Abstract

Test were conducted to find the biopotency of pituitary glands that havebeen dry-preserved for 0, 1, 2 3 and 4 months. Pituitary glands, obtainedfrom the waste of fillet production plant of Pangasius pangasius, weredried gradually in acetone. The treatments were dry-preservation timespan in dessicator: 0, 1, 2, 3 and 4 months. This hormone biopotencytesting used extract of late-stage of gonad maturation from female andfrom induced-spawning. No significant differences were found in thepercentage of late-stage gonad maturation and ovulation, relativefecundity, fertility and hatchability from every treatment. The number offemales reaching late-stage of gonad maturation were 100% (for allmonths of preservation), while the mean percentages of : relativefecundity was 18.05% (for 3 months of preservation), fertility was 95.73%(for 1 month of preservation), 24-hour hatchability was 31.63% (for 0month of preservation). No significant differences were found for allpreservation time span.

Abstract

Penyimpanan jangka pendek mikroba umumnya dilakukan pada suhu dingin non-beku (refrigerator), sedangkan penyimpanan jangka menengah–panjang, harus dalam kondisi beku. Penyimpanan dalam kondisi beku perlu dicegah terbentuknya kristal es di dalam sel, dapat dilakukan dengan pemberian agen krioprotektan seperti gliserol. Telah dilakukan pengujian terhadap 2 koleksi kultur campuran mikroba pendegradasi hidrokarbon yang dimiliki Laboratorium Mikrobiologi, Balai Teknologi Lingkungan BPP Teknologi, yaitu konsorsium A dan B. Hasil menunjukkan bahwa kedua kultur yang disimpan dalam suhu dingin non beku hanya mampu bertahan kurang dari 3 bulan. Sementara jika kultur tersebut disimpan pada suhu beku dapat memperpanjang umur simpan.. Viabilitas kultur yang disimpan beku selama 3 bulan relatif cukup stabil karena hanya turun rata-rata 1 log. Sementara itu penyimpanan beku selama 6 bulan menyebabkan penurunan viabilitas 2 log, berikutnya penyimpanan hingga 2 tahun menyebabkab viabilitas kembali turun 2 log. Pengujian kinerja dalam mendegradasi hidrokarbon hingga hari ke-14 mampu menurunkan kadar minyak mentah hingga masing-masing 44% untuk konsorsium mikroba A dan 47% untuk konsorsium mikroba B. Kemampuan pulih kultur dipengaruhi oleh konsorsium mikroba. Konsorsium A mempunyai kemampuan pulih lebih rendah dibandingkan konsorsium mikroba B, baik untuk penyimpanan di suhu dingin non beku maupun dingin beku.
