Abstract

Escherichia coli is commonly known as nonpathogenic bacteria, but some strains are pathogenic and cause diarrhea, urinary tract infections, and meningitis. Escherichia coli is initially sensitive to antibiotics, but lately began resistant. Alternative antibacterial can be obtained from extracts of certain plants, such as mahogany (Swietenia mahogany). This research aimed to determine the effect of various concentrations of ethanolic extract of mahogany seeds to growth inhibition of  Escherichia coli FNCC 0091 and determine the concentration that could optimally inhibited the growth of Escherichia coli FNCC 0091. Mahogany seeds extract was prepared by maceration method with 96% ethanol. Antibacterial activity was done by disc diffusion method. Data obtained were diameter of clear zone. Data were analyzed using one-way analysis of variance followed by Duncan's test. Results showed that the ethanolic extract mahogany seeds could inhibited the growth of Escherichia coli FNCC 0091. The best treatment was ethanolic extract of mahagony seeds at concentration 100% and 80% with clear zone of 2.33 mm and 2.13 mm.     Key words: Escherichia coli FNCC 0091; ethanolic extract of mahogany seed; discs diffusion method

Abstract

This research aimed to description the chlorophyll content, the metal cadmium accumulations in the leaves, and the growth of water spinach resulting from the furnishing of cadmium metal with various concentrations, detention time, and the interaction between the two treatments. The research was conducted in the Green House Faculty Department of Biology, Faculty of Mathematics and Natural Sciences, State University of Surabaya using a randomized block design (RAK). Cadmium metal concentrations used were 0 ppm, 8 ppm, 12 ppm, 16 ppm, and 20 ppm, with a detention time for 7 and 14 days. Parameters observed chlorophyll content, accumulation of cadmium in leaves of metal, and water spinach plant growth. Data were then analyzed using 2-way ANOVA and Duncan test continued with a significance level of 0,05. Results showed that giving with a concentration of cadmium metal and different detention time can affect the levels of chlorophyll, metals cadmium accumulation in the leaves, and Ipomoea aquatica Forsk growth. The smallest leaf chlorophyll content indicated on providing metal concentration of 16 ppm, and 20 ppm with detention time of 14 days. The mean greatest metal accumulation in leaves was shown at a concentration of 12 ppm, 16 ppm, and 20 ppm which is equal to 6.357 ppm, 6.765 ppm and 6.468 ppm. Giving a concentration of 20 ppm with a 14-day detention time showed the smallest end of the weight is 34.57 grams.   Key words: kadmium concentration; detention time; chlorophyll content; metals kadmium accumulation; growth water spinach

Abstract

Armyworm is one of the important pests of soybean. Entomopathogenic fungi is one of alternative biological agent to control soybean pests such as armyworms. This study aimed to determine the effect of Lecanicillium lecanii on armyworm mortality and the effective concentration of Lecanicillium lecanii to infect armyworms. The design of experiment was a completely randomized design (CRD). The study consisted of 27 treatments with 3 kinds of conidial density, i.e. 107, 108, and 109 conidia/ml. Each treatment repeated 9 times. Observed data were the mortality percentage of larvae and weight of residual feed after 7 days, analyzed using one-way analysis of variance (one-way ANOVA). The results showed that the fungi affects the armyworm mortality. The effective density was 108 and 109 conidia/ml  that could kill armyworms up to 80% and 83%.   Key words: Lecanicillium lecanii; mortality of armyworms

Abstract

Citizens of the pots using contaminated water wells to meet daily needs such as bathing, washing, cooking, watering and raw materials for drinking water. This research aims to test the effectiveness of water purification installation views of physical parameters required (temperature, and Total Disolved Solid), chemistry (pH) and based on the total number of Microbiology coliform and Fecal coliform. to improve the water quality. This research was observation using the approach of cross-sectional and target research is well water residents located next to the septic tank to be processed using simple water purification installation. Physical parameters observed were temperature and solids dissolved total, chemical parameters with degrees acidity (pH) and the parameters of microbiology with the amount of coliform bacteria total and Fecal coliform with the methods MPN. The data obtained were analyzed by qualitative and quantitative descriptive, then compared with Peraturan Pemerintah RI No. 82 tahun 2001 tentang Pengelolahan Kualitas Air dan Pengendalian Pencemaran Air. The results showed the Water Purification Installation can improve the quality of well water. This could be obsetved the total dissolved solids decreased from an average of 700mg/l, 434mg/l being, pH neutral, a decline in the number of total coliform is more than 2400 cells per 100 ml of cells to 140 and Fecal coliform 170 cells per 100 ml of cells to 26. standard meets the criteria so that clean of Peraturan Pemerintah RI No. 82 tahun 2001 tentang Pengelolahan Kualitas Air dan Pengendalian Pencemaran Air. . Abstract

Mud crabs from a natural catchment have a high economic value. From some of the natural catchments, there are soft shell crabs with relatively long moulting periods, about 2 – 3 months. Soft shell crab needs always increase. So, an intensive cultivation is necessary to produce moulting crabs. The purpose of this research was to identify the influence of walking legs mutilations and eyes stalks ablation to the male mud crab moulting periods. This research used experimental research type and completely randomized design (RAL) with three replications. The treatments consisted of mutilation of a pair of the first walking legs, a pair of the second walking legs, a pair of the third walking legs, a pair of claws, a pair of claws and all walking legs, and eyes talk ablation. Crabs used was mud crabs with hard carapaces. The test parameter was moulting period of mud crabs measured from the crabs still had hard carapaces until their moulting were complete. Data obtained in the form of moulting period of mud crabs using mutilation and ablation methods and control mud crabs were analyzed descriptive quantitatively. The results indicated that the methods of mutilation and ablation can accelerate mud crab moulting periods. Mutilation of a pair of claw and all walking legs is the treatment that produces the best moulting time compared with others with their moulting duration are 17-23 days.   Key words: mutilation; ablation; mud crab; moulting period; moulting

Abstract

One of the soft shell crab production technology is using spinach extract to stimulate moulting. Spinach extract contains phytoecdysteroid, a substance which is well known to stimulate moulting in mudcrabs. Ecdysteroid synthesis is a process that requires the presence of Ca2+ as a trigger. The purpose of this study were to determine the effect of various concentrations of lime water and spinach on feed to the duration of moulting and get a combination of lime water and spinach concentration that produces the shortest duration of moulting on male mudcrabs (Scylla serrata). This research used Randomized Completely Block Design (RCBD) with factorial pattern that consists of factor I: 5 levels of lime water concentrations (0, 2, 3, 4, dan 5 ml) and factor II: 5 levels of spinach concentrations (0, 10, 20, 30, dan 40 g) with 3 repetitions. Data obtained were analyzed by Friedman test. The results showed  that the addition of  lime water and  spinach combinations with various concentrations on the feed have  different effects to the duration of moulting of mudcrabs. The shortest duration of moulting was obtained in the treatment of  A4B40 formula, add with 4 ml of  lime water and 40 g of spinach to th feed,  in average of 20 days.   Key words: ecdysteroid; lime water; Ca2+; spinach; “moulting” duration; mudcrab (Scylla serrata)
