Berkala Penelitian Hayati is a half yearly international peer reviewed, an open access life science journal. Since April 2012, the journal was changed into English. This journal is indexed by DOAJ, Crossref, Google Scholar, Academia.edu, and EBSCO Host. This journal publishes original research, applied, review article, and educational articles in all areas of biology. Authors are encouraged to submit complete unpublished and original works that are not under review in other journals. Abstract

Mangrove snail is one of the animal medicines used againts bacterial infection, however, the scientific basic of the activity still void. The aim of this research were to investigate the antimicrobial effect of mangrove snail as the primary step to identify an antimicrobial activity and find out substances responsible to this activity. The study was conducted by collecting 4 snail samples. Extract were made be maceration of methanol and followed by chloroform and n-butanol partision, respectively. Antimicrobial activity has been done using Salmonella typhii, Staphylococcus aureus, Escherichia coli and fungi Candida albicans. The result showed that these mangrove snail capable inhibit bacterial growth either Gram positive or negative, but can not inhibition fungi. Methanol extract of Terebralia sulcata showed inhibition zone has the Rf 0.27, whereas chloroform fractions of Charonia sp. has 6 spots by bioautographic. Two out of six spots showed inhibition zone with Rf 0.16 and Rf 0.4. Methanol extract of N. birmanica has inhibit with Rf 0.56. One spots with Rf 0.42 unclear as inhibition zone suggested negative. Then the methanol extract of Telescopium telescopium with Rf 0.34 showing inhibition zone.

Abstract

Caffeine affects activity of enzyme of polimerase DNA, induce the mitosis of cells mammal of before replication DNA ended the perfection, and also pursue the activity of enzyme fosfodiesterase is hence anticipated by a potential caffeine generate the developmental defect, for example can pursue the process of ossification endochondralis in growth plate. This present research studied the effect of caffeine gift by oral on pregnant dam during organogenesis to structure of histologi growth plate tibia foetus. Twenty four pregnant mice (6 per group) were treated by gavage with 0 (control), 40, 80, 120 mg/kg b.w caffeine from gestation day 6 to 15. On day 18 of pregnancy, fetuses was removed from dams by caecarean section.. Observation of histological structure of the tibial growth plate preparation by paraffin method (Hematoxylin-Eosin staining). The result showed that caffeine cause slightened proliferative zone, maturation zone, and cartilage calcification zone on the mouse tibial growth plate.

Abstract

Brommelain is an enzyme hydrolyze most soluble protein easily and efficiently. This enzyme is used in many industry like food industry. This research aimed to isolation and characterization crude extract brommelain. This enzyme has been extracted from the stems of pineapples to produce crude extract, precipitated with amonium sulfat, and enzyme activity to decided with Bergmeyer methode. The higher activity was 1,996 U/ml in precipitate 40-60% amonium sulfat. Optimum temperature and pH to decided temperature and pH variation was detected based on enzyme activity. Characterization to indicate that bromelain has an optimum temperature at 55°C, optimum pH of 7, KM = 5.074 mg/ml and Vmax = 0.666 mg/ml.second.

Abstract

Study of bioaccumulation of heavy metal at species of macrofauna benthic in Kangean’s archipelago, Madura have been done at February 2004. This study was conducted in the effort data base compiling bioaccumulation of heavy metal, especially in Indonesia. As one of part of “Mussel Watch” program what have been executed by cleft of world angle. Kangean’s archipelago has been selected because its situation was very cloistered and far from modern industrial activity, and also inexistence of data about heavy metal’s accumulation at macrofauna benthic. Sampling has been done in 2 station around island Arjasa (subdivision Arjasa), precisely in Erreng at 6°53’00” S and 115°14’26’’ E and Bilis-Bilis at 6°50’45’’ S and 115°14’54’’ E. Sampling has been conducted by using method of direct collection, and then treated in laboratory to analysis by AAS Spectra AA 250 plus of method flame. Species of Nassarius globosus, Anadara Scapha (big) and Saccostrea cucullata accumulate highest metal of Cu, Cd and Zn (in successively) 192,53 ± 154.65 μg/g dry weight, 13.30 ± 1.55 μg/g dry weight, and 1816.02 ± 367.28 μg/g dry weight.

Abstract

Energy synthesis for sperm motility in the mitochondria with enzymatic reactions in nucleus and mitochondria is called Oxidative Phosphorylation (OXPHOS), in which the respiratory process is coordinated in nucleus and mitochondria. Enzymes in OXPHOS process are complex enzymes I, III, IV and V in the mitochondria and complex enzyme II in the nucleus. If there is OXPHOS dysfunction, the mutation of mitochondria DNA (mtDNA) will decrease energy (ATP, Adenosine Triphosphate) production and increase Reactive Oxygen Species (ROS). The characteristics of ROS are oxidant for lipid, protein and DNA, and all these reactions lead to the dysfunction of OXPHOS and, in the spermatozoa, they will decrease motility. The detection of ROS, Malondialdehyde (MDA), 8-hydroxydeoxyguanosine (8-OH-dG), and sperm motility can use Chemiluminicense method, Spectroflourometric method, High Performance Liquid Chromatography (HPLC) method and Markler method, respectively, as World Health Organization (WHO) Procedures Laboratory. These results indicated there was significant indirect correlation between ROS with sperm motility and direct correlation between ROS with MDA, 8-OH-dG.
