Abstract

Embryonic Stem Cells (ESC) are pluripotent stem cells which has the ablility to self renew and differentiate into form all cells in the body. Inner Cell Mass (ICM) as a source of ESC that can be obtained from blastocyst stage of embryos. Isolation can be done by several methods such as mechanical, enzymatic and immunosurgery. This study aimed to observe the effectiveness of usage immunosurgery method to obtain the ICM from the blastocyst stage of embryo. Blastocyst stage of embryos were obtained from a strain of Swiss Webster female mice that had been stimulated using pregnant mare s gonadotropin (PMSG) and human chorionic gonadotropin (hCG). Inner Cell Mass (ICM) were cultured and observed of the attachment level (attachment rate/AR), the rate of primary colony formation (primary colony/PC) and the morphology of ESC line. The results showed that the ICM were isolated using methods immunosurgery have AR and PC respectively 78,57% and 71,00%. Embryonic Stem Cell (ESC) can differentiate after several passages by forming embryoid body (EB). This study shows that immunosurgery is an effective method for producing ESC line from mice embryos of blatocyst stage.Key words: Blastocyst, Inner cell mass, ICM, ESC AbstrakEmbryonic Stem Cell (ESC) merupakan sumber stem cell yang bersifat pluripoten, yaitu sel yang mampu membelah dan berdiferensiasi menjadi semua tipe sel di dalam tubuh. Inner Cell Mass (ICM) sebagai sumber ESC dapat diperoleh dari embrio tahap blastosis. Isolasi ICM dapat dilakukan dengan beberapa metode seperti mekanik, enzimatik dan immunosurgery. Penelitian ini bertujuan mengamati efektifitas penggunanaan metode immunosurgery untuk memperoleh ICM dari embrio tahap blastosis. Embrio tahap blastosis diperoleh dari mencit betina strain Swiss Webster yang telah distimulasi menggunakan pregnant mare’s gonadotropin (PMSG) dan human chorionic gonadotropin (hCG). Inner Cell Mass (ICM) dikultur dan dilakukan pengamatan terhadap tingkat perlekatan ICM (attachment rate/AR) serta tingkat pembentukan koloni primer ESC (primary colony/PC) serta morfologi ESC line yang terbentuk. Hasil menunjukkan bahwa ICM yang diisolasi menggunakan metode immunosurgery memiliki AR dan PC masing-masing 78,57% dan 71,00%. Embryonic Stem Cell (ESC) dapat mengalami diferensiasi setelah beberapa pasase dengan membentuk embryoid body (EB). Penelitian ini menunjukkan bahwa metode immunosurgery merupakan metode yang efektif untuk memproduksi ESC line dari embrio mencit tahap blatosis.Kata kunci : Blastosis, Inner cell mass, ICM, ESC

Abstract

Hand, foot, and mouth disease (HFMD) is a common disease caused by human enterovirus (HEV). The symptoms and signs of infection among children typically present with vesicular exanthema on the soles of their feet, the palms of their hands and in their mouths, causing discomfort and feeding difficulties. The disease is caused by mainly Coxsackie virus A16 (CVA16) serotype. It has recently been associated with EV-71 serotype which can also cause outbreak and mortality. Currently the data of HFMD cases in Indonesia is not available yet therefore this study aims to identify and determine the distribution of HFMD cases in Indonesia from the outbreak. HFMD can be identified by RT-PCR that recognize PAN-EV gene in the first place and has to be continued to EV-71 in VP1 area. Besides using PCR, we use isolation method using tissue cultures which are RD and 293 cell linesrom. From 48 suspected cases were received in virology lab CBBTH, NIHRD. 26 out of 48 cases (54%) were caused by enterovirus and 3 out of 26 were EV-71 (6,25%). The HFMD cases distribute in all group of ages and sex but children under five is the most susceptible.Key words: HFMD, Enterovirus, Outbreak, EV71 AbstrakHand Foot and Mouth Diseases (HFMD) atau dalam bahasa Indonesianya dikenal sebagai penyakit Kaki Tangan Kuku dan Mulut. Penyakit ini umumnya menyerang anak-anak dan menimbulkan gejala yang khas seperti terbentuknya vesikula di telapak tangan, kaki dan di rongga mulut sehingga menimbulkan rasa tidak nyaman dan susah menelan. Penyebab Utama penyakit ini adalah enterovirus teruma Coxsackie A16. Belakangan ini penyakit HFMD juga berkaitan dengan EV-71 yang bisa menimbulkan Kejadian Luar Biasa dan kematian. Saat ini belum tersedia data penyebab kasus HFMD di Indonesia, oleh karna itu penelitian ini bertujuan untuk mengetahui dan mengidentifikasi penyebab kasus HFMD di Indonesia dari kasus kejadian luar biasa. Identifikasi penyebab penyakit HFMD bisa dilakukan dengan menggunakan methode PCR dengan mengidentifikasi adanya gen Pan EV terlebih dahulu dan dilanjutkan dengan EV-71 spesifik di area VP1. Isolasi virus juga dilakukan dengan mengkultur virus di sel RD dan 293 untuk tujuan identifikasi dan memperbanyak serta memurnikan virus dari spesimen asli. Dari 48 kasus yang diterima laboratorium Virologi Pusat BTDK, Badan Litbang Jakarta, diketahui 26 kasus (54%) disebabkan oleh enterovirus dan 3 diantaranya adalah EV-71 (6.25%). Kasus HFMD dapat terjadi pada semua golongan umur dan jenis kelamin, akan tetapi anak-anak yang berumur antara 1-5 tahun sangat rentan menderita penyakit ini.Kata kunci : HFMD, Enterovirus, KLB, EV71

Abstract

This study aims to determine the persuasive communication strategy of kindergarten teachers "Harapan Bersama" in the character of caring for waste. This study uses the theory of The Meaning Construction Communication Strategy which includes Cognitive Factors and Emotional Factors. And the type of research used is qualitative research. The results of this study indicate that the important role of cognitive and emotional factors in education in the Harapan Bersama Kindergarten environment is that the programs implemented both by teachers at school and parental support at home have had a positive impact in shaping children's understanding and behavior related to caring for waste in their environment. Children's responses suggest that different approaches may be needed to meet individual needs in terms of environmental understanding. This emphasizes the importance of a holistic and diverse approach to environmental education to achieve broader and more integrated outcomes. There were differences in how children responded to environmental issues based on their emotional and cognitive factors, highlighting the complexity of shaping attitudes towards the environment. The results of this study show that persuasive communication strategies through cognitive factors and emotional factors have a positive impact in shaping children's understanding and behavior related to waste care in their environment.
