Abstract

Abstract. The compounds phalerin, kaempferol, quercetin, myrisetin, and catechol are known to have anti-inflammatory effects in vivo, the mechanism of action of these compounds is thought to be inhibition of cyclooxygenase enzymes, but these activities have not been proven in silico. The purpose of this study was to determine the interaction of phalerin, kaempferol, quercetin, myrisetin, and catechol compounds against COX-1 and COX-2 receptors and determine the toxicity of the test compounds. The research was conducted computationally with molecular tethering methods using MGL Tools software version 1.5.6 which is equipped with AutoDock Tools version 4.2. The results of molecular tethering were visualized using the 2019 version of the BIOVIA Discovery Studio Visualizer software. The toxicity test of the test compound is carried out through the Pro Tox II website. The test results are seen from the value of the bond energy (ΔG) and the inhibition constant (Ki). The test results showed that the five test compounds were able to interact with the test receptor, at the COX-1 receptor the kaempferol compound had an ΔG value of -7.34 kcal / mol and a Ki value of 4.20 μM, and at the COX-2 receptor phalerin had a ΔG value of -9.00 kcal / mol and a Ki value of 0.252 μM. Both compounds are compounds that have bond energy values and inhibition constants that are close to natural ligand values. The results of the toxicity test of the test compounds obtained, the five test compounds have non-toxic results. Keywords: Mahkota dewa, cyclooxygenase enzime, anti-inflammatory, molecular docking. Abstrak. Senyawa falerin, kaempferol, kuersetin, mirisetin, dan katekol diketahui memiliki efek antiinflamasi secara in vivo, mekanisme kerja dari senyawa tersebut diduga dengan adanya penghambatan terhadap enzim siklooksigenase, namun aktivitas tersebut belum dibuktikan secara in silico. Tujuan dari penelitian ini, yaitu untuk mengetahui interaksi senyawa falerin, kaempferol, kuersetin, mirisetin, dan katekol terhadap reseptor COX-1 dan COX-2 serta mengetahui toksisitas dari senyawa uji. Penelitian dilakukan secara komputasi dengan metode penambatan molekul menggunakan software MGL Tools versi 1.5.6 yang dilengkapi AutoDock Tools versi 4.2. Hasil penambatan molekul divisualisasikan menggunakan software BIOVIA Discovery Studio Visualizer versi 2019. Uji toksisitas senyawa uji dilakukan melalui website Pro Tox II. Hasil pengujian dilihat dari nilai energi ikatan (ΔG) dan konstanta inhibisi (Ki). Hasil pengujian menunjukkan bahwa kelima senyawa uji mampu berinteraksi dengan reseptor uji, pada reseptor COX-1 senyawa kaempferol memiliki nilai ΔG sebesar -7,34 kcal/mol dan nilai Ki sebesar 4,20 µM, dan pada reseptor COX-2 senyawa falerin memiliki nilai ΔG sebesar -9,00 kcal/mol dan nilai Ki sebesar 0,252 µM. Kedua senyawa tersebut merupakan senyawa yang memiliki nilai energi ikatan dan konstanta inhibisi yang mendekati nilai ligan alami. Hasil pengujian toksisitas senyawa uji yang didapatkan, kelima senyawa uji memiliki hasil tidak toksik. Kata Kunci: Mahkota dewa, enzim siklooksigenase, antiinflamasi, penambatan molekular.
