Abstract

The study about population density and shell size distribution of clam Contradens sp. in Tanjung Mutiara Singkarak Lake, West Sumatra has been conducted on July 2013. This study used stratified purposive sampling method. Sampling site was decided into 3 locations based on human activities, 3 depth strata in each location; <5m, 5-10m and >10-15m. The clams were collected using “dauah” (traditional tool to catch shellfish), 3 sampling plots for each stratum. The result showed that the highest average population density (2.596 ind/m2) was found in location III followed location II (0.055 ind/m2) and location I (0.155 ind/m2). Based on strata, the highest average population density (1.941 ind/m2) was at >10-15m depth, there was no clam found at <5m depth. The largest clam concentrated  at >10-15m depth in location III (without human activity area) whereas small to medium clam concentrated at 5-10m depth.Keywords: clam, Contradens sp., population density, size distribution, Singkarak Lake.

Abstract

The research on vegetation analysis of weed in palm oil plantation (Uncaria gambir (Hunter) Roxb) in Penurunan village, Nagari Kayu Gadang, Sutera, Pesisir Selatan has been conducted from February to April 2013 by using 1 x 1 m 20 plots of purposive quadrate method. The result showed that weed composition in gambir plantation consisted of  15 families, 28 genera and 31 spesies while Index diversity of weed was H’ = 2.54. Axonophus compressus has the highest SDR (Summed Dominance Ratio) 23.02% while Eleusine indica has the lowest SDR 0.40% (2 individuals) in gambir plantation.Keywords : Vegetation analysis, weed, gambir, quadrate method, plot

Abstract

This study aimed of to find out the effect of leaf extract and seed powder containing alkaloid which occured on the production process of nata de coco. An experiment was conducted using Complete Randomized Design (CRD) with factorial pattern. The result showed that the brewed extract of tea, coffee, chocolate affected the weight product of nata de coco. The brewed extract of green tea showed the best result on weight of the nata (24.66 g) which has a significant difference from the treatments of black tea, coffee powder, cocoa powder, coffee leaf, and chocolate leaf. The best dose of brewed extract in obtaining nata was 2 g/100 ml.Keywords : alkaloid, nata de coco, effect.

Abstract

Research about the affectivity of gambier (Uncaria gambir Roxb.) as anti-hypercholesterolemic and stabilizer of blood value on the male white mice (Mus musculus) have been done from August to December 2013 in the Research Laboratory of Animal Physiology, Biology Department, Mathematics and Natural Sciences Faculty, Andalas University, Padang and Sejawat Clinical Laboratory, Padang. This study aimed to determine whether gambier could inhibit the increase in total cholesterol and stabilize blood value of white mice that were given with the leftover frying oil and to determine the best dose of gambier in inhibiting the increase in total cholesterol and stabilize blood value of white mice that were given with the leftover frying oil. This study used an experimental method completely randomized design (CRD) with five treatments and five replications. Treatment with giving the leftover frying oil and gambier dose of 50; 100; 150 and 200 mg/kg and control with giving the leftover frying oil without giving gambier. The result showed that the gambier could inhibit the increase in total cholesterol and stabilize blood of white mice that were given with the leftover frying oil and the best dose is 50 mg/kg body weight. Abstract

The study about community of macrozoobenthos at Batang Ombilin River, West Sumatra has been done from November 2012 until March 2013. This study aimed to know the composition and structure of macrozoobenthic community. This research carried out by using survey method and purposive sampling technique. Sampling site determined in six stations. In each station were collected five samples by using Surber net (30x30 cm2). The result showed that community of macrozoobenthos were found 34 species consist of Insecta 20 species, Gastropoda 5 species, Olighocaeta 4 species, Hirudinea 2 species, Lamellibranchiata, Arachnida and Turbellaria one species respectively. The highest relative density was Gastropoda (46.27 %) and the lower was Arachnida (0.28 %).  Average population was 130.18 ind/m2, and the highest density has found at Station I (391.07 ind/m2), the lower at Station V (37.77 ind/m2). Dominant species in each station were variated. Average diversity index was 1.73, the highest was found at Station V (H’=2.22) and the lowest at Station III and IV (H’=1.19).The composition of macrozoobenthos among stations  were different, with low similarity index ranged from 14.28 % - 36.36 %.Key words : Batang Ombilin, Makrozoobenthos, Composition, Structure.

