Kutai National Park (KNP) in East Kalimantan, Indonesia is blessed with natural mangrove forests. However, there is only very little information relating to the species composition and mangrove diversity. Therefore, this study was conducted to assess the mangrove tree species diversity and richness in Kutai National Park, East Kalimantan, Indonesia. The study results showed that the mangrove trees density was 1,214 trees/ha in the line transect vegetation survey with 1.7 ha plot. The number of mangrove species in KNP was 17 from 6 families. The dominant species with large numbers of individuals were Rhizophora mucronata, Rhizophora apiculata, and Bruguiera sexangula. The diversity index was low in mangroves in Pandan Bay Estuary, and Kaba Bay compared to that in Lombok bay’s mangroves.

Short Communication: Digenea parasites in giant mudskipper (Periopthalmodon schlosseri) in Kuala Lupak Village, South Kalimantan, Indonesia. Biodiversitas 20: 3782-3787. Parasites are organisms that impart detrimental effects on their host. Monogenea which are generally ectoparasites and digenea which are endoparasites are the most common parasites which invade fish. This research was aimed to find out the existence of monogenean and digenean parasites as well as their species in giant mudskipper or timpakul fish (Periopthalmodon schlosseri) in Kuala Lupak Village, South Kalimantan, Indonesia. Employing 25 giant mudskipper which was randomly collected from Kuala Lupak Village, South Kalimantan, this study explored the presence of ectoparasites and endoparasites on the mucous, fins, gills, intestines, stomach, and meat of the fish. The results showed that only digenean parasites were found in the giant mudskipper (Periopthalmodon schlosseri). The digenean parasites found to come from Opecoeloides genus and Buchepalidae family, metacercariae cysts, and parasites of the nematode phylum. Monogenea parasites were not found due to low fish-density, fish behavior that is rarely in the water, and adequate nutrition which is the opposite of growth-supporting factors of monogenean. Parasitic infection could occur because the fish eat some intermediate hosts of digenea, such as snails and other fish. Besides, the condition of waters that have been contaminated with digenean eggs allows the life cycle of the digenean to continue in fish.

Abstract. Andriyono S, Jobaidul Alam Md, Kim HW. 2019. Environmental DNA (eDNA) metabarcoding: Diversity study around the Pondok Dadap fish landing station, Malang, Indonesia. Biodiversitas 20: 3772-3781. Molecular identification of species is now fast growing and currently widely applied method in the diversity estimation of aquatic biota; even though morphological identification is still carried out. The molecular approach is beneficial complementing on regular surveys, e.g. use of nets, traps, fishing rods, and even with poisons. In this study, the eDNA metabarcoding was applied to water samples around the Pondok Dadap fish landing station, Indonesia to determine the diversity of fish around the waters and also to identify marine fish landed in this area. Molecular identification was carried out on fish samples obtained from the fish market improved GenBank database on COI and ITS. While, seawater samples were carried out by using the next-generation sequencing (NGS) platform to obtain the eDNA metabarcoding data for the first time. Molecular identification obtained 34 species (68 sequences of COI and ITS regions) belonging to 28 genera, 18 families, 4 orders, while the eDNA metabarcoding approach identified 53 marine fish species by using the MiFish pipeline representing 38 genera, 27 families, and 7 orders. From the present study, we can able to estimated fish diversity by eDNA metabarcoding, and this finding will be helpful for baseline data preparation for future effective management of resources in this area.

Indonesia is the world's second-biggest producer of cacao after Ivory Coast with its cacao plantations spreads over 1,652 million hectares and mostly managed by smallholders. Cacao as a beneficial commodity can provide job opportunities for over 1.64 million people, with its contribution of more than US-$ 1.6 billion/year to national income. However, pest and disease infections are inevitably common constraints for the cacao cultivators. One current disease is caused by Cacao Swollen Shoot Virus (CSSV). Despite its noticeable symptoms on the cacao trees suffering from the disease, the knowledge on both the virus existence and the prevention steps in dealing with it is lacking. The information on the diversity of mosaic virus will help us to comprehend its epidemiologic development and the needed countermeasures, as well as its evolution. This research is intended to study the mosaic disease virus molecularly, the virus was obtained from DR1 clones from Kalibawang cacao plantation in Kulon Progo, Yogyakarta. The virus morphology was observed using a transmission electron microscope applying quick dipping method. The PCR analysis with conserved region ORF1 primers was conducted to detect the viral existence in the infected trees. The identification of CSSV molecular characters was undertaken using PCR sequencing analysis which was then examined using BioEdit and Mega5 programs to initiate a relationship dendrogram. The result showed that the DR1 cacao tree clones from Kalibawang were infected by mosaic virus with mild visual severity of leaf typical symptom. From the electronic microscope observation, a ±100 nm rod-shaped viral particle with a diameter of 15.3 mm was found. On the molecular level, the cause of mosaic symptom has CSSV amplified at conserved regions with size 375 bp. The results are the first report confirming that the molecular cause of cacao mosaic disease in Indonesia is CSSV. The molecular characters of CSSV in Yogyakarta are very different from those found abroad.

