Abstract

Pumpkin fruit (Cucurbita moschata. D) belongs the Cucurbitaceae family which is a functional vegetable widely distributed in Indonesia because it has nutritional value and health benefits. This study aims to determine the value of total flavonoid content and antioxidant activity in the ethanol extract of unripe, mature and ripened pumpkin. Simplicia powder was extracted by maceration method using 96% solvent. Testing the total flavonoid content with the addition of AlCl3 at a wavelength of 425 nm and the antioxidant activity test was carried out using DPPH (1-1-diphenyl-2-picrylhydrazyl) as a free radical with a maximum wavelength of 515 nm using microplate reader. The results of the study concluded that the ethanolic extract of pumpkin flesh had a total flavonoid content of 0,146 mgQE/100g in unripe fruit, 0,221 mgQE/100g in mature pumpkin, 0,191 mgQE/100g in ripened pumpkin. The antioxidant activity of the ethanol extract of unripe, mature, and ripened pumpkin fruit obtained was not active or did not completely reduce free radicals.

Abstract

Crop cabbage worm (Crocidolomia pavonana) is one of the pests that has a bad effect on the cabbage production in Indonesia. One of the efforts that has been applied to control this pest is by using bayrusil. However, the bayrusil can cause negative effects such as poisoning and environmentally polluting effects. To prevent the undesired effects, it has been developed the use of environmentally friendly insecticide and one of them is the use of insecticidal peptide. Tripsin-modulating oostatic factor (TMOF) and ist analogues is one of the peptides that has an insecticidal activity. The aim of this research is to synthesise a linear tetrapeptide (DPAP) an analogues of TMOF and to test insecticidal activities against crop cabbage worm. A DPAP has been synthesised by Solid Phase Peptide Synthesis on 2-chlorotrytil chloride resin. Fmoc strategy was applied on the synthesis and a combination DIC/Oxime was employed in the coupling reaction.The tetrapeptidyl resin was cleaved by TFA:water:EDT (90:5:5). The peptide was purified by reverse-phase column chromatography and characterized by TOF ES-MS spectroscopy. A linear peptide was biologically tested towards the C. pavonana, showing percentage of mortality values at 1000 ppm.

Abstract

Antioxidant pentapeptides are pentapeptide compounds that have antioxidant activity. One of the pentapeptide compounds that have antioxidant activity is FWKVV. FWKVV is a linear pentapeptide with the amino acid sequence phenylalanine-tryptophan-lysine-valine-valine, which was first isolated to hydrolyzate the muscle protein of Miiuy croaker (Miichthysmiiuy). In addition to isolation, FWKVV compounds can be produced by the peptide synthesis method because this method requires a shorter time than the isolation method from natural materials. Synthesis methods commonly used are solution-phase peptide synthesis and solid-phase peptide synthesis (SPPS). However, the SPSS method is more efficient because it does not require purification in every process. The purpose of this study was to synthesize FWKVV compounds using the SPPS method and test their antioxidant activity. FWKVV has been synthesized using the SPPS method with HBTU/HOBt coupling reagent and Fmoc protective group. The FWKVV crud produced was 148.8 mg and had antioxidant activity against DPPH radicals with an IC50 value of 4.2 mg/mL.

Abstract

Limbah kotoran sapi adalah sumberdaya peternak yang belum termanfaatkan. Peternak umumnya mengatasi limbah kotoran sapi dengan cara membuang ke sungai atau membakar limbah kotoran sapi. Dua hal tersebut dapat menimbulkan permasalahan lingkungan yang kompleks, dengan demikian diperlukan solusi untuk menangani hal tersebut yaitu dengan cara mengolah limbah kotoran sapi menjadi barang yang memiliki manfaat. Salah satu solusinya yaitu mengolah limbah kotoran sapi menjadi pupuk kandang menggunakan bahan Ni35 dimana bahan ini dapat mempercepat dekomposisi limbah padat kotoran sapi menjadi pupuk kandang. Metode yang digunakan dalam pengabdian ini dimulai dari penyuluhan, pelatihan, monitoring, dan evaluasi pelaksanaan produksi pupuk dan hasil uji pada tanaman kangkung dan cabai. Hasil pelaksanaan pengabdian menunjukkan bahwa peserta sangat antusias dalam mengikuti serangkaian kegiatan, serta dapat memproduksi pupuk kandang dengan kemampuan yang dapat bersaing dengan pupuk kandang konveksional/komersil. Cow manure is an underutilized resource among farmers. Generally, farmers address cow manure waste by discarding it into rivers or burning it. Both practices can lead to complex environmental problems. Thus, solutions are needed to manage this issue by transforming cow manure into valuable products. One solution involves processing cow manure into organic fertilizer using a substance called Ni35, which accelerates the decomposition of solid cow manure into usable compost. The method used in this community engagement project includes outreach, training, and monitoring, along with evaluating the production process and testing the fertilizer's effects on plants such as water spinach and chili. The results of this initiative show that participants were highly enthusiastic about the activities and successfully produced compost with competitive quality comparable to conventional commercial fertilizers.

Abstract

Koro kratok adalah kacang lokal di wilayah Jawa Timur yang memiliki keunggulan dikembangkan sebagai olahan sebagai olahan pangan tempe karena tinggi protein dan protein lemak. Tempe sudah dikenal sebagai pangan harian dalam perbaikan sindrom metabolik. Penambahan kunyit bubuk dalam tempe koro kratok diharapkan mampu meningkatkan nilai fungsionalnya. Jenis olahan tempe yang beracun telah ditemukan seperti tempe bongkrek dari ampas kelapa. Sementara itu asam sianida pada koro kratok perlu diteliti keamanannya saat menjadi tempe. Tujuan penelitian ini untuk mengidentifikasi keamanan tempe koro kratok kunyit pada uji toksisitas akut dengan paramater hepar mencit menggunakan metode toksisitas fixed dose. Koro kratok ditambahkan bubuk kunyit di fermentasi selama 24 jam an-aerob dan 4 jam aerob. Sampel tempe koro kratok dilarutkan dalam aquades hingga homogen, kemudian dilanjutkan uji pendahuluan dan uji utama toksisitas akut. Dosis tempe koro kratok kunyit 300 mg/kg BB dilakukan pada uji pendahuluan. Setelah 24 jam hasil menunjukkan tidak terdapat gejala toksik, kemudian dosis ditingkatkan menjadi 2000 mg/kg BB. Pengamatan dilakukan selama 14 hari. Pada hari ke-15, mencit dikorbankan dan dilakukan pemeriksaan histopatologi. Berat badan mengalami perbedaan signifikan hari ke-14 dosis 2000 mg/kg. Berat organ absolut dan relatif juga tidak ada perubahan makroskopis. Hasil SGOT dan SGPT dalam batas normal serta menunjukkan perbaikan signifikan terhadap kontrol mencit. Secara keseluruhan tempe koro kratok dengan penambahan kunyit bubuk LD50 >2000-5000 mg/kg tidak menyebabkan toksisitas.
