Raru (Cotylelobium melanoxylon) is a plant that has been traditionally used in North Sumatra for its potential therapeutic benefits, particularly in the treatment of diabetes. The purpose of this study was to determine the potential of the ethyl acetate fraction of Raru leaves as an antidiabetic through in vitro testing using the α-glucosidase enzyme. The research began with the method of separating compounds from methanol extract and fractionation, followed by looking at the separation of each fraction using the TLC method, glucosidase inhibition test. The results of this study indicate that the ethyl acetate fraction of Raru leaves has α-glucosidase enzyme inhibitory activity (IC50 = 7.19 ppm) which is categorized as a strong inhibitor.
