Abstract

Free radicals are atoms or molecules that have one or more unpaired electrons and are highly reactive. Antioxidants can inhibit the excessive oxidation reaction. The leaves of binahong (Anredera scandens (L.) Moq.) are one part of plants which contain flavonoid compounds and tannins that have scientific activity as antioxidants. This study aims to determine the bioautographic profile and to test the antioxidant activity of the ethanol extract of the leaves of Anredera scandens (L.) Moq. useing 2.2-diphenyl-1-pikrilhidrazil radical capture method. The extraction process produced 19.22% yield, 14.68% (14.684 grams) chloroform fraction, 11.22% (11.224 grams) n-hexane fraction, 3.09% (3.094 grams) ethyl acetate fraction, and 23.90% (23.904 grams) n-butanol fraction. The results of bioautographic profile test using KLT densitometry method proved that ethanol extract, chloroform fraction and ethyl acetate fraction have DPPH radical capture activities. The determination of total flavonoid content found that ethyl acetate fraction contained higher total flavonoids than other fractions. In the ethyl acetate fraction, high total flavonoid level and low IC50 values were obtained, while the n-hexane fraction had low flavonoid level and high IC50 values. Keywords: antioxidants, binahong leaves, free radicals, flavonoids, IC50.

Abstract

The presence of phosphate ions in the aquatic environment can cause disturbance of water balance, such as the increase of fertility of quatic plants resulting in water surface closure. Sunlight will be obstructed and the exchange of oxygen in water is also disrupted. Various attempts to remove phosphate ions have been carried out, adsorption is one of the promising alternative methods to reduce these ions from the waters because of the high effectiveness and the cost required for the preparation of the adsorbent is quite cheap. The general objective of this research was to study the selectivity of limestone in adsorpting phosphate ions. The results obtained can complement the data on the use of limestone which in turn can be used as a reference in the management of wastewater contaminated by phosphate ions. The ability of limestone in adsorping phosphate ions was tested using the Batch method with an investigation on the variations of the adsorption time and the concentration of phosphate ions. Limestone characterization test results before and after activation showed an increase in limestone characteristics as follows: surface acidity increased from 0.50 to 1.40 mmol/g and the specific surface area increased from 12.66 to 25.61 m2 /g. The amount of phosphate adsorped to the activated limestone at the optimum contact time of 15 minutes was of 3.125 mg/g, and at the concentration of 70 ppm was of 3.094 mg/g following the Langmuir adsorption isotherm pattern with an R2 value of 0.9981. Keywords: activation, adsorption, characterization, limestone, phosphate ion

Abstract

Indonesia is quite well known as one of the countries that has tea plantations and uses its own production of tea. As a tea producing country, Indonesia has been able as a tea exporter. The most tea production in Indonesia is black tea product. Black tea products are produced from species Camellia sinensis Var. Assamica which undergoes an enzymatic oxidation process to fresh tea leaves. The most phenolic compounds contained in the black tea leaf are catechins, theaflavins, and thearubigin. The purpose of this study was to determine the phenolic total and catechin content of black tea products and acetone extracts produced by D'wan Tea (DT) and Bali Cahaya Amertha (BCA) plantations which located in Tabanan district, Bali. Determination of total phenol was carried out on black tea leaf products and acetone extract using the Folin-Ciocalteu method compared with gallic acid standards. Determination of catechin content was performed on fresh (before oxidized) tea leaves, black tea leaf products, and acetone extracts using the UV-Vis Spectrophotometry method at a maximum wavelength of 257 nm. The results showed that total phenol content of D'wan Tea black tea products (SDT) and Bali Cahaya Amerta (SBCA) were 1.50 ± 0.02 % mg GAE/g dan 0.97 ± 0.02 % mg GAE/g, respectively. Results of the total phenol content of D'Awan Tea (EADT) black tea acetone extract was 6.92 ± 0.12 % mg GAE/g, while the Bali Cahaya Amerta (EABCA) black tea acetone extract was 7.38 ± 0.07 % mg GAE/g. Catechin content from fresh leaf samples of D'wan Tea (SSDT), SDT, and EADT were 15.30 ± 0.22 % mg/g, 8.37 ± 0.13 % mg/g, and 37.45 ± 0.74 % mg/g. Catechin content of fresh leaf samples of Bali Cahaya Amerta (SSBCA), SBCA, and EABCA were 14.71 ± 0.28 % mg/g, 15.16 ± 0.58 % mg/g, and 35.66 ± 0.89 % mg/g. Total phenol content of DT black tea products is greater than BCA black tea products. DT black tea products appear to have decreased catechins in processing. In the processing of black tea it is necessary to decrease catechins content due to the formation of catechin polymers, namely theaflavin and thearubigin as a determinant of the quality of black tea. Keywords: black tea, Camellia Sinensis, cathecin, total phenol.