The archaeal community in the soil surrounding rice roots plays key roles in biogeochemical cycles. Methanogenic archaea contribute to CH4 production in the rice field and control CH4 emission rates; an important greenhouse gas significantly emitted from the rice field. Characteristic of archaeal and methanogenic community in Indonesian rice field has not been well documented. In this study, rice plants grown in the rice field were treated with two different fertilizer applications, i.e., 100% urea without biofertilizer (250 kg ha-1) (B0), and 50% urea (125 kg ha-1) with biofertilizer consisted of methanotrophic bacteria and N2O-reducing bacteria (B1). Soil archaeal community of the two types of fertilization was characterized over the rice-growing period using 16S rRNA amplicon sequencing. The succession and dynamic of the archaeal population were detected in each of the two treatments before fertilizer application (0 days after transplanting [DAT]) until the rice generative phase (69 DAT). During the period, the archaeal soil community in both treatments was dominated by Crenarchaeota. The relative abundance of putative methanogens was higher in the B0 treatment than that of B1 treatment. Within the methanogenic community, Methanosarcinaceae and Methanomassiliicoccaceae which found as most dominant group in both treatments were presumably to be important contributors to CH4 emission. The application of 50% urea with biofertilizer (B1) changed the structure of the archaeal and methanogenic community in the rice field compared to the application of 100% urea (B0).

Abstract. Taibi W, Belletreche A, Kharsi M, Gaouar SBS. 2019. Phenotypic diversity for quantitative and qualitative characters of barley (Hordeum vulgare) accession from Algeria. Biodiversitas 20: 3794-3803. Little is known about the diversity of barley in Algeria. An inventory has been conducted in Algeria (North and South) to collect local accessions of barley (Hordeum vulgare L.). A collection of 34 traditional and exotic accessions of barley was characterized using 12 quantitative and 18 qualitative agro-morphological traits. The phenotypic diversity was determined by the Shannon-Weaver diversity index (H') at different levels (global, by type of barley and varietal name). The H' estimates showed a wide phenotypic variability for different traits with H’ average of 0.74 and 0.53 from quantitative and qualitative characters, respectively. The results of the multiple correspondence analysis and hierarchical clustering showed a clear distinction between the different accessions. The results of this work have revealed a great phenotypic diversity of barley accessions that only partially matches the names of varieties because of the existence of homonyms and synonyms in the names given by farmers. The information gathered from this study could be used in conventional breeding programs and in situ conservation of diversity.

Abstract. Nashrurrokhman M, Sayekti PR, Safitri A, Purwestri YA, Pratiwi R. 2019. Macronutrient and mineral contents of five local black rice (Oryza sativa) cultivars in Indonesia. Biodiversitas 20: 3647-3653. Nutrient composition is important information that can be used to support the development of black rice as functional food. In this study, the macronutrient and mineral compositions of five black rice cultivars were determined by measuring the contents of total carbohydrates, proteins, and lipids, and levels of the minerals Ca, Fe, and Zn of whole rice, refined rice, and rice bran. Rice samples were obtained from farmers who planted the black rice cultivars of Woja Laka, Toraja, Cempo Ireng “Seyegan”, Melik “Bantul” and Wedomartani. Proximate analysis and spectrophotometric methods were performed to measure the contents of macronutrients and minerals. Statistical analysis of all quantitative data was conducted by using One-way analysis of variance (ANOVA) (p<0.05) using the SPSS-25 program. Results showed that the contents of carbohydrates, proteins, and lipids in the five rice cultivars varied in the range of 61.80%-65.58%, 7.13%-9.10%, and 1.98%-3.23%, respectively. Carbohydrates found in refined rice were in the range of 62.71%-67.89%, while rice bran had protein and lipid contents varying in the range of 10.95%-11.48% and 5.70%-6.86%, respectively. Rice bran also had the highest mineral content and the highest mineral content was 30 ?g/g Ca compared to Fe and Zn. Therefore, the macronutrient and mineral composition varied among the five black rice cultivars and rice components. The highest carbohydrate content was found in refined rice, while the highest proteins, lipids, and minerals (Ca, Fe, and Zn) contents were found in rice bran.

