Abstract

Caulerpa lentilifera merupakan makroalga atau sering disebut dengan anggur laut yang mudah ditemui di Indonesia. Berbagai aktivitas manfaat dari Anggur laut telah ditemukan untuk kegiatan akuakultur salah satunya yakni dalam peningkatan sistem imun ikan. Kajian penggunaan caulerpa sebagai imunostimulan yang efektif perlu dilakukan penelitian untuk menentukan dosis yang tepat untuk meningkatkan imune ikan. Uji toksisitas akut merupakan uji awal yang diperlukan untuk mengetahui dosis lethal ekstrak caulerpa. Hasil uji toksisitas akut dijadikan sebagai dasar penentuan dosis pada uji sub kronis yang selanjutnya akan diketahui dosis efektif dalam peningkatan imunitas ikan. Tujuan penelitian ini adalah untuk menganalisis toksisitas akut ekstrak pelarut yang berbeda yakni pelarut metanol dan pelarut water extract dan menguji pada kadar glukosa darah ikan sebagai indikator tingkat stress ikan. Metode yang digunakan dalam ekstraksi dengan menerapkan metode maserasi dengan pelarut metanol dan pelarut water extract. Uji toksisitas yang dilakukan yaitu metode penentuan LC50-96jamdengan analisis probit.  Metode ini dilakukan diawali dengan uji pendahuluan untuk menentukan ambang lethal atas dan ambang lethal bawah. Selanjutnya dilakukan uji sesungguhnya untuk menentukan nilai LC50-96jam . Pada proses uji toksisitas dilakukan pengamatan kadar glukosa darah ikan komet menggunakan kit blood glucose. Pada penelitian juga dilakukan pengukuran kualitas air seperti suhu, pH dan oksigen terlarut (DO). Hasil penelitian menunjukkan toksisitas akut tertinggi pada ekstrak Caulerpa lentilifera dengan pelarut Metanol sebesar 0,761 ppm. Selanjutnya kadar gula darah tertinggi didapatkan pada ekstrak Caulerpa lentilifera dengan pelarut Metanol sebesar 102 mg/dL pada konsentrasi 0,24 ppm. Parameter kualitas air menunjukkan bahwa pengukuran suhu, pH, oksigen terlarut (DO) didapatkan nilai masih dalam ambang batas untuk mendukung hidup ikan.   Caulerpa lentilifera is a sea grape like-macroalga that grows well in Indonesia. Many beneficial activities of sea grapes have been found for aquaculture, such as to increase fish immune system. However, Caulerpa lentilifera potency as an effective immunostimulant need to evaluate further to determine the proper dose to increase fish immunity. Acute toxicity test is the initial test needed to determine the lethal dose of Caulerpa lentilifera extract. The results then used as the basis to define the dose in the sub-chronic test and further to decide the effective dose in increasing fish immunity. The purpose of this study was to analyze the acute toxicity of different solvent extracts (methanol and water extract) and to evaluate the blood glucose levels of fish studied as an indicator of fish stress levels. The extraction method used was the maceration method . Toxicity test  was carried out to determine LC50-96hours with probit analysis. The toxicity preliminary test was performed to define the upper and the lower lethal threshold. The further toxicity test was carried out to determine the value of LC50-96hours. In the toxicity test process, the blood glucose levels of comet fish were observed using a blood glucose kit. In this study water quality parameters such as temperature, pH, and dissolved oxygen (DO) were also  asesses. The results showed that the highest acute toxicity was found at Caulerpa lentilifera extracted with methanol at 0.761 ppm. Furthermore, the highest blood sugar levels  at 102 mg/dL were found in Caulerpa lentilifera extracted with methanol of at a concentration 0.24 ppm. Water quality parameters indicate that the temperature, pH, and dissolved oxygen (DO) values ​​are still within the threshold to support fish life.

Abstract

Doktor Mengabdi (DM) engages in collaborative efforts to address the issue of escalating feed costs, which result in minimal profits or even financial losses. This is intensified if, during the harvest period, the price of catfish per kilogram decreases. The imperative to address these issues is subsequently addressed through training in the development and production of self-sustaining animal feed utilizing the capabilities of indigenous components. The BUMDES of Labruk Lor Village and the "Mina Agung Mandiri" Fish Farming Group collaborated with the Lumajang District Fisheries Service to carry out the activity. The applied results for this activity encompass: 1) fostering social interaction and promoting comprehension among activity partners; 2) providing guidance and education to enhance understanding of effective and proper fish farming techniques, as well as enabling the production of fish feed using readily available local resources; and 3) implementing the process of fish feed production, which involves grinding the raw materials, blending the various components, and utilizing a fish feed pellet printing machine for shaping. 4). Assessment and surveillance of the execution of tasks 5). Assessing the effects of actions, including their impact on the economy and the growth of fish, This activity empowers farmers to produce feed autonomously and demonstrates a significant upsurge of up to 40% in the profitability of fish farming operations. Moreover, the measurements conducted suggest that the water quality in each partner's pond remains appropriate for the growth of fish and does not exhibit any signs of diseases that could hinder the cultivation and development of the fish.

Abstract

Decreasing water quality and disease outbreaks are negatively affecting catfish farming in Labruk Lor village, Lumajang district, leading to reduced productivity and income for catfish farmers. Service Doctoral Grant programme aims to address issues by offering counselling, training, and optimising yard use through aquaponic activities within the Rumah pangan lestari (RPL) programme. This project is a collaboration between BUMDES Labruk Lor Village, the fish cultivator group "Mina Agung Mandiri" from Labruk Lor village, and the Lumajang Regency Fisheries Service. The methods used in this activity involve socialisation and direct engagement, training and practical application, and monitoring and evaluating. Utilising aquaponics for economic growth can yield the cultivation of 6,984 catfish in 3 m diameter tanks, with an 80% survival rate resulting in a harvest of 6,400 fish weighing 600-700 kg. Vegetables can be harvested 8-10 times per cycle, leading to increased net profit and economic improvement.
