Abstract

ExplorationofSugar Palm (Arenga pinnata Merr) in Tomohon,North SulawesiIndonesia has high genetic diversity of sugar palm, although until now sugar palm germplasm had been collection it’s about 11 acession’s. If sugar palm acession’s not conserved, it apprehensive sugar palm’s accession which is have promising gen’s will occur genetic’s erosion. The research activity was conducted on January, 2009 at Tomohon North Sulawesi, there were Tara-tara, Pinaras and Woloan Village. The purpose of palm sugar germ plasm exploration is to increase genetic diversity of sugar palm germplasm collection which is use as breeding material in compose superior palm sugar and seeds resources in short term. The result showed that on that three villages has been found three accessions of palm sugar namely Tara-tara sugar palm, Pinaras sugar palm and Woloan sugar palm. The highest production and sugar content of toddy to be found in tall sugar palm at  Tara-tara, is about 25-38 lire/bunch/day. Selection for improvement population of three accessions is needs to carry out on production and sugar content of toddy.

Abstract

Genetic Diversity of Bali Tall (DBI) and Sawarna Tall (DSA) based onRandom Amplified Polymorphic DNA (RAPD)Genetic Diversity of Bali Tall (DBI) and Sawarna Tall (DSA) based on Random Amplified Polymorphic DNA (RAPD). Ten Plants were used in each population. The objective of this research were to determine genetic diversity within- and inter-population of Bali Tall (DBI) and Sawarna Tall (DSA) coconuts, and genetic relationship of those population based on RAPD ( ). Research was done in Plant Biology Laboratory of CenterResearch of Genetic Resources and Biotechnology, Institut Pertanian Bogor, February – May 2007. DNA extraction was done by modified Rohde method and to determine the concentration and quality of DNA by Sambrook method. Ten RAPD 10-mer were used namely OPA-02, OPA-08, OPA-10, OPA-13, OPA-20, OPB-08, OPB-11, OPB-12, OPB-15, OPB-20. Genetic relationship between individuals of DBI and DSA coconut populations based on NTSys ver.2.0 programe (Program Numerical Taxonomy and Multivariate Analysis), and clustering analysis based on UPGMA method. These ten RAPD primers used could separate DBI and DSA in each group. Genetic diversity within-population of Bali Tall coconut population range from 2.4% to 30.7% with average 21.7%. So that, opportunity to improve characters in DBI coconut population could be done by selection. Genetic diversity within-population of Sawarna Tall coconut population progressively narrow, range from 1.5% to 12.4% with average 12.7%, so that selection in order to character improvement in this population could be done selectively. Genetic relationship between DBI dan DSA populations far enough (54%), so that crossing between those population will be good for characters improving.

Abstract

Path Analysis of Yield with Vegetative Characters of Arecanut Accession from Bolaang MongondowThe research was conducted from May 2008 to October 2009 at Kayuwatu Experimental Garden, Manado, North Sulawesi Province. The purpose was to evaluate the direct and indirect effect of vegetative characters on yield character of Bolmong arecanut accession. Path analysis method was used in this experiment. The research result showed that from nine characters, there are four characters had direct effect on yield. They were, number of leaves, number of leafscars, upper inter-nodes, and middle internode. These characters had better relationship with yield character than other characters.

Abstract

Use of Commercial Yeasts on Etanol Processing from Palm NeeraThe objective of this research was to find out commercial yeasts which effective for fermentation sugar palm neera become etanol, efficient processing technique and it can be operated by the farmers/the farmer groups. The research was done from March to June 2009, in the Laboratory of Indonesia Coconut and Other Palmae Research Intitute. The research was conducted in factorial experiment 5 x 4 x 2, using completed randomized design. Factor A is commercial yeast, which consists of A1 = Fermipan, A2 = Mauri-pan, A3 = Gold-Pakmaya, A4 = BrewMax, and A5 = Without yeast. (Fermipan, Mauri-pan, Gold-Pakmaya, BrewMax and without yeast. Factor B is fermentation period which consists of B1 = 0 day, B2 = 2 days and B3 = 3 days. Ethanol processing was conducted by using fermentation and distillation process. The result showed that the suitable yeast for fermentation were Gold-Pakmaya and Fermipan yeast. Using a single destillator, laboratory scale, the first and second processes, produced etanol 28-43% and 72-77%, respectively. Processing etanol 30-32% and 72-77% is economically feasible.

