Abstract

Central obesity was accumulation of fat in the abdominal region. Many studies showed correlations between central obesity and cardiovascular diseases, e.g. diabetes and dyslipidemia. This study was conducted to know the difference between HbA1c and lipid profil between the women with and without central obesity. This study was an analytic observational study with cross-sectional design. Subjects of 52 respondents were healthy adult women staff in campus I, II, III Sanata Dharma University in Yogyakarta and selected using purposive sampling technique. The data of waist circumference, pelvic/hip circumference, HbA1c and lipid profile were collected among the subjects and analyzed with computer with 95% confidence interval. The results of this study showed HbA1c levels and lipid profile were different between the women with and without central obesity, although the difference was not statistically significant.

Abstract

Breast cancer is a cancer caused by uncontrolled cell growth at breast tissue. One of the most common triggers of breast cancer is overexpression of estrogen receptor alpha (ER?). This researchs goal is to test the ability of coumestrol as the ligand of ER? with in silico method and to discover coumestrols binding pose inside the ER?s binding pocket. Coumestrols ability as ER?s ligand was tested using structure-based virtual screening (SVBS) method by Setiawati et al. (2014) that had been modified by Istyastono (2015). Results analysis was done using decision tree generated from recursive partition and regression tree method (RPART). If coumestrol is a ligand based on decision tree, it is concluded that coumestrol is active as ligand of ER?. At the end of analysis, coumestrols pose inside ER?s binding pocket was visualized using MacPyMol. From the test acknowledged that the smallest ChemPLP value of coumestrols pose was -83.1487. Coumestrol interacts with GLY420, ARG394, and GLU353 using hydrogen bonds. However, coumestrol were perceived as decoy according to decision tree. Hence, coumestrol could not be recognized as ER?s ligand by the protocol. Therefore, development of proper protocol to indentify ligand for ER? is required.

Abstract

Wounds often leave scars that interfere with skins aesthetic, even lead to health problems, in some of people, scars tend to grow excessively cause of keloid scarring or cuts. One of the causes of scarring wounds is the process of long-term inflammation. Diclofenac sodium is a bitter taste substances. Diclofenac sodium is a selective inhibitor, which inhibit COX-2 in inflammatory processes. Binahong (Anredera cordifolia) is one of the plants in Indonesia, which came from China. Binahong leaves contain saponins, flavonoids, namely quinon, steroids, monoterpenoid, and sesquiterpenoid which has been known as a wound healer. The making of ointment scarless wound healing dosage form is by combining binahong leaves extract and the addition of anti-inflammatory agent diclofenac sodium (F4), which expected to reduce the wound scar on the Switzerland Webster strain mices skin that has bleed with incision method. The test that has been used is histopathology test followed by an extensive calculation of collagen. The calculation data were analyzed using T-test with 95% confidence level. The results from this research indicate that ointment of binahong extract and diclofenac sodium (F4) produce less scar.

Abstract

Wound is a condition where the tissue integrity is damaged so that body will attempt to repair the damaged tissue by wound healing mechanism. This mechanism usually results in the scar formed by its inflammatory phase. Binahong (Anredera cordifolia (Ten.) Steenis) contains ascorbic acid and flavonoids which are important for collagen formation, to improve the rate of the wound healing process. Piroxicam can shorten or detain the inflammatory phase by inhibiting the cyclooxygenase (COX) enzymes in the prostaglandine synthesis process, which play an important role in scar formation. The aim of this research is to discover if the combination of piroxicam and binahong extract in the scarless wound gel could offer scar reduction effect. In this research, a gel preparation with binahong extract was combined with piroxicam to develop the scarless wound gel (BinPirox). The research was purely experimental. It was done by conducting a histopathological test followed by collagen area calculation. The data were analyzed by independent sample t-test with 95% significancy level. In this research, the addition of piroxicam was expected to reduce the scar formation on incisional wound of white Swiss Webster mice (Mus musculus). The result showed that BinPirox formed statistically less scar when compared to Bin (a gel preparation with binahong extract).

