Abstract

Asap rokok merupakan salah satu penyedia radikal bebas eksogen yang dapat menimbulkan stres oksidatif dan berpotensi menyebabkan penyakit degenerative. Tenggulun (Protium javanicum Burm. f.) merupakan salah satu tanaman yang berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak metanol daun tenggulun terhadap kadar malondialdehid (MDA) dan aktivitas superoksida dismutase (SOD) pada darah tikus Wistar yang dipapar asap rokok. Penelitian ini merupakan penelitian eksperimental dengan rancangan acak kelompok dengan posttest-only control group design. 24 ekor tikus putih jantan galur Wistar dibagi dalam 4 kelompok yaitu kelompok kontrol P0 (hanya diberi paparan asap rokok); kelompok perlakuan P1, P2, dan P3 (diberi ekstrak metanol daun tenggulun dengan dosis berturut-turut 50, 100, dan 200 mg/kgBB dan paparan asap rokok). Semua kelompok dipapar asap rokok dengan dosis 3 batang per hari selama 15 menit selama 14 hari. Pada hari ke-15, darah masing-masing tikus diambil untuk dianalisis kadar MDA dan aktivitas SOD. Hasil penelitian menunjukkan bahwa kelompok kontrol P0 memiliki kadar MDA tertinggi yaitu 6,73 ± 0,47 µM dan aktivitas SOD terendah yaitu 1,01 ± 0,12 ng/mL dibandingkan dengan kelompok perlakuan P1, P2, dan P3. Hal ini menunjukkan bahwa paparan asap rokok menyebabkan stress oksidatif. Pemberian ekstrak dengan dosis 50, 100, dan 200 mg/Kg BB secara signifikan (p<0,05) menurunkan kadar MDA dan meningkatkan aktivitas SOD jika dibandingkan dengan kontrol. Semakin tinggi dosis ekstrak yang diberikan, maka kadar MDA cenderung semakin menurun dan aktivitas SOD meningkat. Pemberian ekstrak metanol daun tenggulun dengan dosis 200mg/Kg BB (P3) memberikan hasil terbaik dengan kadar MDA terendah yaitu sebesar 2,91 ± 0,22 µM dan aktivitas SOD tertinggi sebesar 2,73 ± 0,23 ng/mL pada tikus Wistar yang terpapar asap rokok. Pada penelitian ini dapat disimpulkan bahwa ekstrak metanol daun tenggulun secara signifikan mampu menurunkan kadar MDA dan meningkatkan aktivitas SOD pada tikus yang terpapar asap rokok. ABSTRACT Cigarette smoke is a source of free radicals that trigger oxidative stress which can cause inflammation and lung damage. Tenggulun (Protium javanicum Burm. f.) is a plant that is known to have potential as an antioxidant. This study aims to determine the effect of giving tenggulun leaf methanol extract on malondialdehyde (MDA) levels and superoxide dismutase (SOD) activity in the blood of Wistar rats exposed to cigarette smoke. This research is a randomized posttest-only control group design using 24 male white Wistar rats which were divided into 4 groups namely the control group (P0), only given exposure to cigarette smoke; the treatment group (P1; P2, and P3) were exposed to cigarette smoke and given 50, 100, and 200 mg/Kg BW of methanol extract of tenggulun leaves respectively. All groups were exposed to cigarette smoke at a dose of 3 cigarettes per day for 15 minutes for 14 days. On the 15th day, the blood of each rat was taken to analyze its MDA level and SOD activity. The results showed that the control group (P0) had the highest MDA level of 6.73 ± 0.47 µM and the lowest SOD activity of 1.01 ± 0.12 ng/mL compared to the treatment groups P1, P2, and P3. Administration of the extract at doses of 50, 100, and 200 mg/Kg BW significantly (p<0.05) reduced MDA levels and boosted SOD activity when compared to control. The higher the extract dose given, the levels of MDA reduced and SOD activity tends to increase. In this study, the administration of methanol leaves extract of tenggulun with a dose of 200mg/Kg BW (P3) on Wistar rats exposed to cigarette smoke gave the best results with MDA level of 2.91 ± 0.22 µM and SOD activity of 2.73 ± 0.23 ng/mL. The study concluded that the methanol extract of tenggulun leaves significantly reduced MDA levels and boosted SOD activity in rats exposed to cigarette smoke. Abstract

