Abstract

Penelitian ini dirancang untuk melihat pengaruh aerasi dan kepadatan tempat hidup sebagai sumber stres terhadap pertumbuhan ikan nila merah (Oreochromis niloticus). Rancangan penelitian yang digunakan pada penelitian ini adalah Rancangan Acak Lengkap (RAL) faktorial  dengan dua perlakuan, terdiri atas perlakuan dilengkapi dengan satu aerator dan perlakuan dolengkapi dengan dua aerator serta tiga tingkatan padat tebar 4,6,8 ekor ikan. Ikan nila merah dengan berat 4±0,5 g dipelihara dalam kontainer boks kapasitas 15 liter dengan volume air 8 liter. Ikan dipelihara selama 30 hari.  Variabel yang diamati adalah bobot ikan, Panjang dan tinggi ikan, serta bobot karkas dan kandungan protein daging ikan. Faktor lingkungan yang diamati meliputi Oksigen terlarut (DO), pH, suhu, kandungan ammonia, nitrit dan nitrat. Analisis data dikerjakan menggunakan analisis sidik ragam (ANOVA) dua arah. Analisis data dikerjakan dengan program SPSS versi 23. Perbedaan yang nyata antar perlakuan diuji dengan uji Beda NyataTerkecil (BNT). Beda nyata dievaluasi pada taraf P < 0.05. Hasil penelitian menunjukkan peningkatan padat tebar sampai dengan dua kali lipat kapasitas normal dengan menggunakan satu aerator sebagai sumber oksigen terlarut menghambat pertumbuhan dan menurunkan bobot karkas dan kandungan protein daging ikan. Pada penelitian ini dapat disimpulkan bahwa padat tebar ikan masih bisa ditingkatkan sampai dengan dua kali lipat kapasitas normal dengan penambahan aerator sebagai peningkatan oksigen terlarut.This study was designed to examine the effect of aeration and density of living space as a source of stress on the growth of red tilapia (Oreochromis niloticus). The research design used in this study was a factorial Completely Randomized Design (CRD) with two treatments, consisting of treatment equipped with one aerator and treatment equipped with two aerator and three levels of stocking density of 4,6,8 fish. Red tilapia weighing 4±0.5 g was reared in a box container with a capacity of 15 liters with a volume of 8 liters of water. Fish were kept for 30 days. The variables observed were length and height of the fish,carcass weight and protein content of fish meat. Environmental factors observed included dissolved oxygen (DO), pH, temperature, ammonia, nitrite and nitrate content. Data analysis was carried out using two way analysis of variance (ANOVA). Data analysis was carried out using the SPSS version 23 program. Significant differences between treatments were tested using Least Significant Difference test (LSD). Significant differences were evaluated at the level of P < 0.05. The results showed an increase in stocking density up to twice the normal capacity by using an aerator as a source of dissolved oxygen inhibiting growth and reducing carcass weight and protein content of fish meat. In this study it can be concluded that the stocking density of fish can still be increased up to twice the normal capacity with the addition of an aerator as an increase in dissolved oxygen.

Abstract

Salah satu usaha kerajinan logam yang berada di Desa Pasarean Kecamatan Adiwerna Kabuaten Tegal, yaitu UD Berkah Lestari adalah usaha yang bergerak di bidang kerajinan olahan logam, yakni Dandang. Permintaan akan produk Dandang di pasaran sangat besar. Produksi Dandang yang berlebihan akan menimbulkan penumpukan produk yang berakibat menurunkan kualitas barang sedangkan produksi Dandang yang sedikit akan tidak terpenuhinya permintaan pasar sehingga dapat kehilangan pelanggan.Permasalahan yang dihadapi oleh UD Berkah Lestari adalah bagaimana memanajemen inventory (persediaan) Dandang sehingga terjadi keseimbangan antara produksi dengan permintaan akan Dandang. Permintaan produk tetap terpenuhi dengan biaya yang dikelurkan seminimal mungkin sehingga dapat meningkatkan keuntungan.Kegiatan ini bertujuan untuk memberikan masukan akan manajemen inventory suatu usaha dengan biaya operasional yang dikeluarkan seminimal mungkin dan keuntungan yang didapatkan semaksimal mungkin melalui seberapa besar produk optimal yang harus diproduksi.Metode yang digunakan untuk mencapai tujuan adalah mengelompokan masalah dalam aspek bidang yang kemudian dipilih berdasarkan prioritas permasalahan serta IPTEKS yang diterapkan untuk menyelesaikan permasalahan prioritas. Abstract

