Abstract

Propagation through tissue culture by using orchid seed as explants will produce a lot of orchid plants. This study aims to measure the genetic character of orchid plantlets that were regenerated from seeds which have been resulted from in vitro culture. The genetic character of the original orchid plants produced from in vitro culture was determined using Random Amplified Polymorphic DNA (RAPD) molecular markers. The results showed that the primers used in the RAPD analysis showed a polymorphic band pattern of 14 DNA bands, with sizes between 500 bp - 8000 bp. The genetic distance of Grammatophyllum scriptum orchids that was regenerated from seeds is between 0.229 and 0.649. The progenis produced from in vitro culture were clustered into seven groups at a dissimilarity coefficient of 45%.

Abstract

White oyster mushroom (Pleurotus ostreatus) is currently quite popular and much loved by the public because of its delicious taste, high protein content, and low fat. This study aims to measuring the growth of white oyster mushrooms grown on sago dregs media with the treatment of various supplements, measuring the growth power of oyster mushrooms on sago dregs media with the treatment of various kinds of supplements, and measuring the production and efficiency of white oyster mushrooms on the substrate used. This research method consisted of 12 treatments and was repeated 3 times, so that there were 36 experimental units. Research Results White oyster mushrooms grown on MS2 media composition (20 g + MS 80 ml dry sago pulp) showed a good effect on mycelium growth and harvest rapidly. The highest number of fruit bodies was produced in the A3 treatment (20 g of dry sago pulp + 100 ml of water). The highest body weight of fresh mushroom fruit was in the K3 treatment (20 g dry sago pulp + 100 ml coconut water), KN3 (20 g dry sago pulp + 100 ml KN), KN1 (20 g dry sago pulp + 60 ml KN) , and A3 (20 g of dry sago pulp + 100 ml of water). The efficiency of utilization of sago waste organic matter by white oyster mushrooms was 25.84%.

Abstract

Sago palm (Metroxylon sago Rottb.) is one of the carbohydrate-producing crops that has great potential in supporting food security programs. This study aims to determine the morphological and genetic diversity based on the mat-K gene marker of 15 accessions of sago pal that have been collected by the Sago Research Consortium. This research was conducted by preparing plant samples, extracting DNA, amplifying DNA with a PCR tools, visualizing the results of DNA amplification, sequencing, editing DNA sequencing, and blasting. The results showed that the sago palm accessions collected by the UNIPA Sago Research Consortium were morphologically different in the Russet stages. Based on the maturase K (mat-K) gene marker of 15 sago palm accessions were divided into two haplotypes, namely haplotype 1 which experienced deletion at base number 5 (amino acid phenylalanine) and haplotype 2 which did not experience deletion so it had phenylalanine amino acid. Phylogenetic analysis showed that 15 sago palm accessions were divided into 2 groups, namely group 1 and group 2. Sago palm were observed closely related to Metroxylon warburgii with genetic distance of 0.001.

Abstract

Sago palm need to be cultivated intensively for increasing carbohydrate resources to anticipate the deficiency of energy sources of the world. This study aims to measure the influences of sago dregs to the sag palm growth in the field. The experimental method used in this works was Randomized Block Design that consisting of five treatments sago dregs weights in the russet stages of sago palm. The composition of the treatment is follows: without sago dregs (P0), 5 kg sago dregs tree-1 (P1), 10 kg of sago dregs tree-1 (P2), 15 kg sago dregs tree-1 (P3), 20 kg sago dregs tree-1 (P4). The results showed that sago dregs had a significant effect of the spear leaves growth, but not significantly effect of the plant height growth, number of leaflets growth, and number of leaflets. Utilization of 10 kg sago dregs tree-1 as mulch was calculated influence of the spear leaf growth and utilization of 20 kg sago dregs as mulch inhibitory of sago palm growth.
