Abstract

Abstract. Infectious diseases are a major public health problem for developed and developing countries. One of the causes of infection is bacteria. Black bangle contains chemical compounds with pharmacological effects that have potential as antibacterial. This study aims to determine the potential of black bangle rhizome (Zingiber ottensii (Val.)) as an antibacterial and to determine the value of Minimum Inhibitory Concentration (MIC) on the growth of Staphylococcus aureus bacteria. The research was conducted experimentally in a laboratory with the stages of collecting materials, determining plants, making simplicia, standardizing simplicia, screening for phytochemical simplicia and extracts, extracting, testing antibacterial activity, and determining the value of Minimum Inhibitory Concentration (MIC). Antibacterial activity test by agar diffusion method with well with positive control group (tetracycline), negative control (methanol), and test group (extract at concentrations of 40%, 20%, 10%, and 5% w/v). Based on the research conducted, the results showed that the methanol extract of black bangle rhizome with a concentration of 20% and 40% had antibacterial activity with an inhibitory diameter of each of 6.641±0.182 mm and 9.967±0.396 mm respectively. The value of the Minimum Inhibitory Concentration (MIC) was determined using the agar dilution method with a positive control group (media with Staphylococcus aureus bacteria), negative control (media), and a test group (extract at a concentration of 20%, 18%, 16%, 14%, 12%, and 10% w/v). Minimum Inhibitory Concentration (MIC) value set is at a concentration of 18%. Abstrak. Infeksi merupakan salah satu penyakit dengan penderita terbanyak di negara maju maupun negara berkembang. Terjadinya infeksi dapat disebabkan karena adanya bakteri. Bangle hitam memiliki kandungan senyawa kimia dengan efek farmakologis yang berpotensi sebagai antibakteri. Penelitian ini memiliki tujuan untuk mengetahui potensi rimpang bangle hitam (Zingiber ottensii (Val.)) sebagai antibakteri dan menentukan nilai Konsentrasi Hambat Minimum (KHM) terhadap pertumbuhan bakteri Staphylococcus aureus. Penelitian dilakukan secara eksperimental laboratorium dengan tahapan kerja pengumpulan bahan, determinasi tanaman, pembuatan simplisia, standardisasi simplisia, skrining fitokimia simplisia dan ekstrak, ekstraksi, uji aktivitas antibakteri, serta penentuan nilai Konsentrasi Hambat Minimum (KHM). Uji aktivitas antibakteri menggunakan metode difusi agar cara sumuran dengan kelompok kontrol positif (tetrasiklin), kontrol negatif (metanol), dan kelompok uji (ekstrak pada konsentrasi 40%, 20%, 10%, dan 5% b/v). Berdasarkan penelitian yang telah dilakukan, didapatkan hasil yaitu ekstrak metanol rimpang bangle hitam dengan konsentrasi 20% dan 40% memiliki aktivitas antibakteri dengan diameter hambat masing-masing sebesar 6,641±0,182 mm dan 9,967±0,396 mm. Nilai Konsentrasi Hambat Minimum (KHM) ditentukan menggunakan metode dilusi agar dengan kelompok kontrol positif (media yang ditumbuhi bakteri Staphylococcus aureus), kontrol negatif (media), dan kelompok uji (ekstrak pada konsentrasi 20%, 18%, 16%, 14%, 12% dan 10% b/v). Nilai Konsentrasi Hambat Minimum yang ditetapkan ada pada konsentrasi 18%.