Abstract

Engineering Design of Bioethanol Processing Machine from Palm NeeraThe research was conducted in Laboratory and engineering workshop of Indonesian Coconut and Palmae Research Institute, Manado from May to September 2009. The activities consisted of Bioethanol processing equipment, field testingof bioethanol product as machine fuel engine. Raw material characteristics (crude ethanol), material balance, physical conditions and processing system of unit processes and economically processing analysis were observed. Research result showed that use of bioethanol equipment could improved ethanol content of crude ethanol from 13-30% to 80-94% and from 83% to 95-97%. Mass balance varied and depended on characteristic of crude ethanol and heating process. Ethanol 97% could be used as machine engine with ratio of gasoline and ethanol 90:10. Use of bioethanol and gasoline blend can be used more efficient 12.5-29.0% than use of gasoline. Processing of crude ethanol 25-35% to produce ethanol 90-95% is profitable. The bioethanol equipment efficient to be used under farmer scale and medium industries scale.

Abstract

Influence of Fermentation, Filtering and Storage Time on Quality ofVirgin Coconut Oil (VCO)Virgin Coconut Oil (VCO), is a product of the processing without the addition of food additive, that is categorized as natural products. Since 2003, the product is known of consumer because it is very beneficial for health, so its expensive, so many emerging ways of processing. Therefore, in 2004, has conducted research to find practical ways for the farmers, including fermentation of coconut milk (3 hours and 12 hours), filtering (using filter paper 42 mess, straining cloth and only muted 3 days ), then VCO observed at 0-month storage, 2 months, 4 months and 6 months. Observations carried out on the water content, free fatty acid and peroxide numbers. The results showed that water levels are influenced by long storage and interaction of fermentation coconut milk, filtering and storage. Free fatty acids is not affected by any treatment, while the peroxidevalue affected by long storage. VCO water content of 0-6 months of storage ranged 0.07-0.13%, free fatty acids ranging from 0.17-0.23% and the peroxide value ranges from 0.19-3.02 meq/kg oil. The range of water content, free fatty acid and peroxide value, still meet the standards of Asian Pasific Coconut Community (APCC), the water content ranges from 0.1-0.5%, maximum free fatty acids 0.5% and a maximum peroxide number 3 meq /kg. Based on the results of this study, the VCO processing by applying the processing is done, continue to produce VCO that meet the standards set APCC.

Abstract

Inbreeding Depression Analysis Based on Microsatellite (SSRs)Markers in Mapanget Tall CoconutThe objective of this research was to determine inbreeding depression based on microsatellite (SSR) markers of second, third, and fourth generations of selfed Mapanget Tall coconut No.32 (DMT-32). The research was conducted in Plant Biology Laboratory, Research Center of Plant Genetic Resources and Biotechnology, Institut Pertanian Bogor. The research was done since May 2004 until May 2007. Fifteen out of 19 microsatellite ( , SSR) loci (primers) were used to analyze inbreeding depression in each generation of selfed Mapanget Tall coconut No.32. Based on 15 loci of microsatellite (SSRs) markers, showed that inbreeding depression expressed and have tendency to increase from generation to next generation of selfed Mapanget Tall coconut No.32 (DMT-32). Inbreeding depression in DMT-32 S2 (2.78%), DMT-32 S3 (10.54%), and DMT-32 S4 (15.74%). Four out of 15 microsatellite loci used were not in Hardy-Weinberg equilibrium, means that in DMT-32 S4 those loci were homozygote that’s why alleles of those loci were not inherits in equilibrium. DMT-32 S4 coconut can be used as plant materials to find progeny with higher heterosis level.

Abstract

Adoption of Coconut Cultivation Technology on Attacked SexavaPopulation in Talaud Island RegencyThis research was conducted by survey method with descriptive-correlational approach. The research was conducted in Kabaruan and Beo Sub District in Talaud Island Regency. Total of two villages were chosen randomly in each Sub District. In each village, 30 farmers were chosen randomly as respondents, making total of 120 farmers.The result showed that the adoption level of technology of coconut cultivated in Talaud Island Regency could be divided in to three categories, low level (35.83%), moderate (50.83%), and high level of adoption (13.33%).
