Abstract

An experiment on application of immunostimulant bacterin in order to prevent Viral Nervous Necrosis (VNN) in seed production of humpback grouper, Cromileptes altivelis has been conducted in Gondol Research Institute for Mariculture hatchery. The experiment arranged in completely randomized design with two replicates. One thousand and five hundred fry with total length 4 cm were treated as followed ; 1 ml bacterin/L sea water by immersion (Treatment A), 1 ml bacterin/kg pellet by oraly (Treatment B), and without bacterin as a control (C). Bacterin was immersion and oraly every 10 days for 60 days rearing period. Challenge test with VNN was carry out at day 30 and 60. At the end of experiment the non-spesific immune were examined. Result of challenge test performed at day 30 showed that treated fish gave positive response to bacterin treatment. The highest survival showed by treatment B (65.0%) followed by treatment A(50.0%) and C (22.5%). The same result also showed after challenge test at day 60. The highest survival was 72.5% (treatment B) followed by 67.5% (treatment A) and 35.0% (control). Phagocytic activity (PA) of treatment B was 15.5%, and 14.0% from treatment A compared to 8.0% in control. Lysozyme activity (LA) of treated groups were also higher than control namely 1.8 cm for treatment B followed by 1.6 cm for treatment A, and only 1.2 cm in control. Addition of bacterin immunostimulant in feed gives is survival rate higher compared to way of immersion and control. Statistically non-spesific immune response of humpback grouper was significant different among treatment (P<0,05).

Abstract

The aim of this study was to know the optimal salinity level on survival and growth rates of Colossoma macropomum on larvae rearing in different salinity media. The treatment tested with six different salinities were 0; 2; 4; 6; and 10 ‰. This study was conducted from May 15 to June 12, 2006. The experiment design was arranged in completely randoumizes design with six treatments and three replications. Stock density was 1 fish/l with mean initial body weight 0.48 ± 0.03 g and initial total body length 3.04 ± 0.18 cm. The result showed that the optimal salinities between 5.74 – 6.35 ‰, which was giving growth rates 9.31 % and survival rates 100 %. On minimum salinitiy media (0 ‰) showed growth rate 5.45 % and survival rate 19.17 %.

Abstract

The objectives of this research were to find out the distribution and abundance of macrozoobenthos in Kupang Bay and to evaluate the suitability of this water for mariculture activity in the future based on physical and chemical water properties. Samples were collected from 10 stations along shoreline of the bay. Water samples were collected by cammerer water sampler, while benthic organisms were collected by ekman dradge. Samples were transferred to laboratory to identify and analyze. The results showed that the diversity index of the macrozoobenthos was moderately stable, and there were 4 to 10 species found in each station of Kupang Bay. The species dominancy was ranged from 28-82%, and the most dominant was Nassarius pullus. The water quality indicated that the study site was suitable for mariculture activities. It leads to suggestion that for the seaweed and floating net cage culture should be located in the mouth of the bay, while for pearl culture should be located along channel between Semau and Timor Islands.

Abstract

The aim of the study was to know the effect of soy sauce waste as substitution of fish meal in diet on the growth and dietary nutrient digestibility of juvenile tiger grouper (Epinephelus fuscoguttatus). The experiment used completely random design (CRD) with six treatments and three triplicates. The initial body weight of juvenile tiger grouper was 4.03 g and total length of 5-7 cm reared in 18 polycarbonate tanks with each 30 liter volume. Each of tanks was equipped with aeration and flow through water system. Six formulated diet with dry pellet form containing different level of soy sauce waste as substitution of fish meal, i.e., 0%, 7%, 14%, 21%, 28% and 28% added 0.15% methionine amino acid were teste. The result showed that the utilization of soy sauce waste as substitution fish meal influenced absolute growth, specific growth rate, feed conversion ratio and protein retention (P<0.05), but not significantly different (P>0.05) on survival rate of juvenile tiger grouper. Dietary nutrient digestibility (protein) obtained 98.05- 99.69%. Dietary nutrient digestibility decreased with utilization of soy sauce waste in diet. These experiments concluded that the utilization of soy sauce waste as substitution of fish meal could be used until optimal limit 21%, while the substitution of 28% can’t used for diet formulation for tiger grouper (Epinephelus fuscoguttatus).

Abstract

The aim of this research were to know the correlation between water physic-chemistry and the abundance of phytoplankton. The studies were conducted from May to September 2006 at Maitara water and samples were taken from 5 (five) stations for 4 (four) period by filtration method. The result showed that there were 12 genera of phytoplankton from 4 classes: Bacillariophyceae (7 genera), Chlorophyceae (1 genera), Cyanophyceae (1 genera), and Dinophyceae (3 genera). The phytoplankton abundance ranged from 5781 – 39227 individual/L, the highest value was on station 5 period III (39227 individual/L) and the lowest on station 3 period IV (5781 individual/L). The range values of phytoplankton biological index such as diversity index (H’) was 0.6889 – 1.1841, equitability index (E) = 0.4969 – 0.7215, and dominant index (D) = 0.3856 – 0.6183. Physical-chemistry parameters of the water were 0.01 – 2.24 mg/L of nitrate, 0.09 – 0.28 mg/L of orthophosphate, 0.040 – 0.042 of silicate, 6.21 – 8.27 of pH, 30 – 35 ‰ of salinity, 28.4 – 31.5oC of temperature, and 1.10 – 2.75 m of transparency, respectively. Multiple regression analysis showed that there was relationship between waters physical-chemistry parameters and phytoplankton abundance (R2 = 0.796), where the regression equation was Y = 150190.2 + 36.589 nitrate + 597.043 orthophosphate + 1002598 silicate + 25.289 pH - 23.866 salinity – 561.222 temperature – 56.973 transparency.

