Isolation of Escherichia coli from Faunus ater Snails in the Lampanah River Estuary of Aceh Besar

Abstract

Escherichia coli is a normal bacterium in the digestive tract of humans and animals, but it can be pathogenic when it exceeds normal limits. This study aims to isolate Escherichia coli from estuary water and Faunus ater snails in the Lampanah river estuary area, Aceh Besar. The research method was conducted through field observations and laboratory examinations using a modified Carter method. A total of 3 samples of Faunus ater snails and 3 samples of estuary water were inoculated into nutrient broth (NB) and incubated for 24 hours at 37°C. Positive results in the NB medium were transferred to EMBA selective medium, and bacterial growth was observed through changes in the color of the EMBA medium as well as macroscopic and microscopic morphology. The macroscopic results showed that the bacteria on the EMBA medium were metallic green with a colony morphology of 2 mm in size, round shape, serrated margins, convex elevation, smooth surface, and shiny appearance. The microscopic morphology showed short rod-shaped, red bacteria. The conclusion of this study is that the Faunus ater snails and estuary water in the Lampanah river estuary area, Aceh Besar, indicate the presence of Escherichia coli contamination.
