Abstract

The research aimed at studying the effect of moisture content, package and storage period on mitochondrial inner membrane protein of soybean seed. The research was conducted in the laboratory of Seed Technology and Microbiology of Gadjah Mada University from May 2002 to August 2003. A Randomized Complete Block Design was used in the experiment with three factors and three replications. The first factor was moisture content consisted of three levels, i.e. 8%, 10%, and 12%. The second was three kinds of packages namely polyethylene plastic, wheat and aluminium foil. The third was the storage period, i.e. 0, 1, 2, 3, 4, 5 and 6 months. Data collected were analyzed by analysis of variance and Duncan Multiple Range Test. Regression and correlation analysis were done to study the correlation between germination, vigor and protein content of mitochondria inner membrane. Record was made on protein content of mitochondrial inner membrane and its electrophoresis profile, electrical conductivity, germination and vigor. The result showed that moisture content, packaging and storage period had significant influences on protein content of the mitochondria inner membrane and its electrophoresis profile. Inner membrane protein of mitochondrial had a positive correlation with germination and vigor. It was concluded that moisture content, package and storage period affected inner membrane protein content of mitochondria and its protein electrophoresis profile. The protein content of mitochondrial inner membrane, germination and vigor of soybean seed stored in aluminium foil sack at 8%, 10% and 12% moisture content did not decreased within 6 months. Cell membrane damage affected mitochondrial damage.   Key words:  Soybean seed, mitochondria inner membrane protein

Abstract

This  research was aimed to study fatty acid changes during soybean seed storage and to find method of soybean storage to maintain low free fatty acid content and high germination and vigor up to 6 months. The research was conducted at the Chemistry Laboratory of Mathematic and Natural Science Faculty, Pattimura University from May to December 2007. The experimental design used was factorial Experiment in Split- Split Plot Design, consisted of three factors, i.e. moisture content (8. 10 and 12%) as main plot, packaging materials (polyethylene, wheat and aluminium foil) as sub plot, storage period (0, 1, 2, 3, 4, 5, and 6 months) as sub-sub plot. Data were analyzed by analysis of variance and Duncan Multiple Range Test. Regression and correlation analysis were done to study the correlation between germination, vigor and free fatty acid. The results showed that fatty acid of soybean seed during storage at all levels of moisture content in wheat sack increased faster than that of other sacks. The free fatty acid content, germination and vigor of soybean seed stored in aluminium foil at all levels of  moisture content slowly decreased within 6 months. Free fatty acid content had a significant negative correlation with germination and vigor. Keywords: soybean seed, fatty acid, storage, seed viability

Abstract

The quality of soybean seed is easily decreased, so that it is difficult to keep them for a long time. The research aims to study the effect of initial moisture content, packaging material and storage period on succinate dehydrogenase and cytochrome oxidase activity. The experimental design was arranged in Factorial in RCB consisting of 3 factors: moisture content, i.e. 8. 10 and 12 percent, packaging materials, i.e. polyethylene, wheat and aluminium foil; storage period i.e. 0, 1, 2, 3, 4, 5, and 6 months. Changes in seed water content, specific activity of succinate and cytochrome oxidase, respiration rate, germination coefficient velocity germination, and abnormal seedlings were monthly determined. It was found that seed moisture content was increase and showed positive correlation with seed moisture content and abnormal seedlings and showed negative correlation with specific activity of succinate dehydrogenase and cytochrome oxidase, respiratory rate, germination and coefficient velocity germination. From this experiment, soybean seeds were stored in polyethylene and aluminium foil bags observed highly succinate dehydrogenase and cytochrome oxidase specific activity, respiration rate, germination, coefficient velocity germination and keep moisture content in low level could delay seed deterioration followed by polyethylene and wheat bags.  Key words: moisture content, succinate dehydrogenase, cytochrome oxidase, soybean
