Abstract

Abstract

Background: Endometriosis is a chronic estrogen-dependent inflammatory disorder associated with pelvic pain and infertility. Despite its high prevalence, the molecular regulatory mechanisms underlying disease progression remain incompletely understood. Objective: This study aimed to identify differentially expressed genes (DEGs) and prioritize key regulatory candidates involved in endometriosis pathogenesis using integrative bioinformatics analysis. Methods: Publicly available human transcriptomic datasets comparing endometriotic lesions and normal endometrial tissues were analyzed. Data were normalized and subjected to differential expression analysis using adjusted p-value < 0.05 and |log2 fold change| ≥ 1 as thresholds. Functional enrichment analysis and protein–protein interaction (PPI) network construction were performed to identify enriched pathways and hub genes. Results: A total of 326 significant DEGs were identified, including 189 upregulated and 137 downregulated genes. Enrichment analysis revealed predominant involvement of inflammatory signaling, angiogenesis, extracellular matrix remodeling, and steroid hormone response pathways. Network analysis identified IL6, STAT3, VEGFA, and MMP9 as central upregulated hub genes, while PGR and HOXA10 were among the key downregulated regulators associated with progesterone resistance. Conclusion: Endometriosis exhibits coordinated activation of inflammatory–angiogenic networks and suppression of steroid signaling pathways. The identified hub genes represent potential biomarkers and therapeutic targets, warranting further experimental validation.

Abstract

Background: Heart failure (HF) remains a major global health burden associated with high morbidity and mortality. Conventional biomarkers, such as B-type natriuretic peptide (BNP), have limited diagnostic performance because they do not fully capture the molecular complexity of HF. Integrating multi-omics data with artificial intelligence (AI) may improve the identification of novel diagnostic biomarkers. Objective: This study aimed to identify novel diagnostic biomarkers for HF by integrating transcriptomic and proteomic datasets using AI-assisted machine learning. Methods: Public transcriptomic and proteomic datasets from patients with HF and healthy controls were integrated in an in silico bioinformatics study. Differential expression analysis was performed to identify dysregulated genes and proteins, followed by Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment, and protein–protein interaction (PPI) network analyses. Candidate biomarkers were prioritized using Least Absolute Shrinkage and Selection Operator (LASSO), Random Forest, and Extreme Gradient Boosting (XGBoost). Diagnostic performance was evaluated using receiver operating characteristic (ROC) curve analysis. Results: Multi-omics integration identified 1,247 differentially expressed genes, 186 differentially expressed proteins, and 42 overlapping molecules. Enrichment analyses indicated significant involvement in cardiac remodeling, inflammation, oxidative stress, energy metabolism, and calcium signaling. Five hub biomarkers (NPPB, ATP2A2, IL6, PPARGC1A, and TGFB1) were consistently selected by all machine learning models. Individual biomarkers achieved area under the ROC curve (AUC) values ranging from 0.88 to 0.94, whereas the combined five-biomarker panel achieved an AUC of 0.97, with 92.4% sensitivity and 90.1% specificity. Conclusion: AI-assisted multi-omics integration identified five biologically relevant biomarkers with excellent diagnostic performance, providing a promising foundation for future HF diagnostics and precision cardiology.

Abstract

Clean and Healthy Living Behavior (CHLB) is an essential indicator for creating a healthy school environment and preventing environment-related diseases among elementary school children. Preliminary observations at SDIT Masyarul Huda revealed that students' implementation of CHLB indicators was not yet optimal, particularly regarding proper handwashing with soap, personal hygiene practices, and awareness of the importance of maintaining healthy behaviors within the school environment. These findings served as the basis for implementing an interactive educational program aimed at improving students' knowledge of CHLB indicators. This community service program aimed to improve elementary school students' knowledge of Clean and Healthy Living Behavior (CHLB) indicators in the school setting through an interactive educational approach. The program involved 87 students and employed a one-group pretest–posttest design. Students' knowledge was assessed using a pretest–posttest questionnaire administered before and after the intervention. The educational activities consisted of interactive lectures, group discussions, and handwashing demonstrations. Data were analyzed using the Wilcoxon signed-rank test. The results demonstrated that the proportion of students with good knowledge increased from 33.33% during the pretest to 73.56% during the posttest. The normality test indicated that the data were not normally distributed (p < 0.005); therefore, the Wilcoxon signed-rank test was applied, revealing a statistically significant difference between pretest and posttest knowledge scores (p < 0.005). These findings indicate that interactive education effectively improved students' knowledge of CHLB indicators in the school environment. Furthermore, this approach has the potential to serve as a sustainable school-based health promotion model for fostering clean and healthy living behaviors among elementary school students
