Abstract

Caffeine is one of alkaloids which contains in coffee bean, tea leaves and cacao bean. Theaims of this research were to determine caffeine level in coffee powder and the maximum ofcoffee powder level which consumed per day based on SNI. Samples were 6 coffee powdersin Manado city. Identification of caffeine was done using parry reagent, although caffeinelevel was determine using spectrophotometer UV-Vis. The result shows that 6 coffeepowders from Manado contained caffeine. Caffeine level in Manado coffee powder were13,81 mg (sample A), 13,63 mg (sample B), 12,33 mg (sample C), 10,10 mg (sample D),10,13 mg (sample E) and 9,53 mg (sample F). Maximum coffee powder level whichconsumed per day based on SNI were 15,73 g-10,86 g per day.Key words : Caffeine, coffee powder, spectrophotometry UV-Vis, Manado

Abstract

The objective of this research was to determine the species or genus of mercury resistant bacteria culture from mercury polluted waters. Microbiology identification of culture S3.2.2 was done with morphology, physiology and biochemistry test. Molecular analysis was done by amplifying 16S rRNA gene using PCR method. Universal Bact- F1 forward/Uni-B1 Reverse was using as primer. Amplification result was electrophorize with agarose 1,5%, furthermore sequencing was done against 16SrRNA gene. The result shows that culture S3.2.2 were negative grams bacteria, rod shaped, nonmotil, catalase (+), citrat (+), H2S (-), fermentase (+), indol (+) and lisin (+). Blast analysis against nucleotide sequence of 16S rRNA gene possesses 99% similarities with Klebsiella pneumoniae.

Abstract

The dose of dextrometorphan often misused by some people. Excessive doses ofdextrometorphan gives euphoria feeling, sense of calm, sight and hearing hallucination. Theaim of this research was to find out the profile of dextrometorphan abused on sub district EastTombariri Society, Minahasa Regency. This research is a descriptive research. The researchusing interview method to 50 respondents which given 7 same and structured question oneach respondents. The result shows that dextrometorphan abuse was occurred among youth insub district East Tombariri Society, Minahasa Regency. Information about dextromethorphanwere the knowledge to obtained drugs which did not fit distribution procedure and thepurpose of drug uses to relieve stress. Dextrometorphana can be consumed a few youth grainsper day and usually combined with alcohol drinks to accelerate the desired effect such asincrease confidence, happiness, reduce burden, and free feeling.Key words : Drug abuse, Dextrometorphan, East Tombariri, Interview

Abstract

This research to determine the effect of extraction method on free-radical scavengers on shellof yaki pinang fruit. Yaki betel nut shell extraction is done by the method of maceration,percolation and soxhletasi. Activity test done with free-radical scavengers DPPH. Resultmethod were analyzed by using UV-Vis spectrophotometer. Results of research on methodsoxhletasi have free-radical scavengers that activity is quite high in concentrations of 50mg/L (85,16%) and 100 mg/L (92,31%) compared to the maceration extraction method thathas a value of percent inhibition of 74,61% at a concentration 50 mg/L and 81,32% at aconcentration of 100 mg/L and percolation to the value of 14,28% at a concertration of 50mg/L, and 17,17% at a concentration of 100 mg/L using the DPPH test.Key words : pinang yaki (Areca vestiaria Giseke, antioxidant, DPPH

Abstract

The purpose of this study was to determine the antioxidant activity and sunscreens of phenolicfraction of waste corn cobs (Zea mays L.). Corn cob that has been extracted using the winddried reflux for 2 hours. Furthermore corncob extract liquid successively fractionated using nhexane, ethyl acetate, butanol and ethanol 70%. Analysis of total phenolic content, freeradical scavengers activity determination using DPPH (1,1-diphenyl-2-picrylhidrazil) andsunscreen effectiveness by determining in vitro SPF values by spectrophotometric method.The results of this study indicate that the ethyl acetate fraction has a higher content of totalphenolic fraction followed by butanol, 70% ethanol and n-hexane. Total phenolic content of arow is 96.42;35.81;19.18 and 11.52 mg/mL. Ethyl acetate fraction also has a free-radicalscavengers activity is higher than the other factions. The research concludes phenolic fractionof ethyl acetate can act as an antioxidant as well as a sunscreen with the highest SPF valuefollowed fraction 70% ethanol, n-hexane and butanol. Abstract

The purpose of this study which are to identify and determine the rate of borax in meatballs snackssold in the city of Manado. Sampling locations are Bunaken, Malalayang, Mapanget, Sario, Singkil,Tikala, Tuminting and Wanea sub district. Each location is determined each 2 meatballs snacksvendors. Recruitment is done 3 times in each meatball vendor meatball snacks of 20 seeds, until atotal of 60 pieces of meatball samples in a month for each vendor. The samples were identified usinga flame test method and test turmeric paper color method. Identification experiment results of researchmeatballs with borax in the samples and test reaction flame color test please note that all samplestested meatball not harmful preservatives.Key words : Borax, flame test, color test, meatball, Manado

Abstract

This study aimed to determine the effect of ethanol extract eggplant rind (SolanummelongenaL.) on blood sugar levels in white male wistar induced by sucrose. Extraction is done bymaceration with 95% ethanol. The subject of the experiment were 15 white male wistarwhich were divided into 3 (three) groups namely positive control group using metformin 9mg/Kg body weight, negative control group using CMC 1% and treatment group with thedose of 0,02 g/200 g body weight, 0,01 g/200 g body weight and 0,1 g/200 g body weight.The data is processed using ANOVA and LSD test to compare between the positive controlgroup, negative control group, and treatment group. The analysis revealed a significantdifference between negative control group and treatment group, negative control group andpositive control group, but did not show a significant difference between positive controlgroup and treatment group.Key words : Solanummelongena L., Eggplant Rind, Blood Sugar Level, Sucrose

Abstract

Identification and preservation of genetic diversity of a population is important inconservation. This study aims to determine the MATK gene variation among tomato plantsfrom several places in Sulawesi and compare it with the closest relatives in GenBank, as wellas create a phylogenetic tree. PCR technique was used to amplify matK gene with universalprimers. ‘Kinky’ and “Apple” tomatoes were obtained from Minahasa regency of NorthSulawesi province. Both have an identical sequence of partial matK gene. “Cherry” tomatoobtained from North Minahasa regency of North Sulawesi province has a sequence of partialmatK gene which is identical with “cherry” tomatoes obtained from the district of Poso inCentral Sulawesi province. This study implies that both “cherry” tomatoes are the ancestor ofboth “kinky” and “apple” tomatoes.Key words : Genetic diversity, matK gene, Lycopersicon esculentum, phylogenetic tree
