Abstract

Rheumatoid arthritis is a chronic inflammatory autoimmune disease associated with articular and systemic effects. This disease affects synovial joints covered by a special tissue called synovium. Curcumin has a potent antioxidant, antiinflammatory agent, antiangiogenic and anticarcinogenic. Curcumin can downregulate the expression of various proinflammatory cytokines and is reported beneficial effects in arthritis, but has a poor solubility dan bioavailability as well. The purpose of this research was to study the potential of liposomes topikal curcumin in reducing athritis score, reducing the expression of TNF-α and histopathological synovium hyperplasia of hind paw on Wistar rats with CFA that had been treated with topical curcumin. In this study, rats were divided into 7 groups: positive control, negative control, rheumatoid arthritis with topical curcumin therapy of 90 mg/kg BW, rheumatoid arthritis with topical curcumin therapy of 110 mg/kg BW, rheumatoid arthritis with topical curcumin therapy of 200 mg/kg BW, rheumatoid arthritis with methotrexate therapy, rheumatoid arthritis with placebo therapy. Results from this experiment indicated that topical curcumin has no significant to the arthritis score, significantly effect to percentase expression of TNF-α (p<0.05) and could decrease synovium hyperplasia based on histophatology examination. It could be concluded that therapy of topical curcumin could decrease the expression of TNF- α and synovium hyperplasia in rheumatoid arthritis rat. 
Agarose Coated Culture Plate in Tumorsphere Culture of Cervical Cancer Cell Line HeLa: an Alternative to Non Adhesive Culture Plate Putu Juniartha; Muhammad Rasjad Indra; Hidayat Sujuti; Diana Lyrawati; Tatit Nurseta

Abstract

Cervical cancer recurs in 90% cases and linked to cancer stem cells that able to self-renew and responsible for recurrence, metastasis, and mortality of cancer. Isolation and identification of cancer stem cells using serum-free medium needs expensive growth factors and consume time. This study try to grow tumor sphere using culture plate coated with 1% agarose as an efficient and economical alternative to non-adhesive culture plate. HeLa cell line was grew in culture plate coated with 1% agarose and non-adhesive culture plate using similar medium and culture condition. Tumor spheres morphology was observed and the colonies were counted in 7 days followed by single cell assay. Tumor spheres then counted for CD133+, CD34+, and Sox2 expression using flowcytometry. Culture plate coated with 1% agarose can be used as an economic and efficient alternative to culture tumor sphere. Using culture plate coated with 1% agarose, the tumor spheres formed in 7 days with similar morphology to non-adhesive culture plate. Tumorsphere had three dimensional – sphere shape that tightly attached, colonized, and overlapped. The tumor sphere colony counts of two plates were statistically have no significant difference (p=0,667). Single cell assay of a tumor sphere shows that it can grow new tumor spheres with similar morphology. The tumor sphere from culture plate coated with 1% agarose express CD133+ and CD34+ as much as 8.78% ± 2.14 and Sox2 as much as 35.30% ± 23.82 whereas tumor sphere from non-adhesive culture plate express CD133+ and CD34+ as much as 62.36% ± 1.06 and Sox2 as much as 98.86% ± 0.56 (p = 0000).

Abstract

Abstract Objectives: The availability of prevalence data for osteoporosis in Indonesia is limited due to the constrained access to BMD measurements, especially in rural areas. Integrated Health Post for the Elderly (Posyandu Lansia) is one of the Indonesian government's programs to monitor and provide health care for the elderly, including postmenopausal women. This study aimed to determine the prevalence of osteoporosis and related risk factors in postmenopausal women. Methods: Respondents were 249 postmenopausal women from 440 people who attended the Posyandu Lansia from 9 villages in Sidoarjo. Quantitative ultrasound was used for calcaneal BMD measurement; other data were collected via questionnaires and enumerated by trained villagers. Results: Our study revealed that the prevalence of osteoporosis postmenopausal women was 73.5%. The osteoporosis risk factors of age, BMI, weight, and menopause duration were associated with T-scores both individually (p<0.05) or simultaneously (p< 0.001), while menopause age and exercise did not. Conclusions: Groups possessing high-risk factors associated with a high prevalence of osteoporosis are postmenopausal women age ≥ 65 years, BMI < 20 kg/m2, weight < 58 kg and duration of menopause > 6 years. Regardless of the groupings, the prevalence of osteoporosis was very high in postmenopausal women. Dietary and exercise adjustments as well as pharmacotherapy may be required to improve their bone health.
