Aktivitas Antihiperurisemia Ekstrak Etanol Herba Suruhan (Peperomia pellucida (L.) Kunth) Pada Mencit Jantan

Background: Unhealthy eating patterns in a society that often consume animal protein which contains high purine, cause hyperuricemia is increasing. When disease strikes, the patient may experience pain torture, swollen or deformed joints of the hands and feet. Suruhan or Peperomia pellucida (L.) Kunth has been used empirically in the treatment of uric acid by the community. Objective: To find out the effect of ethanol extract of suruhan herbs to decrease levels of uric acid in male mice induced by potassium oxonate intraperitoneally. Results: Analysis of the test results indicate that the effect of decreasing the levels of uric acid ethanol extract of suruhan herb dose of 50 mg / kg BW (24.35%) did not give a significant differences  with allopurinol dose of 10 mg / kg BW (33.61%) with a significance value is 0.450 (p> 0.05).Methods: The decreased of  blood uric acid levels effect test is done by inducing mice with potassium oxonate then given the test preparation and measuring the levels with measure blood uric acid levels tool. Three dossages which tested were 50 mg/kg BW, 100 mg/kg BW and 200 mg/kg BW. All drugs and extracts were administered orally and observation with duration time in 1 hour during 5 hours. Allopurinol at dossage 10 mg/kg BW as positive control and carboxy methyl cellulose 1% as negative control. Each treatment group consisted of 6 male mice. Test result were analyzed by the method of Analysis of Variance (ANAVA) followed by  Post Hoc Duncan method. Conclusion: It is concluded that ethanol extract of suruhan herb has antihyperuricemic effects to the mice. Keywords: ethanol extract of suruhan herb; hiperuricemic; uric acid ABSTRAK Latar belakang: Pola makan yang tidak sehat dalam masyarakat yang sering mengkonsumsi protein hewani yang banyak mengandung purin tinggi, menyebabkan penyakit hiperurisemia semakin meningkat. Bila penyakit ini menyerang, penderita dapat mengalami siksaan nyeri, pembengkakan atau cacat persendian tangan dan kaki. Tanaman suruhan atau Peperomia pellucida (L.) Kunth, secara empiris telah digunakan dalam pengobatan asam urat oleh masyarakat. Tujuan: Penelitian ini bertujuan untuk mengetahui efek ekstrak etanol herba suruhan terhadap penurunan kadar asam urat pada mencit jantan yang diinduksi dengan kalium oksonat secara intraperitoneal. Metode penelitian: Uji efek penurunan kadar asam urat darah mencit dilakukan dengan cara menginduksi mecit dengan kalium oksonat, kemudian diberikan sediaan uji lalu di ukur kadarnya dengan alat pengukur kadar asam urat darah. Dosis ekstrak etanol herba suruhan yang diujikan yaitu 50 mg/kg BB, 100 mg/kg BB, dan 200 mg/kg BB yang diberikan secara oral dengan pengamatan selang waktu 1 jam selama 5 jam. Sebagai kontrol positif digunakan allopurinol dosis 10 mg/kg BB dan CMC 1% sebagai kontrol negatif. Masing-masing kelompok perlakuan terdiri dari 6 ekor mencit jantan. Data hasil pengujian dianalisis dengan metode analisis variansi (ANAVA),  kemudian dilanjutkan dengan Post Hoc Duncan. Hasil: Hasil uji analisis menunjukkan bahwa efek penurunan kadar asam urat ekstrak etanol herba suruhan dosis 50 mg/kg BB (24,35%)  tidak berbeda signifikan dengan allopurinol dosis 10 mg/kg BB (33,61%) dengan nilai signifikansi 0,450 (p>0,05). Kesimpulan: Disimpulkan bahwa ekstrak etanol herba suruhan mempunyai efek sebagai antihiperurisemia terhadap mencit.