Abstract

The goal of this activity was to improve the productivity of Siamese catfish (Pangasius hypophthalmus) backyard hatchery. The research was conducted to identify the optimum density of larvae in the intensive rearing system. Two days post hatching larvae of P. hypophthalmus reared in 18 fiberglass containers size 40x30x30 cm3. The treatment used was 50, 100 and 150 fishes/litre in 6 replicates. A water recirculation system was used in this study and a hi-blower aeration system for oxigen supplay was added in it. The result showed that the density of larvae was not significant difference (P<0,05) for the survival rate at day-30, i.e. 21.34%; 19.78% and 19.71% for 50, 100 and 150 fishes/litre density of larvae, respectively. But the density of larvae was significant difference (P>0,05) for body weight at day-30, i.e. 0.47; 0.34 and 0.25 g/fishes for 50, 100 and 150 fishes/l, respectively.

Abstract

The purpose of this research was to identify the reproductive biology in the yellow rasbora (Rasbora lateristriata) in habitat of the Ngrancah River. To identify spawning events and embryo development, direct observation was employed during the spawning season in their natural environment, and then fertilized egg was incubated insitu. Spawning seasons underwent correspond to the end of rainy season and early dry season which characterized by clean freshly water and low temperature. The result showed that during spawning season, the broodfish migrated from Sermo Reservoir upward to the main river to find out the spawning site. In the early morning between 03.00 and 05.00 AM, the broodstock moved to the spawning site, then making aggregation and both female and male released their gametes in the shallower place. The aggregations were consisting around of 1 female and 3 males. The fertilized eggs would cleavage, and then embryo developed and yolk sac larval hatched within 23 hours at 27oC. The knowledge about spawning events of yellow rasbora may be used for improved management tools in the future.

Abstract

The purpose of this study was to determine allozyme genetic characters of the spotted galaxid in western Victoria, Australia. Two populations, Barongarook Creek (landlocked population) and Dutton Creek (diadromous population), of the spotted galaxias (Galaxias truttaceus) was investigated on their genetic characters by allozyme electrophoresis methods. From 23 system enzymes and general protein, 39 putative enzyme-coding loci were resolved, 3 of which were polymorphic at Esterase-1 (Est-1), Glycerol-3-phosphate dehydrogenase-2 (Gpdh-2), and Phosphogluconate hydrogenase (6pgd). Fixed allozyme differences were found at Est-1 locus indicating that incipient change in genetic character have been started in the landlocked population.

Abstract

The aim of this experiment was to find out the suitable stocking density of sea weed as shelter to growth and survival rate of swimming crabs (Portunus pelagicus) reared in brackishwater ponds. The research was conducted in nine brackishwater ponds with the dimension of 250 m2 for 120 days. The treatments different stocking density of sea weed (Gracillaria sp.) as shelter were applied, namely A: 500 kg/ha, B: 750 kg/ha, and C: 1000 kg/ha, with three replicates, respectively. Crablet 9th with carapace width 5,2 + 0.2 mm and body weight 0.04 + 0.02 g were introduced into each experimental ponds with density of 3 individu/m2. The crabs were fed with trash fish 15% of body weight in the first month, and then decreased to 5% in following month, twice a day. The experiments were design as complately randomized design. The result showed that the shelter were significantly increased to the survival rate and yield of swimming crabs (P<0.05), but not to the growth of charapace width and body weight. The best of survival rate and yield was obtained on the density of shelter at sum 1000 kg/ha.

Abstract

The objective of this research was to know the condition of coral reef in Kulisusu Bay of Muna. Field observation with scuba diving apparatus was done in February 29th, 2000 located in 3 stations and within the depth of 3 to 10 m. Line intercept transect method was used along 100 m of plastic rope. The data of coral reef were analysed by using dBase III+ and Lifeform packet program. The result showed that coral reef condition in two stations were generally in good condition. Fair condition was observed only in station I. The life coral reef was dominated by hard coral Acropora, namely Acropora branching, Acropora Encrusting, Acropora Submassif, Acropora digitate, and Acropora tabulate. While non Acropora namely coral branching, coral encrusting, coral foliose, and coral massive, Milepora and Heliopora were found acrros the coral reef. On the other hand the most abudance benthic fauna were mainly dominated by sponge and soft coral.
