Abstract

Moringa leaves (Moringa oleifera) contain several secondary metabolite compounds related to antibacterial, namely flavono-ids, alkaloids, saponins, terpenoids and tannins. These secondary metabolite compounds can be extracted maximally with the selection of extraction methods, solvents used and optimisation of solvent temperature and dissolving time when extracting in order to obtain good extract yields. Laboratory experimental research with maceration extraction method to see the effect of tem-perature and dissolving time on the extraction of moringa leaves. The temperature variations were T1=30±2°C, T2=40±2°C, T3=50±2°C and the time variations were t1=36 hours, t2=48 hours, t3=60 hours consisting of 3 levels with three repetitions so that 27 samples were obtained. Moringa leaf extract yield was calculated based on the weight of the sample after the extraction process against the initial powder weight of the sample. The results showed that the high yield was obtained from the extract-ion treatment at a temperature of T2=40±2°C for t3=60 hours at 4.93%. It is concluded that the solvent temperature and dis-solving time affect the amount of yield produced, the extract yield can increase followed by high temperature and longer ex-traction time until it reaches the optimal limit.

Abstract

Porphyromonas gingivalis is a gram-negative anaerobic bacterium involved in the pathogenesis of periodontal disease that can lead to tooth loss. One way to control P.gingivalis is by using chlorhexidine, but continuous use can eliminate the sensation of saltiness on the tongue. Avocado (Persea americana Mill) is one of the natural ingredients that can be utilised as medicinal and antibacterial plants because it contains several secondary metabolite compounds such as flavonoids, tannins, saponins and al-kaloids. The use of avocado peel as an antibacterial P.gingivalis is not yet known for certain so this research was conducted to determine the antibacterial activity of ethanol extract of avocado peel against P.gingivalis. The extract was obtained by extrac-tion using the maceration method, then tested for antibacterial activity of ethanol extract of avocado fruit peel against P.gingi-valis with concentrations of 20%, 40%, 60%, 80%, 100%, chloramphenicol 30 μg/μL as positive control and dimethyl sulfoxide as negative control. This test was conducted using the disc paper diffusion method. It is concluded that the ethanol extract of avocado fruit peel had antibacterial activity against P.gingivalis. The 20% concentration had an average inhibition diameter of 9.92 mm and 100% concentration inhibition of 16.36 mm.
