Abstract

Empat flavonoids (Sciadopitysin) (1), Ginkgetin (2), Ishorhamnetin (3), dan Quercetin (4), dua Texanes (Taxinine) (5) dan Taxol (6), dan Phenylisoserine methyl ester (7) dapat diisolasi dari Taxus cuspadata var. nana. Senyawa (7) juga dapat diisolasi dari Taxus chinensis. Senyawa (7) dapat diisolasi untuk pertama kalinya masing-masing dari T. cuspidata var. nana dan T. chinensis.Aktivitas antifungus senyawa-senyawa yang terisolasi tersebut dan tujuh turunan taxinine terhadap tujuh fungi patogenik tanaman (Cochliobolus miyabeanus, Alternaria kikuchiana, Giberella fujikuroi, Cladosporium cucumenium, Fusarium oxysporum, Colletotrichum fragariae dan Corynespora cassiicola) telah diukur. Taxinine dan tujuh turunan taxinine menunjukkan adanya aktivitas antifungus terhadap lima jenis fungi. Diantara tiga jenis kayu tropis (Amboyna,Kempas,Angsana),kayu Amboyna menunjukkan aktivitas antifungus terhadap empat fungi dari basidiomycetes (Le,Pp,G3,dan PLI). Selanjutnya, ekstrak methanolic dan fraksi larutnya nhexane dan ethyl ether dari kayu Amboyna menunjukkan aktivitas antifungus terhadap fungus Pp.Kata kunci: Antifungi, flavonoid, Texanes, phenylisoserine methyl ester, kayu tropis dan temperate, fungi patogenik tanaman, basidiomycetes.

Abstract

The decolourization of the recalcitrant dye RBBR by the culture filtrate of Polyporus sp. S133 and its isolatedlaccase was investigated. The laccase alone decolorized RBBR. A small molecular weight redox mediator (HBT) wasnecessary to increase the decolorization. The purified laccase totally decolorized the dye of 200 mg l-1 initialconcentration of RBBR when only 1.5 U ml-1 of laccase was used in the reaction mixture. The effects of differentphysicochemical parameters were tested and optimal decolorization rates occurred at pH 5 and at a temperature of 50°C. The effect of surfactants on the decolourization of RBBR was tested with Tween 80, Tween 20, and Brij 35. It wasdemonstrated that Tween 80 was inhibiting substrate for the decolorization while Tween 80 and Brij 35 was noinhibiting effect for the decolorization. Provided that all of the condition is included, it is suggested that laccase maybe suitable for the wastewater treatment of similar anthraquinone dyes.Keywords: Decolorization; Laccase; Remazol Brilliant Blue R (RBBR); Polyporus sp. Abstract

Colletotrichum species were reported producing various bioactive compounds that may potentially be used in the development of anti-microbial, antioxidant, anticancer and anti-diabetes drugs. Our previous studies showed that the methanol extracts of mycelia from Colletotrichum sp. TSC13, an endophytic fungus isolated from Taxus sumatrana (Miq.) de Laub., were found to have α-glucosidase inhibitory activities. The aim of present study was to examine the effects of initial inoculum on growth and the content of fatty acids as an α-glucosidase inhibitor in Colletotrichum sp. TSC13 that grew under shake and static condition. The results showed that shake and static conditions had more effect compared to the initial inocculum on growth and glucose consumption in growth and glucose consumption. However in terms of fatty acids contents and α-glucosidase inhibitor activities the initial inoculum showed more effect than shake and static conditions

Abstract

AbstractEndophytic fungi are the microorganisms that spend all or part of their life cycles within plant tissue without causing harmful effects on the plant. In this study, 14 endophytic fungus from Quercus phillyraeoides A. Gray were isolated. Alternaria sp. QPS 05, an endophytic fungi which was isolated from the stem of Q. phillyraeoides A. Gray showed the highest α-glucosidase inhibitory activity. Further separation of ethyl acetate extract from the fungus led to the isolation of active substance from hexane-soluble fraction which give fatty acids mixture consist of palmitic acid, oleic acid, linoleic acid and linolenic acid (1) strong inhibitory activity against α-glucosidase. Isolated fatty acids (1) had inhibitory concentration (IC50) values against Saccharomyces cerevisiae was 12.10 μg/mL. The results of the present study showed that endophytic fungus from Alternaria sp. QPS 05 potentially contained a rich source of natural antidiabetic medicine.
