Abstract

ABSTRAK. Medium tumbuh merupakan salah satu faktor penting dalam menentukan kualitas pertumbuhan planlet anggrek Vanda.  Pada medium tumbuh perlu ditambahkan suplemen nonsintetik yang berperan mempercepat pertumbuhan planlet anggrek Vanda.  Tujuan penelitian ialah mendapatkan suplemen nonsintetik yang tepat untuk mengganti komponen medium Vacin dan Went (VW).  Penelitian dilaksanakan di Laboratorium Kultur Jaringan, Kebun Percobaan Tanaman Hias Pasarminggu, mulai bulan Juni sampai dengan Desember 2007.  Perlakuan yang diberikan ialah sebagai berikut. (1) medium VW  sebagai kontrol; (2) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + pisang 50 g/l; (3) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + ubikayu 50 g/l; (4) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + kentang 50 g/l; dan (5) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + taoge 50 g/l. Rancangan percobaan yang digunakan ialah acak kelompok dengan lima perlakuan dan empat ulangan.  Hasil penelitian menunjukkan bahwa media (1) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + pisang 50 g/l; (2) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + ubikayu 50 g/l; (3)  KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + kentang 50 g/l; dan (4) KNO3 1 g/l + kasein hidrolisat 100 mg/l + ragi 1,25 g/l + taoge 50 g/l  dapat digunakan sebagai media alternatif pengganti medium Vacin dan Went. The Effect of Nonsynthetic Supplement on the Plantlet Growth of Vanda.  Medium is one of the important factors in determining the plantlet  growth of Vanda. Nonsynthetic supplement has to be added in to the medium to accelerate the growth of  Vanda plantlet.  The aim of this experiment was to obtain nonsynthetic supplement to substitute the component of Vacin and Went medium. The experiment was conducted at Tissue Culture Laboratory of Indonesia Ornamental Crops Experimental Garden, Pasarminggu Jakarta, from June through December 2007. The experiment was arranged in a randomized block design with five treatments and four replications. The treatments were : (1) medium VW without addition organic compound; (2) KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1.25 g/l + banana 50 g/l; (3) KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1.25 g/l + casava 50 g/l; (4) KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1.25 g/l + potato 50 g/l; and (5) KNO3 1 g/l + casein hydrolysate100 mg/l + yeast 1.25 g/l + mungbean sprout 50 g/l.  The results showed that  (1) KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1.25 g/l + banana 50 g/l; (2)  KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1.25 g/l + cassava 50 g/l; (3)  KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1,25 g/l + potato 50 g/l; and (4)  KNO3 1 g/l + casein hydrolysate 100 mg/l + yeast 1.25 g/l + mungbean sprout 50 g/l  could be used as an alternative media.  The use of nonsynthetic supplement could reduce production cost of plantlet.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian bubur ubikayu dan ubijalar terhadap pertumbuhan plantlet anggrek dendrobium.  Percobaan dilakukan pada bulan Februari sampai Juni 2001, menggunakan rancangan acak lengkap dengan lima ulangan. Hasil penelitian menunjukkan bahwa pemberian bubur ubikayu putih memberikan hasil yang sama baik dengan pisang ambon terhadap pertumbuhan tinggi plantlet, jumlah, dan luas daun. Akar plantlet pada media mengandung bubur ubikayu kuning lebih banyak dan lebih panjang dibandingkan akar plantlet pada me- dia lainnya, sedangkan pemberian ubijalar merah menghambat pertumbuhan tinggi, luas daun, jumlah, dan panjang akar. Kata kunci : Anggrek  dendrobium ; Pisang; Ubikayu; Ubijalar; Media; Bubur; Pertumbuhan plantlet. ABSTRACT. The aim of this experiment was to investigate the effect of cassava and sweet potatoes pastes incorporated in the growth medium on the growth of plantlets of dendrobium orchid. The result of research showed that addition of white cassava or banana pastes in the media, significantly increased plantlet height, leaf num- ber, and leaf size. The highest number of roots and length of root were found on the media suplemented with yellow cassava paste. Inversely, addition of red sweet potatoe paste inhibited plantlet height, leaf size, and number and length of roots.

Abstract

Penelitian ini bertujuan mengetahui pengaruh ekstrak ragi terhadap pertumbuhan plantlet anggrek dendrobium. Anggrek dendrobium merupakan salah satu jenis anggrek yang disukai konsumen. Tanaman anggrek umumnya menggunakan bibit anggrek asal kultur in vitro. Optimasi komposisi media untuk kultur in vitro sangat diperlukan untuk meningkatkan kemampuan multiplikasi maupun kualitas bibit. Salah satu cara optimasi media yaitu dengan pemberian bahan organik kompleks. Bahan penelitian ini adalah plantlet anggrek dendrobium yang ditumbuhkan dalam media Vacin & Went dengan penambahan air kelapa 150 ml/l, sukrosa 20 g/l, dan bubur pisang 50 g/l. Perlakukan-perlakuan disusun dalam rancangan acak kelompok dengan sembilan perlakuan dan tiga ulangan. Perlakuan terdiri atas penambahan ekstrak ragi ke dalam media kultur dengan konsentrasi: 0 (kontrol); 0,25; 0,50; 0,75; 1,00; 1,25; 1,50; 1,75; dan 2,00 g/l. Hasil penelitian menunjukkan bahwa pemberian ekstrak ragi dalam media kultur dapat meningkatkan pertumbuhan plantlet anggrek dendrobium. Ekstrak ragi konsentrasi 1,25 g/l merupakan konsentrasi terbaik bagi pertumbuhan plantlet dilihat dari tinggi tanaman, luas daun, panjang, dan jumlah akar. Dendrobium is one of the favourite orchid genera. The orchid plantations generally use orchid seedlings that are obtained through in vitro culture. Media optimization is a critical point for improving the capacity of seedling mul- tiplication and quality. One of the methods to enrich the medium was the use of a complex organic mixture of yeast ex- tract. The materials used in this research were dendrobium orchid plantlets that were cultivated in Vacin  & Went medium with the addition of 150 ml/l of coconut water, 20 g/l of sucrose and 50 g/l of banana pulp. The treatments were laid in a randomized block design with nine treatments and three replications. The treatments were the addition of yeast extract into the culture medium of concentrations of 0 (control); 0.25; 0.50; 0.75; 1.00; 1.25; 1.50; 1.75; and 2.00 g/l. The results  showed that the use of yeast in the culture media significantly increased growth of dendrobium plantlets. Concentration of 1.25 g/l was the best for the performance of plantlets in term of plants height, leaf size, root length and number of roots.
