Buletin Plasma Nutfah (BPN) is an open access scientific journal published by The Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development (ICABIOGRAD), The Indonesian Agency for Agricultural Research and Development (IAARD), Ministry of Agriculture. This peer-refereed journal covering the area of genetic resources including exploration, characterization, evaluation, conservation, diversity, traditional knowledge, management and policy / regulation, of all kinds of genetic resources: plants, animals, fishes, insects and microbes. Manuscripts submitted to this journal are those that have never been published in other journals. This journal is published in one volume of two issues per year (June and December). We invite authors to submit the manuscripts to this journal in English or Indonesian. Abstract

AbstractAn experiment on eighteen genotypes of tomato was conducted at Lembang station (RIV) (1250 m asl) in 1999/2000 planting season to characterize their phenotypic performance, growth type and yield. The experiment was arranged in a Randomized Block Design with 18 two replications. Genotypes tested were introduction and landraces. Phenotypic performance growth type fruit color and productive branch number were realtively uniparm while head of plant, fruit diameter, fruit thick and plant age had were different. Tomato lines FMTT 270, FMTT 22 and FMTT 95 had healthy fruit weight per plant, fruit diameter and good thickness at Lembang highland. Lokereal tomato lines PT 4289, PT 4226, PT 4165, and PT 4121 had earlier harvested, to be further use as breeding material.AbstrakDelapan belas galur koleksi plasma nutfah tomat lokal dan introduksi telah dikarakterisasi di KP Balitsa (1250 m dpl) pada MT 1999/2000 untuk melihat karakter pertumbuhan, hasil, dan komponen hasil. Percobaan ditata dalam rancangan acak kelompok dengan dua ulangan. Berdasarkan gugus rata-rata Scott-Knott diketahui bahwa tipe tumbuh, warna buah, dan jumlah cabang produktif genotipe relatif seragam, sedangkan tinggi tanaman, jumlah dan bobot buah per tanaman, diameter buah, ketebalan daging, dan umur tanaman relatif berbeda. Galur FMTT 270, FMTT 22, dan FMTT 95 mempunyai bobot buah sehat dan ketebalan daging buah yang baik di dataran tinggi Lembang, dan galur PT 4289, PT 4226, PT 4165, dan PT 4121 berumur genjah sehingga diharapkan dapat digunakan sebagai bahan pemuliaan lebih lanjut.

Abstract

AbstractExploration was conducted to be used for genetic variation improvement of abaca germplasm. It was performed at Sangir Besar Island in Sangihe Achipilago and Karakelang Island in the Talaud Archipilago, both of them were located in the teritorial of Sangihe-Talaud District of North Sulawesi Province, in September 1999. Result indicated that Sangihe-Talaud District had many species of abaca introduced from Philippina. Fifteen accessions of abaca were collected. The difference characters of each accession were identified by stem colour, bloom shape and colour, stem height and diameter, and fibre strength. Those accessions were conserved in the RITFC’s Experimental Garden for characterization and evaluation.AbstrakUntuk memperluas keragaman sumber daya genetik tanaman abaca, telah dilakukan eksplorasi di Pulau Sangir Besar di gugusan Kepulauan Sangihe, dan di Pulau Karakelang di gugusan Kepulauan Talaud, Kabupaten Sangihe-Talaud, Propinsi Sulawesi Utara, pada bulan September 1999. Dari eksplorasi tersebut diketahui bahwa di Kabupaten Sangihe-Talaud, Propinsi Sulawesi Utara khususnya di Pulau Sangir Besar dan Pulau Karakelang, banyak terdapat jenis abaca yang diduga berasal dari Filipina. Diperoleh 15 aksesi abaca dari kedua pulau tersebut. Perbedaan yang mencolok dari masing-masing aksesi terletak pada karakter warna batang, warna dan bentuk jantung, tinggi dan diameter batang serta kekuatan seratnya. Aksesi-aksesi tersebut ditanam di kebun plasma nutfah Balittas untuk karakterisasi dan evaluasi.