Abstract

Lipsticks are widely used by women to beautify themselves. Among the various colors that make lipstick more interesting, red color lipstick is the most demanded one. Rhodamine B is a synthetic dye that banned for use and certified as a hazardous material according to Minister of Health of Indonesian Republic No. 376/Menkes/Per/1990 because it causes liver damage, kidney and lymph glands damage, followed with organ enlargement. The aim of this study is to identify Rhodamine B in lipsticks in the market. Samples were taken from 3 shops in Denpasar City and Badung Regency. Samples were soaked with amonia solution with using wool yarn to extract the rhodamine B dye and identification using TLC plate silica gel GF254 withn-butanol: ethyl acetate: amonia (1322:5.2:6.5) as mobile phase then detected with UV light 254 and 366 nm. Identification by spectrophotodensitometry where the TLC plate was observed in the TLC Analyzer to observe the AUC in each spot formed. The AUC obtained from the instrument illustrates the concentration of the analyte in each bottle. The result shows that 3 examined samples doesn’t contain rhodamine B. Abstract

Moringa plants is very abundant in Indonesia. Moringa leaves are a source of polyphenol compounds, namely flavonoids. One of the main flavonoids found in Moringa leaves is quercetin. To be able to increase the concentration of quercetin from moringa plants, extraction is necessary. This study aims to determine the most optimal extraction method in extracting quercetin from Moringa leaves. Optimization of quersein extraction methods from Moringa leaves was carried out using maceration, soxhletation, and reflux methods. The solvent used was methanol acidified with 1,2 N HCl. Determination of the concentration of quercetin in the extract used a validated TLC-densitometry method. The highest rendement obtained from the maceration extraction method was 24.08 % w/w. Extraction by reflux and sochletation methods yielded rendement of 23.44 % w/w and 10.12 % w/w respectively. Qualitative analysis of quercetin in the extract was carried out by comparing its Rf value with the standard quercetin which is 0,4. The quercetin level of the extract from maceration extraction was the highest, which was 24.45 % w/w. Extraction using the soxhletation method obtained quercetin as much as 20.95 % w/w. Abstract

Epilachna sparsa is one of the plant pests that damage the epidermal layer on the bottom side of the leaves, so it can lead to damage of leaf tissue and only the skeleton remains. Gayam (Inocarpus fagiferus) is one of the species of the Legumineceae family which has insecticide activity. This study aims to isolate and identify the antifeedant triterpenoid compound from stem of gayam (Inocarpus fagiferus Fosb.) against Epilachna sparsa larvae. Dried powder of 1200 g gayam stems produced 17.82 g of crude methanol extract with antifeedant activity of 60.74% at a concentration of 5% (w/v) which was further separated by partition to obtain n-hexane, ethyl acetate, and water extracts. Extracts of n-hexane, ethyl acetate, and water at 5% (w/v) concentration showed antifeedant activity of 46.74%, 18.09%, and 8.31% respectively. Futhermore, the most active n-hexane extract was identified by phytochemical tests and LC-MS/MS. The phytochemical test showed n-hexane extract contains triterpenoid compounds. The mass spectra from LC-MS/MS chromatogram peaks with MassLynx V4.1 software and Chemspider database for n-hexane extracts which were previously purified by SPE (Solid method) Phase Extraction) using methanol and dichloromethane (DCM) eluents show the presence of components of flavonol pyrogallol, sucrose2,3,3 ', 4', 6-pentaacetate, soyasaponin I, and a compound with a molecular weight of 685,225 g/mol which cannot be identified further because there is no match spectra with the database. Keywords: antifeedant, Epilachna sparsa, gayam, Inocarpus fagiferus Fosb., triterpenoid.
