Abstract

ABSTRACT Immobilization of xylanase with physical adsorption method on sea sand matrix is carried out to increase stabilization of enzyme and can be reused. Xylanase is isolated from Trichoderma viride, purified by precipitation method by using ammonium sulfate with saturated level of 40-80%. This research is to know the shaking time and optimum concentration of xilanase. Variance of shaking time is (1,2,3,4,5) hour and variance concentrate of xylanase (0.5; 1.5; 2.5; 3.5; 4.5) mg/mL on 0.1 g of sand at room temperature. Content of xylanase enzyme used for immobilization is 4.517 mg/ml with activity was 17.029 units . The results of the research showed that the optimum condition of immobilization xylanase is achieved when shaking time is 2 hours and concentrate of xylanase is 4.5 mg/mL within absorbed xylanase is 205.120 mg/g and activity of immobilizied xylanase is 135.886 units. Abstract

ABSTRACT This aims of this study is to determine the optimum conditions and characteristics of fermented suweg flour. The optimum conditions are determined based on the maximum levels of lactic acid produced in the fermentation process. Fermentation conditions are studied based on the variation of pH (3,4,5,6,7), temperatur (30oC, 35oC, 40oC, 45oC, 50oC) and incubation time (3,6,9,12,15 hours) The characteristic of fermented and non fermented suweg flour include starch, amylase, amylopectin and expands power. The optimum fermentation conditions achieved at pH 6, temperatur 45°C, and 12 hours of incubation time. The characteristics of fermented suweg flour at the optimum conditions reach higher starch, amylose, amylopectin and expands power, better than non fermented flour. The contents of starch, amylose, and amylopectin in the non fermented suweg flour are 83.7%, 11.72%, 71.98% respectively. The content of starch, amylose, and amylopectin in fermented suweg flour are 87.3%, 12.15%, 74.82% respectively. Expands power of non fermented suweg flour is 35.29%, while expands power in fermented suweg flour is 41.18%. Key words: Fermentation, Flour, Lactobacillus bulgaricus, Suweg

Abstract

ABSTRACT Pyrophyllite as an inorganic material having an active site that can serve as an ion exchanger, so that potentially to be used as ionophore ion selective membrane. Ion Selective Electrode (ISE) lead (II)-based on pyrophyllite as an ionophore, Polyvinylchloride (PVC) as the matrix polymer, activated carbon and dioktilftalat (DOP) as a plasticizer in tetrahydrofuran solven has been developed. Variables of this research consist of immertion time doing at 10, 20, 30, 40, 50, 60, 70, 80 minutes and composition of the membrane. Basic characterization of ion ISE Lead (II) coated wire based on pyrophyllite were examined Nernst Factor, limit of detection, linear concentration range, response time, and life time. The results of research showed that, the optimum immertion time to 50 minutes, with the optimum membrane composition consist of Pyrophylite : PVC : activated carbon : DOP is 49 : 16 : 3 : 32 (% b/b). Characterization of Lead (II)-based coated wire type pyrophyllite produced include: linear concentration range of lead 10-5 - 10-1 M at a price of 29.33 Nernst Factor mV/decade of concentration. The resulting detection limit is 8.054 x 10-6 M or equal to 1,669 ppm of lead, the resulting response time of 30 seconds and a life time of 45 days. Abstract

ABSTRACT Pectinase isolated from Aspergillus niger was immobilized by entrapping method in a matrix of carrageenan. The aim of this research was to determine the optimum concentration of carrageenan and enzyme. Carrageenan concentration varied (0.017; 0.033; 0.050; 0.067; 0.083) g/mL, while the concentration of enzyme  varied  (0.632; 1.263; 1.895; 2.526; 3.158) mg/mL. Protein content was determined by Biuret reagent  and enzyme activity was determined from the number of reduced sugar as a result of  hydrolyzed pectin per minute. The results showed that optimum concentration of carrageenan was 0.050 g/mL and the optimum enzyme concentration was 1.263 mg/mL with the amount of entrapping enzyme was 3.048 mg/g carrageenan and maximum activity of  348.5 units.
