Abstract

The development of sweet potato cultivation in tidal land has begun. These activities require an appropriate invention in agricultural technology. An integrated management in physical, chemical and biological aspect also needs to be done. The content of soil microbes in tidal land is low, but it’s fairly high diversity can be used to improve the soil fertility. This paper describes some success of microbiological studies on tidal land. Further attempt is by integrate the potential of soil microbes utilization which supported by LEISA (Low External Input Sustainable Agriculture) technology.The success of this activity supports the national food security program and realizing sustainable agricultural system in the utilization of tidal land.

Abstract

This study aims to analyze and describe the relationship between altitude, aerial variables (temperature, light intensity, humidity), water qualities (water temperature, pH, BOD, COD, DO, TOM, and water velocity), and vegetation with the diversity of Odonate assemblages. Odonate samplings were conducted at six survey sites based on altitude and vegetation characteristics. Measurement of altitude, aerial variables, water qualities and vegetation characteristics were replicate in the first day and third day. Analysis of correlations of all environmental factors with the odonate diversity was done through structural equation model using Partial Least Squares (PLS), Open source Smart Software and Microsoft Excel. The aerial variables and water qualities affected indirectly on odonate diversity. The aerial variables directly or with interaction to other factor affected the water qualities and vegetation characteristics. The vegetation characteristics directly influenced to odonate diversity. Water flow affected water quality, light intensity affected the aerial, while morning period observation affected the odonate diversity. Predictive relevance (Q2) for a model designed amounted to 99.95%, while the rest of 0.05% are explained by other variables.

Abstract

Dog conch (Strombus turturella) is one of important sources of fishery commodities with  high economic values in Bangka Belitung. The needs of Dog conch as consumption still depends on fishermen harvest since its aquaculture has not been cultured until now. This study aims at determining behavior and feeding habit of dog conch. The study was conducted in July 2014 to January 2015. The experiments passed some stages 1) samples was taken in the field by hand collecting, 2) the experiment of seagrass consumed preference, 3) feeding behavior, and 4) Analysis of the stomach contents. The results indicate dog conch does not consume seagrass immediately, but detritus and microalgae species of Thalassiosira sp., Synedra sp., Nitzschia sp., Navicula sp. Dog conch eats by put the probiosis of the shell, then it grinds sediment layer, seagrass leaves and shells of other individuals as source if food. Dog conch behavior from beginning to  ending of the observation includes several stages: 1) Initial treatment actively foraging, 2) Individuals dog conch assembled or in pairs, 3) on the second day, the dog conch hoards himself on the base of the substrate and only elastic probiosis which occasionally appear occasionally at the substrate bottom.

Abstract

Pleurotus ostreatus is an edible mushroom that also has potential as medicinal values. In this study, fruiting body of P. ostreatus was tested for its ability to inhibit the growth of fungi and bacteria. The fruiting body powder of P. ostreatus was extracted using methanol by maceration method. Analysis of this compound was done by using anisaldehid sulfuric acid, Dragendorff reagent, and FeCl3. Using the agar well diffusion technique, the extracts were tested for their antimicrobial activity against Staphylococcus aureus (Gram positive), Enterobacter aerogenes (Gram negative) and Candida albican (yeast). The spot results on TLC using crude extract of P. ostreatus is terpenoids. Zone of inhibition for the various extracts varied between 10.9 - 23.2 mm. Ten miligrams extract exhibit maximum antimicrobial activity against most of the tested pathogens

bacteria that cause an illness. Antibiotic treatments to a patient have a purpose to eliminate the pathogen bacteria. Bacteria resistance to antibiotic was influenced by the intensity of antibiotic treatment in a region, the uncontrolled antibiotics treatments would increase the antibiotic resistance of bacteria. Plasmids was an extrachromosomal DNA that encodes a functional protein that would eliminate the antibiotic activity. Plasmid is the determinant of bacteria sensitivity to antibiotics. In this case it would be important to find out the bacterial plasmid profile on the E.coli resistant to metronidazole and nalidixic acid antibiotics. This research was using four different sample from faces of diarrhea, ice block, waters from well, and ketchup to cultivate the E. coli. lasmid isolation method was carried out by lyses alkali method. Plasmid profile of the E. coli that resistant to metronidazole and nalidixic acid antibiotics and analyzed using electrophoresis on 1% agarose. E. coli plasmid DNA profile was observed as a fluorescent DNA band in ultraviolet rays. In result, isolated plasmids from bacteria that resistant to antibiotics metronidazole and nalidixic acid having similar size approximately 500 bp, different from bacteria that sensitive to antibiotics metronidazole and nalidixic acid has a smaller size in region of 100 bp.
