Measuring scientific literacy and cultural awareness is important in science education because both are fundamental competencies for the 21st century. The objective of this research was to create a multiple-choice test instrument to measure scientific literacy and a questionnaire to assess cultural awareness. In addition, this study was oriented towards evaluating the instrument through reliability and validity tests based on expert assessments and empirical trials. The research method used the stages of instrument development according to Oriondo Dallo-Antonio, which included: (1) planning the test, (2) trialing the test, (3) determining validity, and (4) determining reliability. A random sampling technique was used involving 142 students. Data were analyzed using Aiken V and Item Response Theory (IRT) in the QUEST program. Validity results showed a high Aiken V index of 0.99 on the scientific literacy test and 1 on the cultural awareness questionnaire. Reliability results showed high levels of 0.86 and 0.81 for the scientific literacy and cultural awareness instruments, respectively. Most items from both instruments were moderately difficult. These findings demonstrate that the proposed instrument is valid and reliable, making it suitable for measuring scientific literacy and cultural awareness. These findings have an impact by providing an instrument that can serve as a basis for teachers and policymakers to measure students’ scientific literacy and cultural awareness. Â