Abstract

We examined the effectiveness of some insecticide aerosols to German cockroaches. This experiment was conducted at the Laboratory of Animal Physiology Department of Biology, Andalas University from July to October 2013. Animal subjects were three strains of German cockroaches, namely GFA-JKT from Jakarta, PLZ-PDG from Padang and VCRU-WHO from School of Biological Sciences, University Sains Malaysia. Those subjects were treated using spray method of five brands of commercial insecticides found in the market, namely ByWS, HtWS, MtWS, RdWS and VpWS. We analyzed  knockdown and lethal times using probit analysis. The effectiveness were evaluated based on running time to achieve the knockdown and lethal. The results showed that knockdown time (KT90) of ByWS, HtWS, MtWS, RdWS and VpWS were effective for VCRU-WHO and GFA-JKT strains and the only HtWS and RdWS were effective for PLZ-PDG strain. Based on the the lethal time (LT90)  there was no single insecticide effective to kill cockroach.Kata kunci: Blattella germanica, commercial insecticide, Spray Method, Knockdown.

Abstract

Research about partial optimization of thermophilic bacteria isolate M5-24 on protease production was conducted from May 2013 to December 2013 in Laboratory of Microbiology, Biology Department, Andalas University, Padang. Purpose of the research was to find out optimum condition of protease production by isolate M5-24. This study consisted of three steps. Step 1: Investigating the effect of temperature and pH used completely randomized design in factorial with two replications.The first factor was temperature (450C, 500C, 550C, 600C), the second factor was pH (7, 8, 9, 10). Step 2: Investigating the effect of inoculum doses and inducers used completely randomized design in factorial with three replications. The first factor was inoculum doses (1%, 5%, 10%) and the second factor was inducer (yeast extract 0,5%; pepton 0,1%;  kasein 1%; meat extract 0,5%). While Step 3: Investigating the effect of agitation (125 rpm, 150 rpm, 175 rpm, 200 rpm) used completely randomized design with six replications. The result showed that the best duration of growing was at 18th hours after incubation, the highest enzyme activity was at temperature 500 C;  medium’s pH at 8; the best of inoculum dose was at 5%; the best  inducer was casein 1% and optimum agitation was at 175 rpm. Extrinsic optimization can increase protease production of  thermophilic isolate M5-24 up to 1.7 times.Keywords : Optimization, Protease,  thermophilic, bacteria

Abstract

Dogs are the most popular pet animal although they could carry ectoparasites which can cause diseases in human. The purposes of this study were to identify ectoparasites and to determine their prevalence on pet dogs. This study was conducted from March to July 2013 using survey method and collecting ectoparasites directly from 15 pet dogs in Padang. Specimen processing and species identification were done at the Animal Taxonomy Laboratory, Biology Department, Andalas University. This study found 5 species of ectoparasites in pet dogs. There were Ctenocephalides canis Curtis, 1826, Dermacentor sp., Haemaphysalis sp., Rhipicephalus sanguineus Latreille, 1806 and one unidentified species sp. 1 from family Laelapidae. Rhipicephalus sanguineus has the highest prevalence (73,3 %) followed by Haemaphysalis sp. (46,7%), Dermacentor sp.(20%), Ctenocephalides canis (13,3%) and sp. 1 from Family Laelapidae (6,7%).Keywords : ectoparasite species, prevalence, pet dogs

Abstract

An experimental study to figure out the effect of activated charcoal and coconut water to shoot encapsulation of Tetrastigma rafflesiae Miq. has been carried out from October 2013 to February 2014 at Plant Physiology Laboratory, Biology Department, Faculty of Sciences, Andalas University. The experiment used Completely Randomized Design with four treatments and six replications. Treatments consisted of 1g/L activated charcoal, 300 mL/L coconut water and combination between 1g/L activated charcoal and 300 mL/L coconut water. The result showed that addition of activated charcoal and coconut water decreased viability of T. rafflesiae encapsulation. The result indicated that the use of MS (Murashige Skoog) medium with 4% sodium alginate and 50 mM CaCl2.2H2O formed synthetic seeds with high germination percentage (83%).Keywords : Tetrastigma rafflesiae, encapsulation, activated charcoal, coconut water

Abstract

 Research on the “Effect of acidification of coconut water and rice washing water as an composting alternative of growth medium for gray oyster mushroom (Pleurotus Sajor Caju (Fries) Singer)” was conducted from February to March 2014 in Microbiology Research Laboratory, Biology Departement, Faculty of Mathematics and Natural Science, Andalas University. The research aimed to determine the acidity level that can be used as an alternative for the composting of growth medium of gray oyster mushroom as well as the effect of addition of coconut water and rice washing water for the misellium growth. Adjusting pH of coconut water and rice water showed different effects to the growth myselium and the production medium (baglog). Completely Random Design (CDR) with factorial pattern is the method that used in this research. Acidification of coconut water and rice water with pH 3, pH 4 and normal pH have different effect. The fastest growth of mycelium obtained from coconut water with pH 4 liming treatment (9 days).Key Words : acidification, coconut water, rice washing water, Pleurotus sajor caju, composting alternative
