Abstract

Biosurfactants produced by bacteria represent an environmentally friendly alternative for the remediation of petroleum-contaminated soils. This study aimed to isolate, evaluate, and characterize biosurfactant-producing bacteria from petroleum-contaminated soil. The research employed a descriptive survey combined with laboratory experiments, and samples were collected using purposive sampling combined with random sampling. Three bacterial isolates were successfully obtained from a soil sample contaminated with petroleum, collected from a mining site in the city of Pekanbaru, Riau and cultured on Bushnell–Haas Agar supplemented with 1% petroleum oil. Among them, isolate MIB-1 demonstrated the highest potential, showing positive results in hemolysis, oil spreading, and drop collapse assays, with an emulsification index (EI24) of 12.5%. Morphological characterization of the best MIB-1 isolate revealed that it is rod-shaped, Gram-positive, and catalase-positive. The findings highlight the potential of the bacterial isolate as a biosurfactant producer for sustainable bioremediation of petroleum-contaminated soil and contribute to the exploration of indigenous Indonesian microbial resources for environmental applications. However, further optimization at a larger scale is required.
