Abstract

Aloe vera L. contains antiseptic saponins and anthraquinone complexes as antibacterial. This research was conducted to formulate aloe vera powder into nanoemulgel, characterize and test its physical properties, and to know the effect of gelling agent concentration (carbopol and chitosan) on nanoemulgel. Emulsion type used is oil in water (O / W) with surfactant tween 80 and co-surfactant propylene glycol. Nanoemulsion is characterized by percent of transmittance with UV-Vis spectrophotometer, also potential droplet and zeta size distribution with Particle Size Analyzer. Hydrogels consist of 4 combination formulas of carbopol 0.5%; 1%; 1.5%; 2% and 0.3% chitosan. The formula is tested for physical properties including organoleptic, pH, spreadability, adhesion, viscosity and stability with freeze thaw method. The results showed that the nanoemulsion droplet size was 65.05nm ± 13.49 with zeta potential of -0.1mV and the percent of transmittance above 98%. The result of physical properties of the four formulas shows that the different gelling agent concentration gives different physical properties of viscosity and spreadability. Overall, formula 3 has the best physical properties and physical stability compared to formula 1, 2 and 4.

Abstract

Urinary tract infections are infections are characterized by the presence of bacteria that grow and multiply in the urinary tract in excess of the normal amount. Based on the Indonesian Health Profile, urinary tract infections (UTI) are the 10 most common causes of death. Improper use of antibiotics can cause no therapeutic effect and resistance. This study aims to evaluate the accuracy of antibiotic use in patients with urinary tract infections in central hospital Klaten in 2017 covers the right indication, right patient, right medication, and right dosage. Research with non-experimental methods of data retrieval retrospectively and analyzed descriptively. The inclusion criteria in this study were adult patients 18-64 years old, inpatients suffering from urinary tract infections including cystitis and pyelonephritis, receiving antibiotics and complete medical record data. The results showed that patients with urinary tract infections in RSUP in Klaten 2017 had 72 patients with 76 antibiotic prescriptions. Based on the number of patients, the evaluation results showed the right indication is 100% and the right patients is 100%, while the exact evaluation results of the right drug is 96.05% and the right doses is 27,63%.

Abstract

Streptomyces sp. GMR22 is local isolate from Wanagama 1 Forest in Yogyakarta. They have the potential to be developed to produce active compounds because have PKS and NRPS genes.The active compounds from isolation are strongly influenced by various factors, one of them is extraction techniques. Effect difference of extraction technique will be affected by the quality of secondary metabolites produced.The purpose of this study was to compare the cytotoxicity effects of secondary metabolites of Streptomyces sp. GMR22 which have extracted with different stages from previous studies. The extraction technique was carried out by multilevel separatory funnel extraction methods, which was first extracted using non-polar solvent (n-hexane) and then extracted using semi-polar solvent (ethyl acetate). This research is important because in previous studies (separatory funnel only extracted using ethyl acetate) with the use of the lowest concentration in the dengue virus antiviral test (further test) caused 100% of deaths in BHK-21 cells.This study indicate that multilevel extraction result in lower CC50 value than previous studies. There are 49.160 µl/ml (n-hexane extract) and 284.56 µl/ml (ethyl acetate extract) while water extract is 464,38 µl/ml. FTIR compound analysis show that the three extracts produced have different spectrum patterns, especially in the n-hexane and ethyl acetate extract. Value of CC50 is not too high, it is expected that the secondary metabolites contained in the extracts can be used for further analysis such as antiviral testing because it is safe for normal host cells such as BHK-21 cells

Abstract

Kaffir lime peels (Citrus hystric DC.) have been reported that it has activity on HeLa cell. The purpose of this study was to determine the cytotoxic activity of ethanol extract, ethanolic, ethyl acetate, and hexane fraction of kaffir lime peels to T47D breast cancer cells and to identify the compounds of kaffir lime peels (Citus hystrix DC.). Extraction was done by maceration using 96% ethanol. The results of thick extracts were fractionated by the liquid-liquid partition method using solvent with different polarity, namely hexane, ethyl acetate, and ethanol. Cytotoxic activity tests were carried out using the MTT assay. Identification of compounds were carried out by Thin Layer Chromatography (TLC) method with silica gel GF 254 as a stationary phase. The mobile phase used was hexane:ethylacetate (6:4) for ethanol extract, ethyl acetate and hexane fractions, and hexane:ethylacetate (4:6) for ethanol fractions. IC50 ethanol extract, ethyl acetate and hexane fractions obtained were 268.49, 286.82, and 136.27 mg/mL respectively. Ethanol extract, ethyl acetate, and hexane fractions contain alkaloid, flavonoid, terpenoid and phenolic compounds, while the ethanol fraction contains only flavonoid compound. Cytotoxic results of ethanol extract, ethanol, ethyl acetate, and hexane fraction of kaffir lime peels did not active against T47D breast cancer cells.

Abstract

Spirulina platensis contained phycocyanin that has the potential for antioxidant activity. Bentonite contains various minerals that can absorb water and was known having ability for scavenging of free radicals. The purpose of this study was to obtain the optimum formulae of sheet mask essence, containing combination of Spirulina platensis extract and bentonite nanoparticles and evaluated the antioxidant activity using DPPH (2,2-diphenyl-1-pikrihidrazil) assay. Bentonite nanoparticles was prepared by High Energy Milling (HEM) method and characterized using Particle Size Analyzer (PSA). The results of the nanoparticles characterization was 152,8±0,69 nm for the particle size and 0.106±0.03 for PI value. Physical stability tests showed that the formula was homogenous, viscosity between 0.7-1.1 dPas, and pH between 5-6.5. The IC50 value of Spirulina platensis extract was 256.46 ppm with AAI category was medium. The antioxidant test of this fomula that contain Spirulina platensis extract and bentonite nanoparticles did not show any potential antioxidant activity. The concentrations of PEG 400 and Tween 80 were used to formulate the optimum formula were 4.29% w/w and 6.71% w/w.
