Abstract

N‐acetylglucosamine (NAG) is the monomer product of chitin, which has been widely used as a bioactive com‐ pound in applications such as anti‐tumor, anti‐microbial, and antioxidant activities. In production, biological processes using enzymes are preferable to chemicals due to environmental issues. This study aims to determine the activity, purity level, and molecular weight of purified chitinase from Micromonospora sp. AR17 determines the concentration of NAG produced by purified chitinase that has been characterized. Chitinase was produced by fermentation in colloidal chitin broth at 40 °C, pH 7, for 7 days, while chitinase activity was checked every 24 h. The optimal fermentation time was used to produce chitinase for a further purification step. Enzyme purification was carried out by ultrafiltration, ammonium sulfate precipitation, ion exchange chromatography (Q Sepharose Fast Flow), and gel filtration (Sephacryl S‐300). The purified enzyme was then char‐ acterized for optimum time, pH, and temperature to produce NAG. The results suggested that the fourth day was the optimal time for chitinase production, with chitinase activity of 0.0040 U/mL and a NAG concentration of 7.62 µg/mL. The purifica‐ tion step successfully increased the purity by 6.82 times with chitinase‐specific activity at 1.4648 U/mg. Production of NAG with purified chitinase produced a NAG concentration of 32.472 µg/mL with an incubation time of 30 min at 40 °C and pH 7.

Abstract

The probiotic innovation training programme based on market waste implemented at the Krido Yuwono Mina Padi Technopark Group aims to increase tilapia productivity and farming efficiency through the use of organic waste as raw material for probiotics. This training involves participants in the production of probiotics through the fermentation of market waste using lactic acid bacteria starters. The evaluation results show an increase in tilapia productivity of around 15-20% with better feed efficiency and a reduction in ammonia levels in pond water, which was previously a major problem in maintaining fish health. The community involved in this programme also increased their confidence in managing tilapia farming efficiently and received new knowledge that can reduce operational costs. This programme has achieved significant social and economic impacts and is expected to develop into a sustainable community empowerment model in the field of aquaculture and support more environmentally friendly waste management.

Abstract

Panembangan Village has strong potential for rice–fish culture (minapadi) but faces constraints in feeding efficiency, unmonitored water quality, and dependence on grid electricity. This community‐service program offers an integrated solution: an IoT-based auto feeder combined with real-time water-quality monitoring (DO, pH, temperature, conductivity/TDS, turbidity) and powered by off-grid solar energy (PV) to ensure continuous operation. The primary goal was to improve feed efficiency, enhance fish growth, reduce feed waste, and optimize water quality management in aquaculture systems. The implementation of these technologies has demonstrated significant improvements in feed conversion ratios, growth rates of fish, and overall water quality management. The program also provided training to local farmers, enhancing their capacity to manage these technologies effectively. This initiative is expected to contribute to the sustainable development of aquaculture in the region, benefiting both the local community and the environment

Abstract

UMKM olahan ikan di Desa Panembangan menghadapi hambatan kemasan dan pelabelan yang membatasi akses ke ritel modern serta pemasaran digital. Poklaksar Ummi Mandiri mengolah hasil samping lele menjadi kerupuk tulang ikan, namun kemasan lama (plastik bening, informasi terbatas) menurunkan daya saing. Kegiatan pengabdian dirancang untuk memperbaiki mutu kemasan, menyusun panel gizi berbasis uji laboratorium, dan memperkuat pemasaran. Intervensi diharapkan meningkatkan persepsi kualitas, keterbacaan informasi, dan penerimaan pasar. Tujuannya adalah meningkatkan kualitas desain dan fungsi kemasan agar sesuai standar pasar serta menyediakan label gizi akurat sebagai dasar transparansi dan penetrasi ritel. Metode meliputi peninjauan kebutuhan, lokakarya desain kemasan, uji gizi proksimat, penyusunan panel gizi per 100 g, pelatihan barcode/GTIN dan promosi digital, serta monitoring & evaluation melalui survei pra–pasca dan uji pasar A/B shelf test sederhana. Hasil perbaikan kemasan menjadi standing pouch paper metalize menunjukkan kenaikan skor pada enam aspek kemasan (visual, warna, tipografi, labeling, keunikan, fungsionalitas), tersusunnya panel gizi dan kemasan baru, serta peningkatan kesiapan ritel. Uji harga merekomendasikan penyesuaian dari Rp11.800/100 g menjadi Rp14.000–Rp15.000/100 g, sejalan dengan nilai tambah kemasan dan informasi gizi. Disimpulkan bahwa perbaikan kemasan, label gizi, dan strategi pemasaran berhasil menjawab tujuan program dan memperkuat daya saing produk. Model pelatihan–pendampingan kegiatan ini dapat direplikasi sebagai percontohan bagi Poklaksar lain.

Abstract

Cassava opak is a traditional snack that is widely favored by consumers; however, it has a low protein content. Fortification with parang-parang fish (Chirocentrus dorab) meat has the potential to improve product quality and consumer acceptance. This study aimed to evaluate the effect of adding parang-parang fish meat on the sensory characteristics and swelling power of cassava opak, as well as to determine the optimum concentration that produces the highest consumer acceptance. The study employed an experimental method with three levels of fish meat addition, namely P1 (10 g), P2 (20 g), and P3 (30 g), with P0 serving as the control treatment. The parameters observed included appearance, aroma, texture, taste, and swelling power. Sensory evaluation was conducted using a hedonic test involving 25 semi-trained panelists. Data were analyzed using One-Way Analysis of Variance (ANOVA), followed by Duncan’s Multiple Range Test (DMRT). The results showed that the addition of parang-parang fish meat had a significant effect (p<0.05) on the sensory characteristics and swelling power of cassava opak. Treatment P1 produced the highest preference scores for appearance and taste, while aroma preference increased with increasing fish meat concentration. Swelling power decreased from 108.45% in the control treatment (P0) to 40.56% in P3. The best treatment was P1, with a swelling power of 90.34% and the highest overall sensory acceptance among the panelists.

Abstract

The green seaweed from the genus Halimeda is a marine biological resource that holds promise as a source of bioactive compounds, such as antioxidant compounds. The antioxidant activity may differ based on the assay technique since each technique relies on a unique reaction mechanism. This research intended to assess the antioxidant properties of crude Halimeda sp. extract from the Seribu Islands employing three techniques: DPPH, CUPRAC, and FRAP. The sample was obtained through maceration with methanol and subsequently assessed for free radical scavenging activity and metal ion reduction capability. The findings indicated that the CUPRAC method yielded the greatest antioxidant activity, scoring 147.91 µmol Trolox/g extract, while the FRAP method followed with 83.61 µmol Trolox/g extract. The DPPH method generated an IC₅₀ value of 226.09 mg/L. These varying values suggest that the chemical properties of the compounds in the extract react differently to each testing principle. Consequently, employing several antioxidant assays is essential for a more thorough assessment of the antioxidant capacity of Halimeda sp. seaweed extract.