Abstract. Asra R, Witono JR, Fijridiyanto I, Elva, Scheu S. 2019. Community study of dragon’s blood palm (Daemonorops spp.) in Harapan Rainforest, Sumatra. Biodiversitas 20: 3632-3640. Dragon's blood palms are rattan species that produce red-colored exudate on its fruit skin, popularly called exudates 'dragon's blood'. It has a high economic value and plays an important role as an income source for indigenous tribes in Jambi, such as in Bukit Duabelas National Park who live Anak Dalam Tribe and Semerantihan Village live Talang Mamak Tribe. However, overexploitation and unsustainable harvesting method have caused its decline in its population in natural forest. This study aims to make an inventory and map distribution of Dragon's blood palms in Harapan Rainforest, at the boundary between Jambi and South Sumatra Provinces, Indonesia; to determine its sex ratio and identify environmental factors that influence sex of Dragon's blood palms. This research used purposive random sampling method at three areas in Harapan Rainforest, i.e.: Hamparan Bato, Hamparan Batang Kapas, and Hamparan between Block A and B. The results showed that three species of Daemonorops were found in Harapan Rainforest, namely Daemonorops didymophylla, D. draco, and D. longipes. D. longipes has the highest population (18 clumps). The highest number of clumps were found in Hamparan Bato (17 clumps)and Hamparan Batang Kapas (17 clumps) areas. The ratio of male and female plants of D. longipes is 1: 1.84, D. draco is 1: 2 and D. didymophylla is 2.2: 1. Most clumps of Daemonorops species in Harapan Rainforest grow on the river banks which have low organic matters. Soil chemical variables that most contribute to the growth of Daemonorops spp. in Harapan Forest are N, C and pH.

Analysis of stomach content of piscivorous fishes caught in Danau Panggang Peatland, South Kalimantan, Indonesia. Biodiversitas 20: 3788-3793. The purpose of this study was to analyze the stomach content profile of piscivorous fishes caught at the reservaat station and the station without activity as The control station, in the Danau Panggang peatland. Sampling was carried out four times and the index of preponderance stomach content was determined in piscivorous fishes. The results showed there were nine fish species in the reservaat area, including Cryptopterus spp., Pangasius nieuwenhuisii, Chana striata, Anabas testudineus, Pangasius sp., Mastecembelus erythrotaenia, Macrognathus aculeatus, Mystus nigriceps, and Mystus nemurus. Their gut was dominated 89% by Chrysophyta, and 37.5% by Diatom and Navicula. In the area without activity, six species of fish were caught, including Cryptopterus spp, Pangasius nieuwenhuisii, Anabas testudineus, Pangasius sp, Mystus nigriceps, and Mystus nemurus, with 83% of their gut dominated by Diatom. The water temperature was 28.07-33.25º C, brightness (6-115 cm), pH of 4.23-6.97, dissolved oxygen (DO) (1.4-6.5 mg/L) and BOD (7.52-24.92 mg/L) i.e., water quality parameters that tend to be incompatible with the persistence of fishes. These limiting factors of peatland water and extreme conditions led to a limitation at availability of natural food for piscivorous fishes.

Abstract. Sudaryanto FX, Hardini J, Kalih LATTWS, Asrori MM, Suana IW. 2019. Bird communities and vegetation composition in Nusa Penida, Bali, Indonesia. Biodiversitas 20: 3676-3683. A study on bird community and vegetation composition in Nusa Penida was carried out to determine its diversity, conservation status, and habitat as an effort to conserve and develop birdwatching ecotourism in Nusa Penida. The study was conducted in Tembeling forest and Ped agroforestry. By using the point count method, we found 80 species of birds: 70 species in Tembeling forest and 79 species in Ped agroforestry. Fourteen species of birds are protected by Indonesian law. According to the IUCN Red List of Threatened Species, four species are Critically Endangered and one species is classified as Near Threatened. We also found eight species of migratory birds. Diversity of birds in Nusa Penida was high. Individuals of each bird species was spread evenly, except Hirundo rustica, Hirundo tahitica and Streptopelia chinensis were dominant. Twenty-five species of trees were found in Tembeling, while in Ped were found 22 species. These vegetations were provided birds with good resources for foraging, resting, and nesting.
