Abstract

Post-mortem spermatozoa recovery is an important technique for obtaining germplasm reservesfrom genetically valuable animals or endangered species. The purpose of this study was to studywhether ram spermatozoa within epididymides stored at 4º C for 24 and 48 h remain their motility andviability. The characteristic of ram epididymal spermatozoa after freezing and thawing was alsoobserved. Six pairs of ram testes with attached epididymides were used in this study. The motility ofcontrol spermatozoa was well maintained throughout the dilution procedure (83.3±1.1, 80±1.3, and80±1.3% for collection, Niwa and Sasaki freezing -1 extender (NSF-1 and NSF-2 groups respectively);but declined (P<0.05) after freezing and thawing (38.3±3.1%). Motile and viable spermatozoa could berecovered from epididymides up to 48 h of storage, although their quality declined significantly(P<0.05) as post-mortem storage time increased (motility: 83±1.2, 67±3.0, and 46±5.1; viability:84.2±2.4, 73±2.8, and 66.6±2.6 % for control, 24 h and 48 h group respectively). These data indicatethat ram epididymides could be stored at 4º C for 48 h when epididymal spermatozoa cannot beimmmediately collected and cryopreserved. These storage conditions might be possible to use forepididymal sperm recovery in wild ruminants.

Abstract

Lulur merupakan kosmetika tradisional, yang salah satu bahan dasarnya adalah ketan hitam. Pada penelitian ini pembuatan lulur beras ketan hitam ditambahkan dengan minyak kelapa, teh hijau dan susu. Saat penyimpanan dibutuhkan kestabilan formulasi, untuk itu peneliti menambahkan antioksidan berupa tokoferol (vitamin E). Pada penelitian ini kestabilan 2 formula, yaitu formula tanpa dan dengan tokoferol diamati. Pengamatan dilakukan selama 2 minggu, dengan mengukur kadar keasaman (pH) dan viskositas. Hasil penelitian menunjukkan kedua formulasi yng dibuat tidak menunjukan perbedaan signifikan. Pada formula dengan tokoferol didapatkan penurunan kadar keasaman sebesar 0,03 %, sedangkan pada formula tanpa tokoferol 0,017%. Sedangkan untuk data viskositas didapatkan tidak ada perubahan viskositas pada formula dengan tokoferol dan perubahan sebesar 0,18 % pada formula tanpa tokoferol. Kesimpulannya adalah kedua formula memiliki kestabilan yang hampir sama selama penyimpanan. Hal tersebut disebabkan karena pada kedua formula terdapat bahan campuran berupa minyak kelapa yang memiliki kemampuan sebagai antioksidan setara dengan tokoferol. Kata kunci: anti oksidan, lulur ketan hitam, minyak kelapa ABSTRACT Lulur is a traditional cosmetic, and one of its ingredients is black sticky rice. In this study, black sticky rice was mixed with coconut oil, green tea, and milk to prepare the body scrub. To keep the formula stable during storage, adding an antioxidant in the form of tocopherol (vitamin E) is needed. The stability of 2 formulas, namely formulas without and with tocopherol, was observed. The observations were carried out for two weeks by measuring the acidity levels (pH) and viscosity. The research results showed that the two formulas did not show any significant differences. In the formula with tocopherol, the acidity level was reduced by 0.03%, while without tocopherol by 0.017%. However, there was no change in the viscosity of the formula with tocopherol, while in the formula without tocopherol, there was a slight change of 0.18%. In conclusions, both formulas had almost the same stability during storage. These were because both of the formula consisted of coconut oil that can act as an antioxidant equivalent to tocopherol.
