Abstract

[ANTAGONISTIC ACTIVITY OF A Bacillus sp. Bth22 AND Paenibacillus sp. CONSORTIUM AGAINST Ralstonia solanacearum IN CHILI (Capsicum annuum L.) UNDER IN VITRO AND GREENHOUSE CONDITIONS]. Bacterial wilt caused by Ralstonia solanacearum is a major constraint in chili cultivation. This study evaluated the antagonistic activity of Bacillus sp. Bth22, Paenibacillus sp., and their consortium against R. solanacearum under in vitro conditions, and their ability to delay symptom onset and suppress bacterial wilt intensity in chili under greenhouse conditions. The antagonism assay followed a completely randomised design with four treatments — pathogen without biocontrol agent, Bacillus sp. Bth22 + pathogen, Paenibacillus sp. + pathogen, and consortium + pathogen — with three replicates each; the greenhouse experiment used six replicates of five plants per replicate, with the replicate mean as the unit of analysis. Treatments significantly affected inhibition zone width (p < 0.001), with consortium + pathogen producing the widest zone (18.833 mm), followed by Paenibacillus sp. + pathogen (16.567 mm) and Bacillus sp. Bth22 + pathogen (13.967 mm), while pathogen without biocontrol agent produced none. The compatibility assay showed no inhibition zone at the intersection of the two isolates, indicating compatibility. In the greenhouse, first symptoms appeared at 2.97, 8.80, and 15.03 days in pathogen without biocontrol agent, Bacillus sp. Bth22 + pathogen, and Paenibacillus sp. + pathogen, whereas no symptoms appeared in consortium + pathogen until the end of the 28-day observation (censored value, >28 days). Disease intensity in consortium + pathogen remained 0% up to 28 days after inoculation, whereas P0 reached 95.57%. The consortium therefore showed higher antagonistic activity and disease suppression than the single isolates and is a candidate for biocontrol formulation; field efficacy and the nature of the interaction between the two isolates require further study.

Abstract

[ORGANIC PRIMING FOR THE INVIGORATION OF DETERIORATED SORGHUM SEEDS (Sorghum bicolor (L.) Moench)]. Seed deterioration in new superior varieties (VUB) is one of the constraints in crop development, including sorghum Bioguma 3 Agritan. This study aimed to evaluate the effects of organic priming and soaking duration on the viability of deteriorated sorghum seeds. The experiment was conducted at the Seed Technology Laboratory of UPN “Veteran” Jawa Timur using a factorial completely randomized design with two factors, namely type of organic priming (sprout extract, spirulina extract, and combined extract) and soaking duration (6, 12, 18, and 24 h). The observed variables included germination percentage, maximum growth potential, fresh weight of normal seedlings, and dry weight of normal seedlings. The results showed that, in severely deteriorated sorghum seeds, priming with 10% mung bean sprout extract for 6 h produced the highest germination percentage, increasing germination by 97.17% compared with the untreated control, and gave the best result for maximum growth potential. This treatment was comparable to 10% spirulina extract for 12 h and combined extract (sprout extract +spirulina) for 12 h. The highest fresh weight of normal seedlings was obtained from 10% spirulina extract for 12 h, whereas dry weight of normal seedlings was relatively similar among treatments. Soaking for 24 h in all types of organic priming was not recommended because it tended to reduce the viability of deteriorated sorghum seeds.

Abstract

[LAND SUITABILITY EVALUATION FOR PADDY RICE DEVELOPMENT IN KARTINI VILLAGE, JAGEBOB DISTRICT, MERAUKE REGENCY, SOUTH PAPUA, INDONESIA]. Jagebob District is one of the land optimization program sites in Merauke Regency, yet village-level information on land suitability for paddy rice remains limited. The objectives of this research were to establish the actual land suitability class for paddy rice in Kartini Village, Jagebob District, Merauke Regency, South Papua, to determine the factors limiting that class, and to propose land management directions. A survey covering 383.10 ha was carried out between January and April 2026. Six land units were delineated by overlaying slope, soil type, land use, and landform maps. In each land unit, soil was sampled purposively at five points and composited into a single sample. Sixteen land characteristics were assessed by matching them against paddy rice growth requirements: temperature, water availability, drainage, texture, soil depth, pH (H₂O), cation exchange capacity (CEC), organic carbon, total nitrogen, P₂O₅, K₂O, slope, erosion hazard, flood hazard, surface stones, and rock outcrops. Annual rainfall in the area reaches 2,237.54 mm, the mean temperature is 26.85 °C, and the Oldeman agro-climatic type is C2. Climatic and physical conditions support paddy rice cultivation: clay texture, somewhat poorly to poorly drained soils, 120 cm soil depth, 0–3% slopes, and very low erosion hazard. In contrast, soil chemical properties are limiting: very acidic to acidic pH (4.13–5.14), low CEC (9.03–14.88 cmol/kg), very low organic carbon (0.78–0.88%), low to moderate total nitrogen, and very low P₂O₅. All land units were classified as marginally suitable with the subclass S3nr,na. The limiting factors were nutrient retention, namely organic carbon in all land units and pH in land units 1, 2, 4, 5, and 6, and nutrient availability, namely P₂O₅ in all land units. Paddy rice development in this area requires organic matter addition, soil acidity management, and balanced N and P fertilization.