Traktus alimentarius merupakan saluran pencernaan yang digunakan dalam rangka penyederhanaan makanan melalui mekanisme fisik dan kimiawi. Peningkatan pertumbuhan organ cerna ini merupakan bentuk pengoptimalisasian fungsi fisiologis organ tubuh ikan berdasarkan pengendalian faktor lingkungan dengan pemberian aerasi dan filter. Penelitian ini bertujuan untuk menganalisis penggunaan aerasi, filter, serta kombinasi aerasi dan filter pada morfometri traktus alimentarius ikan nila merah (Oreochromis niloticus). Penelitian dilakukan selama 60 hari dengan proses awal aklimatisasi. Hewan uji dipelihara dengan memperhatikan pakan dan faktor lingkungan. Dilakukan pengukuran panjang dan bobot  awal, serta bobot akhir total maupun kompartemen pada traktus alimentarius. Data penelitian kemudian dianalisis dengan uji Anova dan uji BNT menggunakan aplikasi spss release 26. Hasil menunjukkan bahwa aerasi dan filter memberikan pengaruh yang nyata (P<0,05) terhadap panjang dan berat traktus alimentarius ikan. Kesimpulan penelitian ini bahwa penggunaan aerator ganda dilengkapi dengan filter memberikan dampak positif terhadap pertambahan panjang dan bobot traktus alimentarius.The alimentarius tract is a digestive tract that is used to simplify food through physical and chemical mechanisms. This increase in the growth of digestive organs is a form of optimizing the physiological functions of fish organs based on controlling environmental factors by providing aeration and filters. This study aims to analyze the use of aeration, filter, and combination of aeration and filter on the morphometry of the tract alimentarius of red tilapia (Oreochromis niloticus). The study was conducted for 60 days with the initial process of acclimatization. The test animals were kept by paying attention to feed and environmental factors. Measurements of length and initial weight, as well as total final weight and compartments in the alimentarius tract were carried out. The research data were then analyzed by Anova test and BNT test using spss release 26 application. The results showed that aeration and filter had a significant effect (P<0.05) on the length and weight of the alimentarius tract of fish. The conclusion of this study is that the use of double aerators equipped with filters has a positive impact on the increase in length and weight of the alimentarius tract.

Abstract

Physiological stress due to nutritional deficiency accompanied by excessive activity can cause problems of energy deficiency required by the body. Energy shortage can affect the body weight, as well as the weight and histology of the liver. This study was conducted to examine and analyze the effect of snakehead fish meat supplementation in feed toward body weight, weight and hepatic histology in animals experiencing physiological stress. This study used 20 male Wistar rats consisting of 5 treatments with 4 replications. The treatment in this study consisted of control (P0) and treatment (P1: 5%, P2: 10%, P3: 15%, P4: 20%) groups. This research used Completely Randomized Design (RAL). The variables measured in this study were body weight, hepatic weight, central venous diameter and hepatocytes in the test animals. The data obtained were analyzed using Analysis of Variance (ANOVA) at 5% significance level and continued with Duncan test with 5% significance. The result of data analysis showed that fish snakehead fish supplementation in feed gave a significant effect to body weight, weight and histology of animal liver. Concentrations of snakehead fish supplements can increase the body weight, hepatic weight, and hepatic histology which was indicated by higher central venous and hepatocyte diameter values compared to other controls and treatments. The conclusion of this research is the provision of snakehead fish supplement (Channia striata) can increase the body weight, hepatic weight, central venous and hepatocytes diameter of Wistar rats with the most influential concentration of 20%. Abstract

The world is facing the challenge of the COVID-19 disease, which is now stated as a pandemic. Inside the host cell, spike envelope protein (spike) of SARS-CoV-2 interact with the Angiotensin-converting Enzyme 2 (ACE2) receptor. It can be inhibited by bioactive compounds such as flavonoids which have anti-viral and broad pharmacological effect. This study aimed to determine the spike protein inhibitory activity by quercetin against the ACE2 receptor using the molecular docking method. This study focused on the inhibitory of the penetration activity of s proteins in ACE2 by utilizing natural material compounds that have the potential to be used as anti-SARS-CoV-2 drug development agents. The flavonoid compound quercetin was extracted by the maceration method. The quantitative analysis was carried out using a UV-Vis spectrophotometer to prove the presence of quercetin content. Molecular docking simulations were carried out to look for the binding affinity between the spike protein and quercetin. Docking was carried out using the Autodock, PyRx, and visualization using Discovery Studio. Indicators that prove that quercetin forms binding affinity and protein complexes with spikes are the Vina Score and RMSD. It is also supported by ADME analysis and conformity to Lipinski's rules of five. That matter becomes a success indicator of the spike activity inhibition by quercetin, which makes it possible to be used as an anti-SARS-CoV-2 drug development agent. The novelty from this study is molecular docking method that used to show that quercetin in red onion had inhibitory activity on the penetration process of protein spike in SARS-CoV-2. Results obtained from this study can be used as a recommendation for advanced research in invitro and invivo studies as a drug which has potential to inhibit protein spike of SARS-CoV-2.

