah tangga untuk memenuhi kebutuhan pangan keluarga melalui budidaya lele. Pelaksanaan program dibagi ke dalam tiga tahap, yaitu pelatihan warga, pendampingan, dan evaluasi.Pelatihan warga meliputi pelatihan budidaya lele dan kangkung secara hidroponik menggunakan pot dan pelatihan pembuatan pakan lele dari limbah rumah tangga. Pendampingan dilakukan secara berkala setiap minggu hingga lele bisa dipanen. Evaluasi dilakukan di akhir kegiatan untuk mengetahui efektivitas dan potensi teknologi untuk dikomersilkan. Hasil dari program ini menunjukkan bahwa budidaya lele dan kangkung menggunakan pot dapat digunakan untuk memenuhi kebutuhan pangan keluarga dengan harga yang murah. Desain teknologi kolam portabel dari pot ini juga berpotensi untuk dikembangkan sebagai industri rumah tangga. Pakan lele dari limbah rumah tangga mampu menekan biaya pakan dalam budidaya lele hingga 20% dengan nilai rasio konversi pakan (FCR) sebesar 1.11 atau efisiens pakan hingga  90%. Pakan lele ini membantu pengurangan sampah sisa makanan dan sampah dapur hingga 100%.
