Abstract

ABSTRACTSponges are one of the biota components that make up coral reefs that have bioactive potential, which has not been widely utilized. The secondary metabolite content of sponges is known to be able to ward off and inhibit the pathogenic bacteria that interfere with it. The purpose of this study was to determine whether the extracts and fractions of the Liosina paradoxa sponge collected from Mantehage Island waters able to inhibit the growth of Escherichia coli and Staphylococcus aureus bacteria. The sample was extracted by maceration method with ethanol solvent and the fractionation method used was liquid - liquid fractionation. Antibacterial activity was tested using the agar diffusion method (Kirby and Bauer disc diffusion). The results obtained from the antibacterial activity test against the Escherichia coli bacteria, the methanol fraction got an average result of 9.94 mm and the average result of the chloroform fraction was 8.06 mm. In the Staphylococcus aureus bacteria, the methanol fraction got an average yield of 11.04 mm and the chloroform fraction an average yield of 7.19 mm, while the ethanol extract and n-hexane fraction did not have antibacterial activity on the two tested bacteria. Methanol fraction has moderate inhibitory power against Escherichia coli bacteria and strong inhibition against Staphylococcus aureus bacteria, while chloroform fraction has moderate inhibitory against both tested bacteria. Keywords: Liosina paradoxa, Antibacterial, Escherichia coli, Staphylococcus aureus ABSTRAKSpons merupakan salah satu komponen biota penyusun terumbu karang yang mempunyai potensi bioaktif yang belum banyak dimanfaatkan. Kandungan metabolit sekunder dari spons diketahui mampu menangkal dan menghambat bakteri patogen pengganggunya. Tujuan penelitian ini adalah untuk mengetahui apakah ekstrak dan fraksi dari spons Liosina paradoxa yang diperoleh dari perairan Pulau Mantehage memiliki aktivitas antibakteri terhadap bakteri Escherichia coli dan Staphylococcus aureus. Sampel diekstraksi dengan metode maserasi dengan pelarut etanol dan metode fraksinasi yang digunakan yaitu fraksinasi cair – cair. Aktivitas antibakteri diuji menggunakan metode difusi agar (disc diffusion Kirby and Bauer). Hasil yang didapat dari uji aktivitas antibakteri pada bakteri Escherichia coli, fraksi metanol mendapatkan hasil rata–rata 9,94 mm dan pada fraksi kloroform hasil rata – rata 8,06 mm. Pada bakteri Staphylococcus aureus, fraksi metanol mendapatkan hasil rata - rata 11,04 mm dan pada fraksi kloroform hasil rata – rata 7,19 mm, sedangkan ekstrak etanol dan fraksi n – heksan tidak memiliki aktivitas antibakteri pada kedua bakteri uji. Fraksi metanol memiliki daya hambat sedang terhadap bakteri Escherichia coli dan daya hambat kuat terhadap bakteri Staphylococcus aureus, sementara fraksi kloroform memiliki daya hambat sedang terhadap kedua bakteri uji.Kata kunci : Liosina paradoxa, Antibakteri, Escherichia coli, Staphylococcus aureus
