ABSTRAKPendahuluan: Periodontitis kronis merupakan penyakit peradangan destruktif jaringan periodontal yang disebabkan oleh bakteri patogen jenis anaerob yang dapat ditemukan di plak subgingiva dan cairan sulkus gingiva. Terapi periodontal perlu dilakukan untuk mengeliminasi bakteri periodontopatogen dengan perawatan non-bedah scaling & root planing didukung dengan mengkonsumsi obat yang bersifat antimikroba sebagai terapi adjuvan. Probiotik seperti Lactobacillus reuteri berpotensi sebagai terapi adjuvan karena mampu memproduksi zat antibakteri berupa bakteriosin. Penelitian ini bertujuan untuk menganalisis efektivitas tablet hisap probiotik Lactobacillus reuteri terhadap penurunan populasi bakteri anaerob dalam cairan sulkus gingiva pasien periodontitis kronis pasca scaling & root planing. Metode: Studi eksperimental klinis uji coba terkontrol acak pendahuluan (preliminary RCT) dengan rancangan pre-test post-test with control group design melibatkan 8 subjek yang terbagi menjadi kelompok kontrol (hanya SRP, n=4) dan kelompok perlakuan (SRP + konsumsi tablet hisap L. reuteri 2x/hari selama 7 hari, n=4). Sampel CSG diambil pada hari ke-0 (pasca SRP) dan hari ke-8 (setelah 7 hari masa perlakuan). Perhitungan koloni bakteri dilakukan menggunakan colony counter dengan metode total plate count (TPC) (CFU/mL), lalu dianalisis statistik menggunakan Uji Paired t-test dan Unpaired t-test. Hasil: Hasil rerata penurunan jumlah koloni bakteri anaerob (105 CFU/mL) pada kelompok perlakuan dan kontrol secara berturut-turut sebesar 242,25 ± 10,1 dan 165,75 ± 52,6 (p≤0,05). Hasil penelitian menunjukkan terdapat perbedaan bermakna rerata penurunan jumlah koloni bakteri anaerob antara kelompok perlakuan dengan kelompok kontrol (p≤0,05). Simpulan: Scaling & root planing dikombinasikan dengan mengkonsumsi tablet hisap probiotik Lactobacillus reuteri efektif dalam menurunkan populasi bakteri anaerob penyebab periodontitis kronis.KATA KUNCI: probiotik lactobacillus reuteri, bakteri anaerob, periodontitis kronis, scaling and root planing, cairan sulkus gingiva.Effectiveness of Lactobacillus reuteri probiotic lozenges in reducing anaerobic bacterial population in gingival crevicular fluid after scaling and root planning: Study experimentalABSTRACTIntroduction: Chronic periodontitis is a destructive inflammatory periodontal disease caused by anaerobic pathogenic bacteria found in subgingival plaque and gingival crevicular fluid. Periodontal treatment is needed to eliminate periodontopathogenic bacteria through nonsurgical treatment with scaling and root planing, supported by the use of antimicrobial medicine as adjuvant therapy. Probiotics such as Lactobacillus reuteri have potential as adjuvant therapy because they can produce bacteriocins, which are antibacterial substances. This study aimed to analyze the effectiveness of L. reuteri probiotic lozenges in reducing anaerobic bacterial populations in the GCF of patients with chronic periodontitis after scaling and root planing. Methods: A preliminary randomized controlled trial (RCT) with a pre-test post-test with control group design was conducted involving 8 subjects, divided into a control group (SRP only, n=4) and a treatment group (SRP + use of L. reuteri lozenges twice daily for 7 days, n=4). GCF samples were collected on day 0 (post-SRP) and day 8 (after the 7-day treatment period). Bacterial colony counting was performed using a colony counter with the total plate count (TPC) method (CFUs/mL), and the data were statistically analyzed using paired and in paired t-tests. Results: Anaerobic bacterial colonies (105 CFU/mL) decreased at 242.25 ± 10.1 in the treatment group and 165.75 ± 52.6 in the control group. The results showed a significant difference in the mean reduction in the number of anaerobic bacterial colonies between the treatment group and the control group (p≤0.05). Conclusion: Scaling & root planing combined with the use of probiotic Lactobacillus reuteri lozenges is effective in reducing the population of anaerobic bacteria associated with chronic periodontitis.KEY WORDS: Lactobacillus reuteri probiotic, anaerobic bacteria, chronic periodontitis, scaling and root planing, gingival crevicular fluid.