Adas (Foeniculum vulgare Miller) adalah tumbuhan herbal yang mengandung minyak atsiri dengan kandungan senyawa anetol sebagai antimikrobial. Penambahan minyak atsiri adas pada edible coating. merupakan cara untuk melapisi permukaan produk pangan dengan tujuan mencegah kontaminasi patogen sehingga dapat memperpanjang masa simpan. Tujuan dari penelitian ini adalah mengetahui pengaruh penambahan minyak atsiri adas (Foeniculum vulgare Miller) sebagai edible coating pada bakso. Destilasi minyak atsiri adas dilakukan dengan metode Microwave Assisted Extraction (MAE). Destilasi ini menggunakan variabel daun segar dan biji adas. Variabel yang digunakan dalam destilasi adalah daya (80, 240, 400) watt dan waktu (20, 25, 30) menit. Hasil penelitian menunjukkan, rendemen tertinggi dihasilkan dari minyak atsiri adas yaitu sebanyak 1% pada daya 400 watt dengan waktu 20 menit. Minyak atsiri adas memiliki sifat mudah menguap sehingga identifikasi senyawa dapat diketahui dengan metode Gas Chromatography-Mass Spectrometry (GC-MS) . Berdasarkan hasil analisa gugus fungsi diketahui bahwa minyak atsiri mengandung senyawa utama yaitu trans anetol yang memiliki sifat antimikrobial. Kata kunci: adas, antimikrobial, edible coating, Microwave Assisted Extraction (MAE) ABSTRACT Fennel (Foeniculum vulgare Miller) is an herbal plant consists of essential oil containing anethole as an antimicrobial. Adding fennel essential oil to edible coatings is a way to coat the surface of food products to prevent pathogen contamination, thus extending the shelf life. This research aimed to analyze the effect of adding fennel essential oil (Foeniculum vulgare Miller) as an edible coating on meatballs. Distillation of essential oil from fennel leaves and seeds was carried out using the Microwave Assisted Extraction (MAE) method. This distillation uses variable fresh leaves of 100 grams and fennel seeds of 50 grams with variable power of 80 watts, 240 watts, and 400 watts. The time used in distillation is 20 minutes, 25 minutes, and 30 minutes. The results showed that the highest yield obtained was as much as 1% of fennel essential oil at a power of 400 watts with a time of 20 minutes. The identification of chemical compounds contained in the funnel essential oils was done by using the Gas Chromatography-Mass Spectrometry (GC-MS) method. The results showed that the funnel essential oil contained the main compound of trans-anethole with antimicrobial properties. Abstract

In the case of Rubisco Like Protein (RLP) Chromohalobacter salexigen BKL 5 (RLP CS), which is halophilic, the presence of salt bridges (SB) is not too numerous but efficient enough to maintain stability in high salt stress. This study was to determine the effect of mutations SB at position 185 (D185/WT) on the stability and activity of RLP CS through an in-silico approach by replacing aspartic acid with glutamic acid and alanine (D185E and D185A). The methods used were Molecular Dynamics Simulations (MDS) and Molecular Docking (MD). The MDS was used to study molecular characteristics at the atomic level, while MD was for the interaction patterns of proteins and ligands. The results of the MDS analysis carried out at 10ns showed that the mutation at position 185 with alanine and glutamic acid changed the size/dimensional of RLP CS and affected its overall geometric structure. Interestingly, even though the structure has changed, the activity of the protein remains relatively constant, which is indicated by the results of MD, and has a relatively similar binding energy value of around -6.3 Kcal/mol.
