ISOLASI DAN IDENTIFIKASI KITIN DAN KITOSAN DARI KULIT UDANG WINDU (Penaeus monodon) DENGAN SPEKTROSKOPI INFRAMERAH

Abstract

ABSTRAK Kitin dan kitosan merupakan biopolimer alami pada Krustasea dan Jamur. Kitosan digunakan secara ekstensif sebagai bahan baku di berbagai industri. Penelitian bertujuan untuk mengekstraksi kitin dan kitosan dari kulit udang melalui reaksi deproteinisasi, demineralisasi, dekolorisasi dan deastilasi. Rendemen kitin dan kitosan  yang dihasilkan secara berturut turut adalah 60,5% dan 63,0%. Hasil analisis terhadap spektrum FTIR kitin memperlihatkan beberapa puncak utama pada bilangan gelombang 3554,45 cm-1 yang menunjukkan vibrasi pembengkokan amida sekunder dan amina (NH) sekunder pada bilangan gelombang 1670.35 cm-1 serta 1427,32cm-1 menunjukkan adanya vibrasi peregangan CH. Hasil analisis spektrum FTIR kitosan menunjukkan adanya vibrasi peregangan simetris pada 3302,20 cm-1 akibat adanya tumpang tindih OH dan amina (NH), vibrasi peregangan 1577.71 cm-1 disebabkan oleh perambatan C = O peregangan (amida I) dan vibrasi peregangan 1666,30 cm-1 yang menunjukkan amida sekunder. Hasil karakterisasi dengan spektroskopi inframerah menunjukkan bahwa senyawa hasil ekstraksi adalah kitin dan kitosan. Kata kunci : Ekstraksi,kulit udang, kitin, kitosan, spektroskopi inframerah. ABSTRACT Chitin and chitosan are natural biopolymers in crustaceans and fungi. Chitosan is used extensively as raw materials in various industries. The study aimed to extract chitin and chitosan from shrimp shells through deproteinization, demineralization, decolorization and deastilation reactions. The yields of chitin and chitosan which were produced successively were 60.5% and 63.0%. The analysis of FTIR chitin spectrum showed some major peaks at wavelength 3554,45 cm-1 indicating secondary bending amide and secondary amine (NH) vibration at wave number 1670.35 cm-1 and 1427,32cm-1 indicating stretch vibration of CH. The results of FTIR spectroscopy analysis showed a symmetrical stretch vibration at 3302.20 cm-1 due to overlapping of OH and amine (NH), stretching vibration 1577.71 cm-1 caused by propagation C = O stretching (amide I) and stretch vibration 1666, 30 cm-1 indicating secondary amide. The results of characterization by infrared spectroscopy showed that  the extraction compound was chitin and chitosan. Key words : Extraction, shrimp skin, chitin, chitosan, infrared spectroscopy.
