Abstract

An oxytetracycline-tolidin-bovine serum albumin (OTC-tolidin-BSA)-conjugate was synthezed as immunogen for producing specific antibodies in immunized rabbits that would be used as reagent for development of OTC residue detection with enzym-linked immunoassays technique. The immunogen was prepared through diazotization tolidin and subsequently reacted with OTC. The red purple immunogen of OTC-tolidin-BSA absorbed at wave lengths of 277 nm and 488 nm under UV screening absorbances and confirmation with the high performance liquid chromatography (HPLC) showed the absence of peak at retention time of 3.46 minutes. Characaterized result with SDS-PAGE showed the molecular weight of the OTC-tolidin-BSA at 69.79 kDA. Subsequently, the immunogen was immunized into New Zealand rabbits in order to produce the polyclonal antibodies. The antibodies were purified using a protein A sepharose column. The OD optimum responses of 0.92 to 1.20 were obtained from the second fractionation at dilution of 1/1000 by titrating the antibodies and OTC-tolidin-BSA coating antigen at concentration of 10 µg/mL on several bleeding times. Abstract

Chloramphenicol (CAP) is a broad spectrum antibiotic that has been banned in many countries due to its serius side effect to human. Detection of CAP residue in food has been determined to a minimum required performance limit (MRPL) of 0.3 ng/g. The purpose of this research was to conduct the analysis of CAP residue in bovine meat by using LCMS and to study the presence of CAP residue in marketed bovine meat samples. LC separation was done on a Shimpack column C18 with ammonium acetate 10 mM/water as mobile phase, and ESI-MS analysis in negative ion mode. The coefficient of determination, R2 = 0.9981 at concentration of 0.125, 0.25, 0.63, 1,00 and 2.00 ng/g. Recovery at three fortification levels (0.25, 0.50 and 1.00 ng/g) was in the range 77.5, 97.3 and 83.4%. The decision limit and the detection capability were 0.15 ng/g and 0.17 ng/g respectively. Analysis results of 52 marketed samples showed that CAP residue were detected in 9 samples in the concentration range of 0.14 to 2.70 ng/g and 6 among those positive samples were above the MRPL value. Therefore, it is important to increase the awareness and also to monitor regularly CAP residues in food originated from animal to provide safe food for the consumers. Abstract

IKM anyaman agel adalah salah satu sub sektor IKM kerajinan yang mampu menggerakkan ekonomi danmenjadi ikon kabupaten Kulon Progo. Agel diperoleh dari hasil pembelahan bagian atas pucuk daun CoryphaGebanga .Teknologi yang digunakan masih sangat sederhana namun mampu merambah ke pasar mancaNegara seperti USA, Eropa, Australia, Malaysia, Jepang. Tahun 1998, meski kondisi Negara Indonesiamengalami krisis moneter, dimana banyak perusahaan besar sedang tiarap , justru para pelaku industri agelmengalami “golden era”. Tahun 2009 Realisasi ekspor kerajinan agel dari Kulon Progo mencapai volume831.000 kg dengan nilai $ 1.138.800. Luas panen tanaman gebang di Kulon Progo hanya tinggal 47 Ha denganproduktivitas 0,83 ton/Ha/tahun . Hasil dari luas panen tanaman agel di Kulon Progo diperkirakan hanya mampumensuplay kurang dari 1 % kebutuhan bahan baku padahal pasar kerajinan anyaman agel masih sangat terbuka.Oleh karena itu Model Bauran Pemasaran dari Craven,1994 diharapkan dapat menggambarkan strategi kelolausaha pada IKM anyaman agel di Kulon Progo. Hasil penelitian menunjukkan bahwa secara alamiah IKManyaman agel sudah membentuk klaster industri tersendiri untuk memenuhi bahan baku, menerapkan strategiagar proses produksi dapat berlanjut, mempromosikan & memasarkan produk, menerapkan strategi harga,kesemua hal tersebut bermuara pada kepuasan pembeli /pelanggan.Kata kunci: agel, Bauran Pemasaran, Kelangkaan Bahan baku klaster industri, nilai tambah, tenaga kerja.