Abstract

AbstractDirect induction of somatic embryos in Arabica coffee (Coffea arabica L.) using plant groth regulators (PGR's) has been successful. The concentration and combination of different kinds of PGR's can influence the response and success in embryo induction. An experiment was conducted to determine the optimal concentration of 2.4-D in combination with kinetin for direct induction and proliferation of somatic embryos. The plant material used was Arabica coffee var. Kartika-l originating from The Indonesian Coffee and Cacao Research Institute, Jember. Explants were taken from young leaves of reddish-green in color. Somatic embryos were induced directly on a Murashige-Skoog (MS) standard medium containing 30 g/l sucrose and supplemented with 0, 1, 2, 4, and 8 mg/l 2.4-D in combination with 0.1 mg/l kinetin each. The cultures were incubated in the dark at temperature 26oC and RH +60% for 6 weeks with 10 replications. The results showed that somatic embryogenesis in Arabica coffee was best induced in a culture medium wiyh 2.4-D at 4 mg/l, combination with 0.1 mg/l kinetin. Induction of somatic embryos was achieved at 100% 4 weeks after culture. Three morphological stages of embryo development were identified: globular, early heart, and middle heart. The embryos were of three distinct colors such as, yellowish, yellowish-white, and white. The highest rate of proliferation of somatic embryos was achieved at 2 mg/l, 2.4-D in combination with 0.1 mg/l kinetin averaging 68.53 embryos per explant 6 weeks after subculture.

Abstract

AbstractStudy on the performance of two quality protein maize (QPM) varieties (Srikandi Putih-1 and Srikandi Kuning-1) plus two Indonesian open pollinated varieties (Bayu and Lamuru) was conducted at seven environments in Java and Bali. The experiments were arranged in a randomized block design with three replications. Each variety was grown in a four-row plot with 5 m length, 75cm apart and 25 cm within row spacing and one plant per hill. Observations were made on yield, biomass, plant height, ear height, flowering dates, yield components, plant and ear aspect, husk cover, diseases incidence of Puccinia polysora and Helminthossporium maydis, protein contains, lysine, and tryptophan-amino acids contains. In general, Srikandi Putih-1 and Srikandi Kuning-1 varieties had a good adaptations under all environments. They contained nearly twice the lysine and tryptophan-amino acids compared to Bayu and Lamuru varieties. Therefore, they have future prospect for the development of protein synthesis in humans and monogastric animals.AbstrakPenelitian penampilan varietas jagung unggul baru bermutu protein tinggi (Srikandi Putih-1 dan Srikandi Kuning-1) dan dua varietas unggul nasional (Bayu dan Lamuru) telah dilaksanakan pada tujuh lokasi di Jawa dan Bali pada tahun 2003 sampai 2004. Penelitian ditata dalam percobaan acak kelompok dengan tiga ulangan. Setiap varietas ditanam pada petakan empat baris, panjang 5 m, jarak antar baris 75 cm, jarak dalam barisan 25 cm, dan ditumbuhkan satu tanaman per rumpun. Pengamatan dilakukan terhadap hasil biji, biomas segar, tinggi tanaman, panjang tongkol, umur berbunga, komponen hasil, skor penampilan tanaman, penutupan kelobot, penyakit karat dan hawar daun, serta kandungan protein kasar, asam amino lisin, dan triptofan. Secara umum varietas Srikandi Putih-1 dan Srikandi Kuning-1 dapat beradaptasi baik pada semua lingkungan. Selain itu, kedua varietas tersebut mempunyai kandungan lisin dan triptofan hampir dua kali lipat dibandingkan dengan varietas Bayu dan Lamuru. Oleh karena itu, kedua varietas tersebut mempunyai prospek untuk dikembangkan sebagai sintesis protein pada ternak monogastrik dan manusia.