Abstract

Physiological stress for a long time can lead to impaired kidney organ function. Snakehead fish meat are known to contain important nutrients such as albumin, glutamine, glycine, cysteine, and various minerals that can improve kidney function. This study was conducted to examine and analyze the effect of snakehead fish supplementation in feed towards body weight, kidney weight, diameter and number of glomerulus in Wistar rats with physiological stress. This study used 20 male Wistar rats divided into 5 groups consisted of 1 control group (P0) and 4 treatment groups (P1: 5%, P2: 10%, P3: 15%, P4: 20%). The treatments were performed with 4 times repetition. This study used Completely Randomized Design (CRD). The variables measured were body weight, kidney weight, diameter and number of renal glomerulus. The data obtained were analyzed using Analysis of Variance (ANOVA) at 5% significance level and continued with Duncan test with 5% significance level. The results of this study were: the supplementation of snakehead fish with 20% of concentration can increase the body weight; concentration 10; 15; and 20% can increase the weight of the kidney; concentrations of 5%, 10%, 15%, and 20% can increase the glomerular diameter in Wistar rats with physiological stress. The supplementation of snakehead fish with concentration of 5% to 20% did not affect the number of glomerulus of Wistar rats with physiological stress. Results of this study is useful as information for people who study the mechanism of kidney repair due to physiological stress.

Abstract

Flora biodiversity has been known to have a vital function in reducing carbon emissions in the air and contributing oxygen in significant levels to the environment. PT. Pertamina (Persero) Fuel Terminal Boyolali (FTB) has an important role in the conservation programs, protection of flora biodiversity, and environmental management. This study aims to analyze carbon stocks found in trees, plants cover soil, litter, necromas, and carbon in the soil and evaluate conservation of flora. This study uses a purposive random sampling method of carbon sources followed by the determination of carbon stocks in three conservation areas managed by PT. FTB, namely in the Wonopotro Forest, in the Pepe River watershed, and Pertamina's operational area. The samples from carbon sources include trees, litter, ground cover plants, necromas, and soil. The trees and necromas carbon sources sampling at each location was carried out at 2 stations with a size of 10 x 50 m2. Meanwhile, the sampling of litter, ground cover plants, and soil was carried out at 2 stations, each consisting of 3 plots. The results showed that the total carbon stock in the conservation areas was 1,688,0791 tons. The Pepe River watershed has the highest total carbon stock, which was 1,466,0928 tons/ha. The FTB and Wonopotro Forest, respectively, have about 180.7569 and 41.2294 tons/ha. The novelty of this research is the method of calculating carbon stocks in a conservation area that can be used as a database for assessing environmental management performance to the preservation of biological resources.

Abstract

Cholesterol in quail meat is one of the factors causing consumers to control consumption of quail meat. On the other hand, the antioxidant content in quail meat could attracts the consumers. Cinnamon (Cinnamomum sp.) bark and gotu kola (Centella asiatica) leaves are medicinal plants that contain many polyphenol compounds. The objective of this study was to obtain the most optimum formula made from cinnamon and gotu kola as a feed supplement to produce the quail meat which has low cholesterol but rich in antioxidants. This study used a completely randomized design which consisting of six groups with three replications. The six groups included one control (P0), 5 treatments consisted of 5% cinnamon (P1), 5% gotu kola (P2), combination of cinnamon and gotu kola powder with ratio 5%:5% (P3), 5%:10% (P4) and 10%:5% (P5).  The results showed that food supplements made from cinnamon bark and gotu kola leaves could increase the body weight, antioxidant levels, and reduce cholesterol levels in quail meat. Cinnamon as feed supplement with a concentration of 5% or a combination of cinnamon and gotu kola with a ratio of 5%: 10% provided the best effect on antioxidants and cholesterol level in meat. The use of cinnamon and gotu kola supplementation to produce high antioxidant and low cholesterol of quail met have not studied yet. The result of the study would be beneficial for developing the healthy, safe and good quality of quail meat.

Abstract

Anthrax is a deadly disease caused by Bacillus antracis that damages the lymphatic and vascular systems of animals and humans. Anthrax prevention in Indonesia has been done but has many shortcomings among them is a lot of dead animal raises after vaccination and relatively expensive. The azadirachtin bioactive compound produced by neem endophytic microbes is known to inhibit the growth of B. anthracis bacteria, thus potentially as an anti-anthrax agent. Other bacteria that have the same pathogens as B. antrachis are Methycillin Resistant Staphylococcus aureus (MRSA) so that it is used as a research object. The aim of this research is to obtain the active biomaterials from selected neem leaf endophytes to be tested in S. aureus MRSA as bacteria with Anthrax pathogens. The method used in this research is the isolation, selection, and extraction of Neem  endophytes. The process of isolation is done by growing on the general media and the selection process on special media. The bioactive extraction process uses 1000 rpm centrifuge and screening on MRSA culture medium. Inhibitory zone tests were used to determine the effect of endophytic biopsy in inhibiting MRSA growth. The result of the research is that the bioactive endophytes of neem are able to inhibit MRSA growth which is characterized by the formation of drag zone around the endophytic bioactive. Thus it can be concluded that the use of neem endophytes can be a new solution to eradicate the presence of anthrax disease in the livestock and people in Indonesia with safe research methods, relatively inexpensive and ineffective vaccine replacements.
