Abstract

The objective of this study was to determine the capability of coconut water, coconut water-honey, palmyra juice, palmyra juice-honey in maintaining sperm motility and viability of landrace boars during storage at refrigerator (5 °C ) and room temperature (32 °C). This experiment used a completely randomized design (CRD) with 8 treatments and 3 replications. Semen was collected twice a week from landrace boars. Semen Characteristics and their quality were evaluated macroscopically and microscopically. Good quality semen have motility >70%, sperm concentration >200x106 sperm/ml and morphological abnormalities less than 20%, these semen were added with Coconut Water (P1), Coconut Water-Honey (P2), palmyra juice (P3) and palmyra juice-honey (P4). Sperm motility and viability were evaluated every 2 hours until 40% motility were evaluated. The results showed that fresh semen characteristics were good, with the presentage of sperm motility 85%, sperm concentration 266.67 ± 13.33% and morphological abnormalities 12.67±2.90%. The best diluent stogare at 5 °C was palmyra juice-honey better than palmyra juice, coconut water and coconut water-honey with sperm motility 55.70±7.63%during 28 hours observation. The best dilluent storage at 32 °C was palmyra juice with sperm motility 46.67±6.66% during 18 hours observation. In conclusion that palmyra juice-honey and palmyra juice effective to maintain semen quality of landrace boars during preservation

Abstract

The objective of this study was to determine the the effect diluent of commercial is diluent BTS® and MIII® in maintaining sperm motility and viability of landrace boars during storage at different temperatures at refrigerator (5 °C), AC (28 °C) and room temperature (32 °C). This experiment used a completely randomized design (CRD) with 6 treatment and 4 replications. Semen was collected twice a week from landrace boars. Semen Characteristic and their quality were evaluated macroscopically and microscopically. Good quality semen have motility >70%, sperm concentration 200 x 106 sperm/ml and morphological abnormality less than 20%, these semen were added with diluent commercial is diluent BTS® and MIII®. Sperm motility and viability were evaluated every 2 hours until 40% motility were evaluated. The result showed that fresh semen characteristic were good, with the presentage of sperm motility 85% with sperm concentration 242.50±16.5% and morphological abnormality 12.50±1.70%. The results showed that the liquid semen of landrace boar were stored at a temperature of 28 °C is better results temperature with the percentage motility of sperm 54.50% during 38 hours and temperature of 5 °C showed the second best result with the percentage of 46.32% motility during 36 hours of storage in the diluent BTS® . In conclusion : BTS® diluent is an appropriate diluent used to maintaining semen quality of landrace boars quality during preservasion.

Abstract

The study aims to determine the effect of commercial thinning is thinning Beltsville thawing Solution (BTS) and Mulberry III (MIII) with a water jacket method and non-water jacket against landrace pig semen quality after stored at 5° C. This study uses a completely randomized design (CRD) with 4 treatments and 2 control within five repetitions. Fresh semen that fulfill the criteria diluted with Beltsville thawing Solution (BTS) and Mulberry III (MIII), then stored using non-water jacket and water jacket at 5 °C of temperature. Motility and viability observations of spermatozoa performed every 2 hours after storage for up to a decline in the percentage motility of at least 40%. Research result shows the characteristics of fresh semen motility is 84.50 ± 0:50, the concentration is 250 ± 8:36 sperm cells / ml and 13.60 ± 1.72 abnormality. Semen observations which diluted with Beltsville thawing Solution (BTS) at 5 °C temperature shows the water jacket method is better than the non-water jacket method with a percentage of 57.00 ± 1.22 % motility during 32 hours storages. Semen observations which diluted with Mulberry III (MIII) at 5 °C temperature shows the water jacket method is better than the non-water jacket method with a percentage of 51.00 ± 1.00% motility during 32 hours storages. Semen observations which diluted with Beltsville thawing Solution (BTS) and Mulberry III (MIII) shows the BTS water jacket liquid semen is better than BTS non water jacket, miii water jacket and miii non water jacket with motility percentage 57.00 ± 1.22% for 32 hours storages. It was concluded that the water jacket method is better than the non-water jacket method and water jacket BTS is more effective to maintaining the spermatozoa quality during storages at 5 ° C.

Abstract

This research was aimed to defined the morphology of boar’s sperm and using carbofuchsin staining. The semen sample were collected with technique glove hand method from three healty Landrace and Duroc boars belong to UPT pembibitan Ternak Babi Dinas Peternakan Provinsi NTT. Semen were stained using Carbofucsin staining technique, morphology observation was evaluated by microscop on 500 cells for each sample. From macroscopic and microscopic evaluation the result showed that the Landrace boar’s semen volume was308.33±23.63 ml, the pH was 7,4 and the sperm concentration was 230±20x106 cell/mL. Macroscopic and microscopic evaluation the result showed that the Duroc boar’s semen volume was 213.33±41.63 ml, the pH was 7,4 and the sperm concentration was 455±198.30x106 cell/mL. The result of morphology observation in Landrace and Duroc boars showed that the normal sperm was 95.91±1.14% and94.02±2.76%. Abnormal sperm in Landrace and Duroc was4.13±1.11% dan 5.97±2.76%. The abnormality of theLandrace and Duroc sperm consist of narrow (0.07% and0,26%), acrosome defect (0,27% and 0,1%), pear shapered (0,49% and 0.18%), double head (0,07% and 0,04%), variable size (0.11% and 0,12%), lose abnormal head (0.06% and 0,32%) and tail abnormalities (1,72% dan 4,36%). Abstract

This study aims to determine the quality of Landrace porkspermatozoa with the addition of natural thinners of lontar fruit juice and Moringa oleifera Lam. Leaf extract as antioxidants stored at different storage temperatures, namely 5°C (freezer),15°C (refrigerator) and 22°C (air-conditioned room). Semen was collected six times and evaluated both macroscopically and microscopically. The best quality of semen was semen with spermatozoa motility> 70%, concentration> 200x106 cells / ml and abnormal spermatozoa <20%. In this study, there were three treatments, in which the cement with antioxidant diluents was stored at 15°C. (treatment I), 22°C (treatment II) and 5 oC (treatment III). The results showed at the 36th hour of storage the temperature was 22°C with a motility of 55.00 ± 0.20% and a viability of 72.50 ± 0.27%, a temperature of 15°C with a motility of 55.00 ± 0.28% and a viability of 64.50 ± 0.20%, and a temperature of 5°C with a motility of 50.00 ± 0.50 % and viability 57.50 ± 0.33%. It was concluded that the water thinner of lontar fruit and the antioxidant of Moringa oleifera Lam. Leaf extract were able to maintain the quality of the liquid Landrace pig semen at different storage temperatures. A significant difference (P≤0.05) occurred between the 5°C and 22°C treatment, while the 15°C temperature was not significantly different (P≥0.05) with the 5°C and 22°C temperatures.
