This study aimed to analyze the effect of combined picloram and kinetin treatments on callus induction from young leaf roll explants of sugarcane variety Bululawang. The experiment was conducted using a completely randomized design with one factor consisting of 16 treatment combinations of picloram (0, 2, 4, and 6 ppm) and kinetin (0, 0.5, 1, and 1.5 ppm). The observed parameters included the time of callus initiation, percentage of explants forming callus, fresh weight, dry weight, and callus morphology. The results showed that the addition of picloram and kinetin combinations did not significantly affect the time of callus initiation but significantly increased the percentage of explants forming callus (averaging 86.72%) compared to the control (12.50%). The highest fresh and dry weights of callus were recorded at the treatment of 4 ppm picloram + 1 ppm kinetin, with a fresh weight of 0.4412 g and a dry weight of 0.0650 g. Callus morphology was predominantly compact in texture with a yellow to yellow-reddish color. The combination of picloram and kinetin effectively increased the percentage of callus induction, fresh weight, and dry weight of callus in sugarcane, indicating its potential application in tissue culture and secondary metabolite production.

This research aims to isolate actinomycetes that produce the enzyme L-asparaginase from Olo Beach and to determine the activity of the L-asparaginase enzyme produced. Actinomycetes producing the enzyme L-asparaginase were obtained through the following steps: isolation on SCA medium using the spread plate method; selection of L-asparaginase-producing actinomycetes on M-9 liquid medium with phenol red as a color indicator; production of L-asparaginase based on the principle of fermentation in production medium, followed by measurement of L-asparaginase activity and protein content. Protein content was determined using the Lowry method with Bovine Serum Albumin (BSA) as the standard. The data obtained were analyzed using ammonia and BSA standard curves. The results showed that 11 actinomycetes isolates from Olo Beach were capable of producing L-asparaginase. Qualitative selection showed a change in media color from yellow to pink, indicating the presence of L-asparaginase activity. Quantitatively, the highest L-asparaginase enzyme activity was 40.688 U/mL and the highest specific activity was 123,677 U/mg, found in isolate SPO 6. The lowest enzyme activity was 31,651 U/mL and the lowest specific activity was 25,874 U/mg. Specific activity is influenced by enzyme activity and protein content.

This study aims to stimulate the effectiveness of antibacterial solutions and methanol extracts of lo'i keta, a traditional medicine from Bima-Dompu, West Nusa Tenggara, against the growth of both bacteria. Lo'i keta solutions and methanol extracts were tested at concentrations of 25%, 50%, 75%, and 100% using the agar well diffusion method. The inhibition zones formed were measured to assess antibacterial activity. Ciprofloxacin was used as a positive control, and distilled water as a negative control. The results showed that lo'i keta solutions and methanol extracts had inhibitory power against Staphylococcus aureus and Streptococcus pyogenes. Its effectiveness increased with increasing concentrations of the solution and extract. The best activity was shown at a concentration of 100%, approaching the positive control. This study shows that lo'i keta has the potential as a natural alternative to fight gram-positive bacteria, because resistance to antibiotics is increasing and lo'i keta can be used as a traditional herbal medicine, but there has not been much scientific exploration.
