Antibiotic resistance represents a global health crisis that has driven the exploration of natural products as alternative antibacterial agents. This research aimed to evaluate the efficacy of combining white pomegranate peel (Punica granatum) and red betel leaf (Piper crocatum) extracts in inhibiting the growth of Staphylococcus aureus in vitro. A completely randomized design (CRD) was employed using the disc diffusion method. The treatments consisted of combination ratios of 1:1, 1:2, and 2:1 at concentrations of 25, 50, 75, and 100%. Chloramphenicol served as the positive control, while distilled water was used as the negative control. The findings revealed that the 1:2 ratio combination produced the highest inhibition zone (25.50 mm), which was significantly different (p < 0.05) from the positive control. This combination demonstrated a mutually reinforcing effect. It was concluded that the combination of white pomegranate peel extract and red betel leaf extract holds promise as a natural antibacterial agent.