Abstract

Previous studies has shown that estrogen receptor alpha that binds to estrogen may increase cancer cell proliferation, thus estrogen receptor alpha can be targeted to cure breast cancer. The aim of this in silico study is to test whether phytoestrogen 2,6-dihydroxyanthraquinone is a ligand of estrogen receptor alpha. Setiawati et al. (2014) has established a valid protocol for Structure Based Virtual Screening using molecular docking software PLANTS 1.2. This protocol is used to test 2,6-dihydroxyanthraquinone as ligand for estrogen receptor alpha. Output from the protocol is analyzed using a post-docking analysis method developed by Istyastono (2015) in R 3.0.2. Visualization of binding pose is generated with PyMOL 1.2. Results show that 2,6-dihydroxyanthraquinone cannot be identified as a ligand of estrogen receptor alpha. The method used in this study is not suitable to identify 2,6-dihydroxyanthraquinone, which is a marginal active substance, as active ligand. Further study to develop a suitable method to identify marginal active substances as ligands in estrogen receptor alpha is needed.

Abstract

Breast cancer is a disease with abnormal cell proliferation at breast tissue that can invade the surrounding tissue and spread to another organ. Based on WHO (2014) at 2012, there are 48,998 breast cancer cases on women in Indonesia with 19,730 death cases. In breast cancer, overexpression estrogen receptor alpha (ER-?) usually observed. Hence ER-? became the focus of prevention and therapy for breast cancer. In vitro study of emodin shows IC50 2.7 M and Ki 0.77 M on ER-?. In silico research using docking protocol and post docking analysis protocol shown that emodin was not an active ligand on ER-?. The outcome shown as ChemPLP score with average -75.292 and PLIF bitstring. Emodin binds with LEU346, LEU387, and ARG394 residue. Currently, the protocol that used in this research has yet identified marginal compound like emodin as an active ligand on ER-?.

Abstract

Wound is a defect of skin caused by physical or thermal damage. The inflammatory phase in the wound healing usually causes scars. Aspirin is a nonsteroidal anti-inflammatory drug (NSAID) that has the ability to inhibit the activity of cyclooxygenase (COX) leading to reduced prostaglandin amount. Binahong (Anredera cordifolia) is one of the plants that is used as a wound healer. Binahong contains ascorbic acid which has an important role in collagen formation phase. In this study, binahong leaf extract ointment will be combined with aspirin to produce scarless wound ointment. The method used is a purely experimental method. The test method used is histopathology tests then processed by the method of calculating the area of collagen. The data are analyzed using T-test. The addition of aspirin in the preparation of wound healing ointment cant reduce scar formation allegedly with an incision method of white mice (Mus musculus) Swiss Webster. Statistically, the results showed that binahong ointment (UB) produces the least scar than ointment base (B), followed binahong-aspirin ointment (UBA), and aspirin ointment (UA).

Abstract

Wound is a condition where the tissue was damaged. The body will repair this damage by wound healing mechanism which often leads to the formation of scar. The scar is caused by the inflammation phase of wound healing mechanism. Ibuprofen is one of the antiinflammatory agent that can inhibit or shorten the inflammatory phase by inhibit cyclooxygenase enzyme in prostaglandin synthesize. Prostaglandin has an important role on the formation of inflammatory phase. Binahong (Anredera cordifolia (Ten.) Steenis.) has a potential activity as a wound healing agent. Binahong contains ascorbic acid which is impotant for the activation of prolyl-hydroxygenase enzyme that support hydroxylation phase in the process of collagen formation, so that the wound healing process can be acceletared. In this research, a gel preparation with binahong extract was combined with ibuprofen to form a scarless wound gel. This research was purely experimental. The test method used is a histopahological test which continued with a calculation of collagen area and a physical properties test. The collagen area calculation data were analyzed by independent sample t-test with 95% confidence interval. In this research, the addition of ibuprofen was expected to reduce the scar formation on incisional wound of white Swiss Webster mice. The result showed that the gel preparation with binahong extract and ibuprofen formed statistically less scar when compared to the gel preparation with binahong extract only.
