Abstract

The objective of this research was to determine the genetic variation of the Maleo (Macrocephalon maleo) based on blood protein polymorphism. Material of the research was blood of Maleo coming from different original habitat, coastal and inland area. PAGE-TYLE (Polyacrilamide Gel Electrophoresis-Thin Layer Electrophoresis) was used in this research. Parameters observed were genotype at four locus Pre Albumin (PAlb), Albumin (Alb), Ceruloplasmin (Cp) and Transferrin (Tf). The obtained various data were used to analyze gene frequency, individual heterozygosity as well as average heterozygosity. The result showed that gene frequencies at allele A and B of four locus Maleo both  in coastal and inland habitat were the same at PAlb locus and Tf locus, that was 0.5; Alb locus was 0.33 vs 0.67 and 0.67 vs 0.33, respectively; Cp locus was 0.5 vs 0.67 and 0.5 vs 0.33, respectively. Individual heterozygosity of four locus Maleo in coastal and inland habitat were the same at PAlb and Tf  that was 0.5; Alb was 0.4555 respectively; Cp was 0.5 vs 0.4555 respectively. Average heterozygosity in coastal and inland Maleo were 0.3911 and 0.3822, respectively. In conclusion, Maleo in coastal and inland habitat showed the same value of diversity relativity due to uncontrolled assortative mating.Key words: Maleo (Macrocephalon maleo), genetic variation, polymorphism blood protein

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh penyuntikan hormon PGF2α terhadap siklus estrus domba palu. Penelitian dilaksanakan pada bulan Januari-Maret 2022 di Laboratorium Pengembangan Agribisnis Peternakan dan Perikanan, Universitas Tadulako. Variabel yang diamati meliputi: lama onset estrus setelah disuntik (jam), lamanya estrus (jam) dan interval siklus estrus (hari). Ternak yang digunakan adalah domba ekor gemuk betina sebanyak 15 ekor berumur 18 bulan. Rancangan yang digunakan adalah Rancangan Acak Lengkap dengan 3 perlakuan dan 5 ulangan. Perlakuan dosis hormon PGF2α yang digunakan secara berturut-turut yaitu 0,5 ml (P1), 1,0 ml (P2) dan 1,5 ml (P3). Apabila perlakuan berpengaruh nyata maka dilakukan uji lanjut menggunakan Uji Beda Nyata Terkecil (BNT). Hasil penelitian menunjukan nilai onset estrus, lama estrus dan interval siklus estrus pada masing-masing perlakuan P1 sebesar 39 jam-30,8 jam-15,5 hari, P2 36 jam-31,6 jam- 15,4 hari dan P3 34,6 jam-34,8 jam-14,8 hari. Hormon PGF2α berpengaruh sangat nyata terhadap nilai onset estrus dan lama estrus (p<0,01) tetapi tidak berpengaruh terhadap interval siklus estrus (p>0,05). Pemberian hormon PGF2α sudah efektif dan lebih ekonomis dalam sinkronisasi estrus untuk menghasilkan gejala-gejala estrus yang berkualitas.