Abstract

AbstractThere are several sources of publication could be used as database. The sources could be articles that have been published in the periodicals as well as unpublished data (idle data). Database manager should be the person who know the background, objectives, and the principles to manage the data. In addition, the coordinators should be the persons who have background of animal geneticist by training. Data on the characteristics of breeds or strain within species are extracted and presented on specific format. The format are either in the free or fixed format, for computerization. The presentation could be separated into two form of Master Records containing physical characteristics, and Slave Records containing performance characteristics and environment. Each breed or strain has only one Master Record, but it could have several Slave Records. The number of slave records are dependent upon the number of publication available.AbstrakDi dalam pembuatan pangkalan data (database) yang merupakan bank data, bermacam-macam sumber dapat digunakan, mulai dari artikel yang telah dipublikasi maupun data yang tidak dipublikasi (idle data). Pengelola database harus mengetahui latar belakang, tujuan serta prinsip-prinsip dasar penanganan data. Koordinatornya paling tidak, telah mendapat pendidikan di bidang pemuliaan ternak (animal geneticist by training). Data karakteristik suatu rumpun (breed) atau strain dalam spesies diekstrak dan dipresentasikan dalam suatu format, baik dalam bentuk bebas (free format) maupun tetap (fixed format), sehingga mudah untuk dilakukan komputerisasi. Presentasinya dibedakan menjadi dua, yakni dalam bentuk karakteristik fisik (master record) serta untuk karakteristik performa dan lingkungan (slave record). Setiap rumpun atau strain mempunyai sebuah master record dan beberapa slave record. Jumlah slave record bergantung pada jumlah publikasi yang tersedia.

Abstract

AbstractSelf sufficiency in rice has always been a heavy task due to limited number of sustained high yielding varieties available. Therefore, effort must be put on the breeding for high yielding varieties adapted to different agroecosystems. Development of high yielding variety should use available genetic resources at ICABIOGRAD as well as at IRRI. So far, accessions collected at ICABIOGRAD gene bank were 3563 cultivated rice and 100 wild rice. To be better able to utilize those accessions, dynamic conservation consisting of in situ and on farm conservation are needed. Wild rice should be conserved at the National Park while the traditional varieties by the farmers as well as NGO’s. Wild germplasm should be used to develope high yielding variety by using molecular marker. Development of better database will certainly facilitate the base of available genetic resources for the purpose of better high yielding variety. So far, more than 160 varieties of rice suitable for lowland, upland and tidal swamp have been released.AbstrakUpaya untuk memenuhi kebutuhan beras penduduk mendapat tantangan berat mengingat varietas unggul padi yang tersedia hanya sedikit yang mampu beradaptasi baik dan bertahan lama. Oleh karena itu, para pemulia harus lebih giat dalam merakit atau memperbaiki varietas sesuai dengan agroekosistem pengembangan, karena setiap varietas unggul menuntut sejumlah persyaratan untuk dapat menampilkan keunggulannya secara maksimum. Perakitan varietas unggul harus lebih memanfaatkan secara luas sumber keanekaragaman plasma nutfah padi yang terdapat dalam Bank Gen BB-Biogen. Hingga kini Bank Gen BB-Biogen telah melestarikan sebanyak 3563 plasma nutfah padi dan 100 aksesi padi liar. Agar plasma nutfah dapat lebih diberdayakan maka perlu dilakukan cara konservasi yang lebih dinamis seperti pelestarian in situ dan lekat lahan (onfarm conservation). Spesies padi liar sebaiknya dilestarikan di Taman Nasional dan padi tradisional dilestarikan oleh masyarakat termasuk petani dan LSM di daerah-daerah tertentu untuk dikembangkan. Penggunaan tetua yang lebih beragam diharapkan dapat dihasilkan varietas unggul dengan sifat-sifat yang diinginkan termasuk pemanfaatan spesies padi liar dalam persilangan kerabat jauh dengan menggunakan teknologi marka molekuler. Akses terhadap informasi sumber daya genetik perlu lebih dipermudah dengan mengembangkan database yang lengkap dan membangun jaringan website. Sampai Agustus 2003 di Indonesia telah dilepas lebih dari 160 varietas unggul padi yang sesuai untuk lahan sawah, lahan kering (gogo), dan lahan rawa pasang surut.
