Indonesia is rich in natural resources, so many people use plants as medicinal plants empirically. Traditional medicine must be used properly in order to produce a good therapeutic effect as well. Guava and lemon are thought to have an analgesic effect because they contain steroids, saponins, flavonoids and alkaloids. This study aims to determine the analgesic effect of the combination of lemon extract and guava leaf extract using the hot plate method. In this study, a posttest only design with a control group was used. This study used 25 mice where in each group there were 5 mice. Group I was given CMC 0.5% Na, group II was given diclofenac sodium suspension at a dose of 0.13 mg/20 gBW, group III was given ethanol extract of lemon peel and guava leaves (75%:25%), group IV was given ethanol extract of peel lemon and guava leaves (50%:50%), and group V was given ethanol extract of lemon peel and guava leaves (25%:75%). After 30 minutes of administration of the test solution, the mice were placed on a heated hot plate. Observations were made by looking at the latency time for 2 hours with an interval of 30 minutes. The results showed that group III had a percentage of analgesic activity of 51%, group IV had a percentage of analgesic activity of 52%, while group V had a percentage of analgesic activity of 55%. It was concluded that group V had a higher percentage than the other combined extracts.Keywords: analgesic; hot plates; extractABSTRAK Indonesia kaya akan sumber daya alam, sehingga masyarakat banyak menggunakan tumbuhan sebagai tanaman obat secara empiris. Obat tradisional harus digunakan dengan baik agar manghasilkan efek terapi yang baik juga. Jambu biji dan lemon diduga memiliki efek analgesik karena di dalamnya terkandung steroid, saponin, flavonoid dan alkaloid. Penelitian ini bertujuan untuk mengetahui efek analgesik pada kombinasi ekstrak lemon dan ekstrak daun jambu biji dengan metode hot plate. Dalam penelitian ini digunakan desain posttest only with control group. Penelitian ini menggunakan 25 ekor mencit dimana dalam setiap kelompok terdapat 5 ekor mencit. Kelompok I diberi Na CMC 0,5%, kelompok II diberi suspensi natrium diklofenak dosis 0,13 mg/20 gBB, kelompok III diberi ekstrak etanol kulit lemon dan daun jambu biji (75%:25%), kelompok IV diberi ekstrak etanol kulit lemon dan daun jambu biji (50%:50%), dan kelompok V diberi ekstrak etanol kulit lemon dan daun jambu biji (25%:75%). Setelah 30 menit pemberian larutan uji, mencit ditempatkan di atas hot plate yang sudah dipanaskan. Pengamatan dilakukan dengan melihat waktu latensi selama 2 jam dengan interval waktu 30 menit. Hasil penelitian menunjukkan bahwa kelompok III memiliki persentase aktivitas analgesik sebesar 51%, kelompok IV memiliki persentase aktivitas analgesik sebesar 52%, sedangkan kelompok V memiliki persentase aktivitas analgesik sebesar 55%. Disimpulkan bahwa kelompok V memiliki persentase yang lebih besar dibandingkan dengan ekstrak kombinasi lainnya.Kata kunci: analgesik; hot plate; ekstrak
